Project *** discuss a high-throughput automation framework for FEFF cal.ul.tions,buil. on robust,rigorousl. benchmarked *** is a computer program uses a real.space Green’s function approach to cal.ul.te X-ray absorption *** wil. demonstrate that the El.IE al.orithm,which combines 33 weak“l.arners”comprising a set of preprocessing steps and a simil.rity metric,can achieve up to 84.2% accuracy in identifying the correct oxidation state and coordination environment of a test set of 19 K-edge XANES spectra encompassing a diverse range of chemistries and crystal.*** XASdb with the El.IE al.orithm has been integrated into a web appl.cation in the Material. 全球气候变化将严重影响北极的生态系统,而地区特有的模式和非均匀趋势使得对这些影响进行预测变得复杂。24个无冰水面越冬地区(或称为"小生境")被认为对加拿大高纬度北极地区和巴芬湾地区的8种海鸟及海洋哺乳动物具有特殊的重要性。我们在1979～2001年间的每年3月对无冰水面面积的局部发展趋势进行了研究,这些趋势是从卫星微波遥测的海冰密集度近似值中获取的。在巴芬湾、戴维斯海峡、格陵兰岛西岸和兰开斯特海峡的小生境中发现了无冰水面面积缩小的趋势。而在哈得逊湾、哈得逊海峡和福克斯湾中观测到了无冰水面面积增加的趋势。我们通过物种分布和丰度对每一个小生境的生物重要性进行了研究。主要海洋食肉动物赖以生存的水域面积的减少可能带来的后果包括对觅食效率和氧气及猎物的供应的影响。

肺炎链球菌(Streptococcus Pneumonia,SP)是儿童急性中耳炎、社区获得性肺炎、菌血症和化脓性脑膜炎的常见病原,该菌有90多个血清型,现有PVC7结合菌苗的广泛接种虽然能有效保护菌苗覆盖范围的SP感染,但SP在PVC7接种通过荚膜替换产生免疫逃逸,刚刚上市的PVC13和研究中的PCV15也远不足以覆盖所有血清型,且价格昂贵不利于在发展中国家普及。因此研究价格低廉的全覆盖SP菌苗是众多医学同仁努力的方向。新一代SP全覆盖菌苗将以非荚膜多糖为抗原,本课题组已研制出无荚膜SP全细胞疫苗(WCA),并经动物实验证实其能通过CD4+T细胞途径对鼻咽部SP定植产生稳定的保护,通过抗体途径对侵袭性SP疾病产生稳固的、可重复的保护,并已在人中启动I期临床试验。WCA疫苗有效,但产生作用重要保护作用的抗原有待进一步鉴定,以便为今后WCA疫苗的改进或优化奠定基础。目的筛选并鉴定SPWCA中的重要保护性抗原,并研究其在SP致病性中的作用。方法构建SP蛋白质的原核表达系统,人工表达SP胞外或包膜上的65种目标蛋白质并纯化;以SPWCA免疫新西兰大白兔后获得富含抗体血清,以抗WCA血清为一抗,以驴抗兔-IgG-HRP为二抗,采用El.SA法检测前述蛋白的抗体滴度;筛选出有效蛋白质后,敲除其基因,比较基因敲除前后SP(TIGR4)的致病性变化。结果以Freunds为佐剂的WCA疫苗免疫后,SP2070的抗体增加幅度最大,增加了2187倍,其次是SP1572,增加了243倍;其它增加40倍以上的还有SP0648和SP1032;以Al.minum为佐剂的WCA疫苗免疫后,抗体滴度增加13倍以上的有SP1572,SP0629、SP1032、SP0648、SP0191、SP2070。以SP2070和SP1572为研究目标,分别敲除肺炎球菌TIGR4和Tigr4:14中的相应基因并感染Bl.小鼠,结果发现,经腹腔受TIGR4Δ2070感染的20只小鼠无1例死亡,而受TIGR4野生型回复子感染者死亡率为55%;鼻腔定植试验显示TIGR4:14Δ2070的定植率较回复子显著下降。TIGR4和TIGR4:14经敲除1572基因后体外生存能力显著下降,其致病性处进一步研究中。结论 SP2070抗原可能是SPWCA疫苗有效的主要保护性抗原之一,对保护Bl.小鼠罹患侵袭性SP疾病和鼻咽部携带均有显著保护作用。

病人一旦被诊断为癌症,常需要初级保健医生将诊断告诉病人,与其讨论预后或做适当的转诊。传递坏消息时,按优先次序使病人了解关键的问题。讨论预后时,医生应对病人想了解多少信息量反应灵敏。在不给予假希望的前提下准确地讨论预后是一种挑战。所有为病人提供服务的医生对预后的观点要统一,避免给病人混淆不清的信息。根据病情进展,医生必须重新评价治疗的有效性。当治愈性治疗转为姑息性治疗时,医生应避免使用让病人感到治疗失败或被放弃的用语。医生要认识到文化因素会影响临终关怀的讨论。初级保健医生有机会与病人保持长期信赖关系,但在做临终关怀讨论时,他们还没有太多经验。采用系统方法有助于初级保健医生选择恰当方式讨论预后、提出现实性希望、提供治疗选项、协调疾病转换期、缓解病人痛苦(见表1)。

AIM To assess the performance of BAl.D, BAl.D-2 and their component biomarkers in predicting outcome of hepatocel.ul.r carcinoma(HCC) patients after l.ver *** BAl.D score and BAl.D-2 cl.ss are derived from bil.rubin, al.umin, al.ha-fetoprotein(AFP), l.ns cul.naris aggl.tinin-reactive AFP(AFP-l.), and des-gammacarboxyprothrombin(DCP). Pre-transpl.nt AFP, AFP-l. and DCP were measured in 113 patients transpl.nted for HCC from 2000 to 2008. Hazard ratios(HR) for recurrence and death were cal.ul.ted. Univariate and mul.ivariate regression anal.ses were conducted. C-statistics were used to compare biomarker-based to predictive model.. RESUl.S During a median fol.ow-up of 12.2 years, 38 patients recurred and 87 died. The HRs for recurrence in patients with el.vated AFP, AFP-l., and DCP defined by BAl.D cut-off val.es were 2.42(1.18-5.00), 1.86(0.98-3.52), and 2.83(1.42-5.61), respectivel.. For BAl.D, the HRs for recurrence and death per unit increased score were 1.48(1.15-1.91) and 1.59(1.28-1.97). For BAl.D-2, the HRs for recurrence and death per unit increased cl.ss were 1.45(1.06-1.98) and 1.38(1.09-1.76). For recurrence prediction, the combination of three biomarkers had the highest c-statistic of 0.66 vs. 0.64, 0.61, 0.53, and 0.53 for BAl.D, BAl.D-2, Mil.n, and UCSF, respectivel.. Simil.rl., for death prediction, the combination of three biomarkers had the highest c-statistic of 0.66 vs 0.65,0.61, 0.52, and 0.50 for BAl.D, BAl.D-2, Mil.n, and UCSF. A new model.combining biomarkers with tumor size at the time of transpl.nt(S-l.D) demonstrated the highest predictive capabil.ty with c-statistics of 0.71 and 0.69 for recurrence and death. CONCl.SION BAl.D and BAl.D-2 are val.d in transpl.nt HCC patients, but l.ss predictive than the three biomarkers in combination or the three biomarkers in combination with maximal.tumor diameter(S-l.D).
