Interplay between nitric oxide and VIP in CCK-8-induced phasic contractile activity in the rabbit sphincter of Oddi

AIM: the sphincter of Oddi (SO) plays an important role in delivery of bile into the duodenum. to establish whether vasoactive intestinal polypeptide (VIP) and nitric oxide (NO) were involved in phasic contractile activity of the rabbit SO stimulated by cholecystokinin-octapeptide (CCK-8). MEtHODS: Isolated SO muscle rings were cleaned of fat and mounted horizontally on two small L-shaped hooks one of which was connected to a force transducer for the measurement of isometric *** experiments were carried out in a thermostatically controlled (37±0.2℃) organ bath (5 mL) containing Krebs *** organ fluid was gassed with 95% O2 and 50 mL/L CO2 to keep the pH at 7.40±0.05. Contractile responses to CCK-8 (1 μmol/L) were evaluated in the presence and absence of NG-nitro-L-arginine (LNNA), an inhibitor of NO synthase (100 μmol/L), and (p-chloro-D-Phe6-Leu17)-VIP (VlPa, 30 μmol/L), a VIP receptor antagonist. RESULtS: CCK-8 stimulated the phasic activity of the SO. NO synthase inhibition increased the frequency and amplitude of contractions with a slight increase in developed tension. Pre-incubation with VlPa also attenuated this CCK-8 effect. the combined application of LNNA and VlPa abolished the phasic activity of the muscle rings with a marked increase in tension in response to CCK-8. CONCLUSION: VIP and NO together contribute to an increase in phasic activity of SO.

AIM:the rote of the sphincter of Oddi(SO)in ethanol (EtOH)-induced pancreatitis is *** aim was to characterise the effect of E-I-OH on basal and stimulated SO motility. MEtHODS:SOs removed from white rabbits were placed in an organ bath(Krebs solution,pH7.4,37℃).the effects of 2 mL/L,4 mL/L,6 mL/L and 8 mL/L of EtOH on the contractile responses of the sphincter were determined. SOs were stimulated with either 0.1 μmol/L carbachol,1 μmol/L erythromycin or 0.1 μmol/L cholecystokinin(CCK). RESULtS:EtOH at a dose of 4 mL/L significantly decreased the baseline contractile amplitude from 11.98±0.05 mN to 11.19±0.07 ***,no significant changes in the contractile frequency were ***(0.6%) significantly decreased both the baseline amplitude and the frequency compared to the control group(10.50±0.01 mN, 12.13±0.10 mN and 3.53±0.13 c/min,5.5±0.13 cycles(c)/min, respectively).Moreover,0.8% of EtOH resulted in complete relaxation of the ***(0.1 μmol/L)or erythromycin (1 μmol/L)stimulated the baseline amplitudes(by 82% and 75%,respectively)and the contractile frequencies (by 150% and 106%,respectively).In the carbachol or erythromydn-stimulated groups 2-6 mL/L of E-IOH significantly inhibited both the amplitude and the ***, a 4-5 min administration of 6 mL/L EtOH suddenly and completely relaxed the ***(0.1 μmol/L)stimulated the baseline amplitude from 12.37±0.05 mN to 27.40±1.82 mN within 1.60±0.24 *** this peak,the amplitude decreased to 17.17±0.22 mN and remained constant during the *** frequency peaked at 12.8±0.2 c/min, after which the constant frequency was 9.43±0.24 c/min throughout the rest of the *** at a dose of 4 mL/L significantly decreased the amplitude from 16.13±0.23 mN to 14.93±0.19 ***,no significant changes in the contractile frequency were *** at a dose of 6 mL/L inhibited both the amplitudes and the frequencies in the CCK-stimulated group,while 8 mL/L of E

