肺癌是全球肿瘤发病率和病死率的主因,且预后很差。加强对肿瘤生物学的认识对肺癌研究至关重要。被誉为"下一代测序技术"的NGS技术(next-generation sequencing)是一种针对全基因组鉴定的有力工具,可以对致癌体细胞突变进行全面检测。大多数的NGS技术是基于平台特异性DNA文库进行多重聚合酶链反应(polymerase chain reaction,PCR),从而对目的基因扩增后测序。这种技术适用于高通量测序,可以检测出肿瘤中出现的全部基因组变异。缺点是这种技术需要在时间、实验设备、计算机数据分析、生物信息技术等各方面的大量投入。NGS技术已广泛应用于全基因组、外显子组、转录组和表观基因组的研究中,为肺癌研究和医疗模式带来改变。这项新技术的开展将转变当前对致癌信号通路的认识,可为癌症诊疗提供新的分子靶点。肺癌体细胞突变已有NGS技术的分析报道,但大规模基因组研究仍在进行中。个体化治疗策略将改善那些潜在获益患者的治疗方式,避免"无辜"患者受无效治疗带来的高额费用和不良反应。NGS的组织化、计算机化和生物信息化技术推动了科技的进步,同时,患者知情权和数据发布的相关伦理问题也浮现出来。信号通路中,驱动基因(driver gene)突变和传递基因(passenger gene)突变的区别,需要对测序结果进行细致解读。解读准确与否取决于DNA提取的样本类型、样本处理技术和样本含量。肿瘤异质性也会降低肿瘤基因突变的检测效能。NGS技术将推动对肿瘤基因突变的基础和临床研究,而且,也可应用于单细胞和游离的循环DNA,未来还将用于从体液和肿瘤亚群中获取的DNA样本。如果能进一步降低费用、提高检验速度和精度,NGS技术无疑将会成为肺癌研究的绝佳选择。

Capturing flue gases often require multiple stages of scrubbing, increasing the capital and operating costs. So far, no attempt has been made to study the absorption characteristics of all the three gases(NO, SO_(2) and CO_(2)) in a single stage absorption unit at alkaline p H conditions. We have attempted to capture all the three gases with a single wet scrubbing column. The absorption of all three gases with sodium carbonate solution promoted with oxidizers was investigated in a tall absorption column. The absorbance was found to be 100% for CO_(2), 30% for NO and 95% for SO_(2) respectively. The capture efficiency of sodium carbonate solution was increased by 40% for CO_(2) loading, with the addition of oxidizer. Absorption kinetics and reaction pathways of all the three gases were discussed individually in detail.

Marginal contact binary systems are an important class of binary systems in exploited by astronomers to understand various astrophysical phenomena as they bridge both semi-detached and overcontact *** studied the photometric data of CW Aqr using the JCB 1.3 m telescope(JCBT),Kepler,and TESS to understand the period variations and constrain the light curve *** noted that the period of CW Aqr is increasing with a mass transfer rate of m=7.53×10^(-8)M_(⊙)yr^(-1)from the secondary to the primary *** also ascertained cyclic deviations along with a parabolic trend in the O-C *** residuals displayed a sinusoidal trend which was fitted with an LTTE,indicating the presence of a third body with an orbital period of P_(3)~10 yr and e=*** fitted the JCBT,Kepler,and TESS photometric normalized light curves and noted the mass ratio to be q~0.32 for both contact and semi-detached *** identified a temperature difference between the primary and secondary components of~2500 *** activity signatures were found in the observed light curves which were also evident from the Hαline studies where the filled-in effect was not *** our study suggests that CW Aqr is a marginal contact binary with a low mass ratio evolving toward a semi-detached ***,it may merge if the period increases due to ongoing mass transfer accompanied by a lowering in the mass ratio and later oscillate back to the period-decreasing trend due to angular momentum loss over a few cycles of thermal relaxation oscillations followed by instability.

