Perceptual and Sensory-Functional Consequences of Skin Care Products

Skin care products are often designed to provide tangible, physical benefits to skin health. Alleviation of dry skin and minimization of the signs of aging and post-injury scarring are important benefits targeted by many products on the market. Equally important to these benefits are favorable, desirable sensory attributes, without which products are unlikely to be used and repurchased. Other products are designed primarily to deliver sensory—or sensual—benefits (e.g., many cosmetic creams). This review considers the tactile sensory experience delivered by skin care products by examining: 1) their instrumentally-measured rheology and tribology;2) their influence on the skin’s mechanics (e.g., compliance);3) their implications for changing sensory function (e.g., tactile sensitivity);and 4) the possibility that skin care products alter their own perception. Products that contain chemosensates (e.g., capsaicin, menthol) or pharmaceutical actives are not considered here. Although numerous perceptual-physical links have been reported, formulation rules by which products can be designed for optimal skinfeels are currently unavailable from the existing literature. This is because of inconsistencies among studies in the perceptual attributes investigated, the physical characterizations chosen to describe the products, and analysis methods employed. To provide a robust method for designing products with beneficial and desirable skinfeels, we propose the use of 1) a consistent lexicon that fully describes the perceptual experience of any product investigated, 2) a means of recording the mechanical events at the fingertip skin that occur when a skin care product is manually applied to the body. This approach contrasts with previous instrumental (in vitro) methods that may not generalize well to product-treated human skin (in vivo). Regulation of intestinal permeability: The role of proteases

The gastrointestinal barrier is-with approximately 400 m^2-the human body's largest surface separating the external environment from the internal milieu. This barrier serves a dual function: permitting the absorption of nutrients, water and electrolytes on the one hand, while limiting host contact with noxious luminal antigens on the other hand. To maintain this selective barrier, junction protein complexes seal the intercellular space between adjacent epithelial cells and regulate the paracellular transport. Increased intestinal permeability is associated with and suggested as a player in the pathophysiology of various gastrointestinal and extraintestinal diseases such as inflammatory bowel disease, celiac disease and type 1 diabetes. The gastrointestinal tract is exposed to high levels of endogenous and exogenous proteases, both in the lumen and in the mucosa. There is increasing evidence to suggest that a dysregulation of the protease/antiprotease balance in the gut contributes to epithelial damage and increased permeability. Excessive proteolysis leads to direct cleavage of intercellular junction proteins, or to opening of the junction proteins via activation of protease activated receptors. In addition, proteases regulate the activity and availability of cytokines and growth factors, which are also known modulators of intestinal permeability. This review aims at outlining the mechanisms by which proteases alter the intestinal permeability. More knowledge on the role of proteases in mucosal homeostasis and gastrointestinal barrier function will definitely contribute to the identification of new therapeutic targets for permeability-related diseases.

Visceral hypersensitivity in inflammatory bowel diseases and irritable bowel syndrome: The role of proteases

Proteases, enzymes catalyzing the hydrolysis of peptide bonds, are present at high concentrations in the gastrointestinal tract. Besides their well-known role in the digestive process, they also function as signaling molecules through the activation of protease-activated receptors(PARs). Based on their chemical mechanism for catalysis, proteases can be classified into several classes: serine, cysteine, aspartic, metallo- and threonine proteases represent the mammalian protease families. In particular, the class of serine proteases will play a significant role in this review. In the last decades, proteases have been suggested to play a key role in the pathogenesis of visceral hypersensitivity, which is a major factor contributing to abdominal pain in patients with inflammatory bowel diseases and/or irritable bowel syndrome. So far, only a few preclinical animal studies have investigated the effect of protease inhibitors specifically on visceral sensitivity while their effect on inflammation is described in more detail. In our accompanying review we describe their effect on gastrointestinal permeability. On account of their promising results in the field of visceral hypersensitivity, further research is warranted. The aim of this review is to give an overview on the concept of visceral hypersensitivity as well as on the physiological and pathophysiological functions of proteases herein.

The frequency of co-infections with bacterial or fungal pathogens has constantly increased among critically ill patients with coronavirus disease 2019(COVID-19)during the *** was the most frequently reported invasive fungal *** onset of candidemia in COVID-19 patients was often delayed compared to non-COVID-19 ***,Candida invasive infections in COVID-19 patients were more often linked to invasive procedures(e.g.,invasive mechanical ventilation or renal replacement therapy)during the intensive care stay and the severity of illness rather than more“classic”risk factors present in patients without COVID-19(e.g.,underlying diseases and prior hospitalization).Moreover,apart from the increased incidence of candidemia during the pandemic,a worrying rise in fluconazole-resistant strains was reported,including a rise in the multidrug-resistant Candida *** outcomes,the development of invasive Candida co-infection had a negative impact,increasing morbidity and mortality compared to non-co-infected COVID-19 *** this narrative review,we present and critically discuss information on the diagnosis and management of invasive fungal infections caused by Candida *** critically ill COVID-19 patients.

Efficient and safe cell engineering by transfection of nucleic acids remains one of the long-standing hurdles for fundamental biomedical research and many new therapeutic applications,such as CAR T cell-based *** has recently gained increasing attention as a more safe and versatile alternative tool over viral-or DNA transposon-based approaches for the generation of adoptive T ***,limitations associated with existing nonviral mRNA delivery approaches hamper progress on genetic engineering of these hard-to-transfect immune *** this study,we demonstrate that gold nanoparticle-mediated vapor nanobubble(VNB)photoporation is a promising upcoming physical transfection method capable of delivering mRNA in both adherent and suspension *** transfection experiments on HeLa cells showed the importance of transfection buffer and cargo concentration,while the technology was furthermore shown to be effective for mRNA delivery in Jurkat T cells with transfection efficiencies up to 45%.Importantly,compared to electroporation,which is the reference technology for nonviral transfection of T cells,a fivefold increase in the number of transfected viable Jurkat T cells was ***,our results point toward the use of VNB photoporation as a more gentle and efficient technology for intracellular mRNA delivery in adherent and suspension cells,with promising potential for the future engineering of cells in therapeutic and fundamental research applications.

Subjects with blood type A had a higher PG Ⅱ level than subjects with other blood types. Both PG Ⅰ and Ⅱ levels rose with dally consumption of cigarettes. Alcohol consumption was not related to PG levels. The PG Ⅰ/Ⅱ ratio declined with increasing consumption of sour pancakes a fermented staple food found to contain N-nitroso compounds and to be a risk factor for stomach cancer in this population.

The reduced diameter of skeletal myofibres is a hallmark of several congenital myopathies,yet the underlying cellular and molecular mechanisms remain *** this study,we investigate the role of HACD1/PTPLA,which is involved in the elongation of the very long chain fatty acids,in muscle fibre *** humans and dogs,HACD1 deficiency leads to a congenital myopathy with fibre size disproportion associated with a generalized *** of HACD1-deficient Labradors,Hacd1-knockout mice,and Hacd1-deficient myoblasts,we provide evidence that HACD1 promotes myoblast fusion during muscle development and *** further demonstrate that in normal differentiating myoblasts,expression of the catalytically active HACD1 isoform,which is encoded by a muscle-enriched splice variant,yields decreased lysophosphatidylcholine content,a potent inhibitor of myoblast fusion,and increased concentrations of≥C18 and monounsaturated fatty acids of *** lipid modifications correlate with a reduction in plasma membrane *** conclusion,we propose that fusion impairment constitutes a novel,non-exclusive pathological mechanism operating in congenital myopathies and reveal that HACD1 is a key regulator of a lipid-dependent muscle fibre growth mechanism.