AIM:In previous experiments we have demonstrated that by administering low doses of cholecystokinin-octapeptide (CCK-8),the process of regeneration following L-arginine (Arg)-induced pancreatitis is *** rats that were also diabetic(induced by streptozotocin,StZ),pancreatic regeneration was not *** aim of this study was to deduce whether the administration of exogenous insulin could in fact restore the hypertrophic effect of CCK-8 in diabetic-pancreatitic rats. MEtHODS:Male Wistar rats were used for the experiments. Diabetes mellitus was induced by administering 60mg/kg body mass of StZ intraperitoneally(i.p.),then,on d 8, pancreatitis was induced by 200mg/100 g body mass Arg *** at an interval of 1 *** animals were injected subcutaneously twice daily(at 7 *** 7 p.m.)with 1 μg/kg of CCK-8 and/or 2 IU mixed insulin(300g/L short- action and 700g/L intermediate-action insulin) for 14 d after pancreatitis *** this the animals were killed and the serum amylase,glucose and insulin levels as well as the plasma glucagon levels,the pancreatic mass/body mass ratio(pm/bm),the pancreatic contents of DNA,protein,amylase,lipase and trypsinogen were *** tissue samples were examined by light microscopy on paraffin-embedded sections. RESULtS:In the diabetic-pancreatitic rats treatment with insulin and CCK-8 significantly elevated pw/bm and the pancreatic contents of protein,amylase and lipase vs the rats receiving only CCK-8 ***-8 administered in combination with insulin also elevated the number of acinar cells with mitotic activities,whereas CCK-8 alone had no effect on laboratory parameters or the mitotic activities in diabetic-pancreatitic rats. Background Proper parent-child interaction is crucial for child development, but an assessment tool in Chinese is currently lacking. this study aimed to develop and validate a parent-reported parent-child interaction scale for Chinese preschool children. Methods the Chinese parent-child interaction scale (CPCIS) was designed by an expert panel based on the literature and clinical observations in the Chinese context. the initial CPCIS had 14 parent-child interactive activity items. Psychometric properties of the CPCIS were examined using the Rasch model and confirmatory factor analysis (CFA). Convergent validity was investigated by the associations between CPCIS and family income, maternal education level, and children's school readiness. Results the study recruited 567 Chinese parent-child pairs from diverse socioeconomic backgrounds, who completed the CPCIS. Six out of the 14 items in the initial CPCIS were dropped due to suboptimal fi t values. the refined 8-item CPCIS was shown to be valid and reliable by Rasch models and CFA. the person separation reliability and Cronbach's α of the CPCIS were 0.81 and 0.82, respectively. the CPCIS scores were positively associated with family's socioeconomic status (η2 = 0.05,P ternal education level (η2 = 0.08,P tered, valid, and reliable tool for the assessment of parent-child interactions in Chinese families.

针对固定间隙的上海深紫外自由电子激光(SDUV-FEL)混合型波荡器的端部,用Rad ia程序进行了模拟计算。在端部不加任何电磁线圈补偿的情况下,通过减小端部磁铁、磁极的体积和变动端部磁极位置的方法对波荡器磁场进行了优化,优化以后波荡器横向磁场的边缘场强度降到5×10-4t(距离端部磁块边缘10 mm处),边缘场波形没有了明显突起,优化后的横向磁场的一次积分曲线和二次积分曲线都有很大改善,端口处的一次积分值、二次积分值接近于零。

慈善行为既出自人们对弱者和贫困者同情的善良意志,反映了人性的光辉,也是整个社会文明程度的体现。慈善事业的发展既受到政府鼓励和推进,也需要全社会民众的积极参与。目前,官办慈善组织在组织公益慈善活动方面起了主导作用(甚至是支配作用),但近日诸多公共事件的发生使这一体制遭到许多批评,并使慈善议题成为大众舆论的焦点。因此,在有关慈善事业发展的研究中,我们既要考察目前慈善事业发展中政府所起的作用,更要思考如何调动社会组织和广大民众的积极性来共同参与。分析我国的官办慈善体制所存在的问题,思考我们如何推动民间慈善和私人慈善活动的发展,使慈善事业得到全面发展,是十分有必要的。

Magnetic Properties of Nd-Fe-B Sintered Magnet Powders Recovered by Yb Metal Vapor Sorption

Fine ground powders of Nd-Fe-B sintered magnet bulks (particle size=46～125 μm in diameter) were coated and alloyed with Yb metal by sorbing them. A significant recovery of the decreased magnetic properties of the ground powders (remanence B r=～0.95 t, coercivity H cj =～227 ka·m -1 and maximum energy product (BH) max=～48 8 kJ·m -3) was observed in accordance with increasing temperature up to 800 ℃. the sorbing temperature and time for Yb metal vapor were optimized and after heating at 800 ℃ for 90 min and annealing subsequently at 610 ℃ for 60 min, the B r, H cj and (BH) max values were increased to be 0.98 t, 712 ka·m -1 and 173 kJ·m -3, respectively. From the microstructural characterizations of resulting samples by using X-ray diffraction (XRD), scanning electron microscopy (SEM), and electron probe X-ray microanalyzer (EPMA), it is found that the sorbed Yb metal uniformly covers the surface and diffuses to the Nd-rich grain boundary of fine ground powders of Nd-Fe-B sintered magnet bulks forming a (Nd,Yb)Fe 2 phase.