In post-earthquake surveys,it is difficult(and often infeasible)to observe and quantify displacements beyond line-of-sight(LOS),given seismic force-resisting and gravity systems exist completely or partially within a building′s *** overcome this limitation,we develop a novel framework that generalizes graph-based state estimation towards structural joint localization via engineered *** landmarks provide an indirect means to estimate residual displacements where direct LOS is *** our framework,engineered landmarks define topologies of uniquely identifiable landmarks that are either visible or non-visible to a robot performing simultaneous localization and mapping(SLAM).Within the SLAM approach,factors encoding robot odometry and robot-to-visible landmark measurements are formulated for the cases of wireless sensing and fiducial object detection and *** landmarks are rigidly attached to non-visible landmark subsets for each engineered landmark,where the complete set of non-visible landmarks form globally rigid and localizable connectivity graphs via range-based *** subsets of non-visible landmarks are embedded within the base structure and uniquely define joint pose via geometric *** factors are unified within a common graph to solve for the maximum a posteriori estimate of robot,landmark,and joint states via nonlinear least squares *** demonstrate the applicability of our approach,we apply the Monte Carlo method over a parameterization of system noise to calculate residual joint pose error distributions,maximum average inter-story drift ratios,and related summary statistics for a 19-story nonlinear structural *** performing nonlinear time history analyses over sets of service-level and maximum considered earthquakes,our parametric study gives insight into our method′s application towards post-earthquake building evaluation in non-LOS conditions.

The light curve solutions of two W UMa binary systems in the intermediate open cluster NGC 7789 are presented in this paper. These variables were observed using the 2 m telescope of the IUCAA-Girawali Observatory. The V passband photo- metric solutions of the two W UMa binaries were obtained using the latest version of the W-D program. The result shows that both systems are H-subtype W UMa binaries with high mass ratios.

Marginal short-period contact binaries are important to understand as they pose a different physical scenario than the predicted theoretical model based on the thermal relaxation oscillation mechanism due to their shallow degree of *** we present the optical and X-ray studies of a contact binary source RW Dor using the Transiting Exoplanet Survey Satellite(TESS)and XMM-Newton *** the first time we report the varying O'Connell effect and explain the asymmetry with a spot *** on the new times of minima,we make a robust estimate of the orbital period of the third body at 47.01~0.52 yr with an eccentricity e=*** show that the period-decreasing trend observed in O-C variation can be explained by both conservative mass transfer from primary to secondary and AML via stellar *** X-ray luminosity exhibited by RW Dor did not vary significantly on three different occasions and was found to be about 3.34×10~(29)erg s~(-1).Assuming that the quiescent X-ray emission is emitted from an undisturbed loop structure,the loop size is estimated to be 0.6-1×10~(10)cm which is≤Alfvén radius r_A~8×10~(10)cm.

The addition of cement for stabilization of expansive soils is one of the most commonly used *** with every calcium-based stabilizer,the time delay between the physical mixing of the stabilizer and compaction plays an important role in achieving the desired results after ***,a clear insight on the determination of optimum time delay for achieving the maximum desired compaction properties of cement-stabilized soils is yet to be ***,the recent studies highlighted the use of sulfate to mitigate the negative effect of compaction time *** only drawback with the use of sulfate along with calcium-based stabilizers is the formation of ettringite,which deteriorates the stabilized soil *** view of this,the present study is aimed at using the sulfate resistant cement(SRC)as a stabilizer along with the controlled addition of sulfate solutions to mitigate the negative effect of compaction time delay in stabilizing the expansive *** bring out the above effects,three periods of time delays(0 h,6 h and 24 h)and three sulfate concentrations of 5000 parts per million(ppm),10,000 ppm and 20,000 ppm were *** experimental results showed that the delay in compaction resulted in the formation of clogs and reduction of strength of SRC-stabilized expansive *** sulfate addition to SRC-stabilized expansive soil,the formation clogs was not curtailed and resulted in the formation of ettringite *** formations were captured with the help of scanning electron microscope(SEM)images and validated with electron dispersive X-ray spectroscopy(EDAX)***,an attempt is also made to explain the mechanism of density and strength reduction with the aid of physico-chemical properties and mercury intrusion porosimetry(MIP)studies.