Although lanthanide double-decker complexes with hetero-macrocyclic ligands as functional luminescent and magnetic materials have promising properties, their inferior water solubility has negated their biomedical applications.[1] Herein, four water-soluble homoleptic lanthanide（Ln = Gd, Er, Yb and La） sandwiches with diethylene-glycol-disubstituted porphyrins（DD） are reported, with their structures proven by both quantum chemical calculations and scanning tunneling microscopy. Our findings demonstrate that the near-infrared emission intensity and singlet oxygen（1 O2） quantum yields of YbDD and GdDD in water are higher than those of the reported capped lanthanide monoporphyrinato analogues, YbN and GdN;the brightness and luminescence lifetime in water of YbDD are greater than those of YbN. this work provides a new dimension for the future design and development of molecular theranostics-based water-soluble double-decker lanthanide bisporphyrinates.[2]

the Permian Basin is a unique ecosystem located in the southwest of the *** unanswered question is whether the bacteria in the Permian Basin adapted to the changing paleomarine environment and survived in the remnants of Permian *** our previous study,a novel bacterial strain,Permianibacter aggregans HW001t,was isolated from micro-algae cultures incubated with Permian Basin waters,and was shown to originate from the Permian *** this study,strain HW001t was shown to be the representative strain of a novel family,classified as‘Permianibacteraceae’.the results of molecular dating suggested that the strain HW001t diverged~447 million years ago(mya),which is the early Permian period(~250 mya).Genome analysis was used to access its potential energy utilization and biosynthesis capacity.A large number of transporters,carbohydrate-active enzymes and protein-degradation related genes have been annotated in the genome of strain *** addition,a series of important metabolic pathways,such as peptidoglycan biosynthesis,osmotic stress response system and multifunctional quorum sensing were annotated,which may confer the ability to adapt to vari-ous unfavorable environmental ***,the evolutionary history of strain HW001t was reconstructed and the horizontal transfer of genes was predicted,indicating that the adaptation of *** to a changing marine environment depends on the evolution of their metabolic capabilities,especially in signal *** conclusion,the results of this study provide genomic information for revealing the adaptive mechanism of strain HW001t to the changing ancient oceans.

Reduction of tat-interacting Protein 30 Expression Could be a Prognostic Marker in Bladder Urothelial Cancer

Background： tat-interacting protein 30 （tIP30） has been reported to be a tumor suppressor, with reduced or absent expression in various tumors. However, its role in bladder urothelial cancer （BUC） has not been investigated. therefore, herein, we investigated the expression of t1P30 protein in BUC and normal bladder mucosa and the clinical significance of tIP30 expression in the prognosis of BUC. Methods： We reviewed data from 79 cases of BUC and 15 adjacent tissue samples from 79 patients treated at our institution between 2004 and 2007. tIP30 expression was examined by immunohistochemistry. the relationship between tIP30 expression and tumor stage, histological grade, and survival was analyzed. Differences between groups were evaluated using the t-test or matched-pairs test, and differences in the survival rates were analyzed with the log-rank test. Results： tIP30 protein expression was significantly reduced in BUC tissue （t = -6.91, P 〈 0.05） compared with normal tissue samples, and in invasive bladder cancer （t = 10.89, P 〈 0.05） compared with superficial bladder cancer. tIP30 protein expression differed significantly among different differentiated groups classified either according to the World Health Organization （2004, F = 17.48, P 〈 0.01） or World Health Organization （1973, F = 10.68, P 〈 0.01）. tIP30 protein expression was significantly reduced in high-grade papillary urothelial carcinoma compared with papillary urothelial neoplasm of low malignant potential （P 〈 0.05） and low-grade papillary urothelial carcinoma （P 〈 0.05）. Meanwhile, tIP30 protein expression was significantly reduced in Grade 111 BUC, compared with Grade I （P 〈 0.05） and Grade 11 （P 〈 0.05）. Patients with low t1P30 expression showed a higher incidence of disease progression than those with high tIP30 expression （t - 2.63, P 〈 0.05）. kaplan-Meier survival analysis showed a strong positive relationship between tIP30 expression and overall survival （OS）（x^2 = 17.29, P 〈 0.05）.
