Immuolocalization of nestin in pancreatic tissue of mice at different ages

AIM： To localize nestin positive cells （NPC） in pancreatic tissue of mice of different ages. METHODS： Paraffin sections of 6-8 um of fixed pancreatic samples were mounted on poly-L-lysine coated slides and used for Immunolocalization using appropriate primary antibodies （Nestin, Insulin, glucagon）, followed by addition of a fluorescently labeled secondary antibody. The antigen-antibody localization was captured using a confocal microscope （Leica SP 5 series）. RESULTS： In 3-6 d pups, the NPC were localized towards the periphery of the endocrine portion, as evident from immunolocalization of insulin and glucagon, while NPC were absent in the acinar portion. At 2 wk, NPC were localized in both the exocrine and endocrine portions. Interestingly, in 4-wk-old mice NPC were seen only in the endocrine portion, towards the periphery, and were colocalised with the glucagon positive cells. In the pancreas of 8- wk-old mice, the NPC were predominantly localized in the central region of the islet clusters, where immunostaining for insulin was at a maximum. CONCLUSION： We report for the first time the immunolocalization of NPC in the pancreas of mice of different ages （3 d to 8 wk） with reference to insulin and glucagon positive cells. The heterogeneous localization of the NPC observed may be of functional and developmental significance and suggest（s） that mice pancreatic tissue can be a potential source of progenitor cells. NPC from the pancreas can be isolated, proliferated and programmed to differentiate into insulin secreting cells under the appropriate microenvironment.

The precise detection and measurement of dopamine（DA）,a crucial neurotransmitter in the human body,plays a significant role in diagnosing,preventing,and treating neurological diseases associated with its levels.A highly sensitive DA electrochemical sensor was constructed by combining molybdenum disulfide quantum dots（MSQDs） with multiwalled carbon nanotubes（MWCNTs）.The MSQDs were synthesized using the shear exfoliation *** sensors consist of MSQDs with Mo-S edge catalytic centers for the DA redox reaction,and MWCNTs amplify the sensor *** linearity of the sensor for the detection of DA was tested in the presence of ascorbic acid(AA,50 μmol·L-1) and uric acid(UA,200 μmol·L-1),and exhibited linearity from 2 to 966 μmol·L-1of DA with 0.097 μA(mol·L-1)-1sensitivity and a low limit of detection of0.6 μmol·L-1（the ratio between signal and noise,S/N=3）.Moreover,the sensitivity and selectivity of the sensor were also studied using *** is no increase in amperometric current after adding the most potentially interfering *** sensor was successfully applied to recover DA in human blood sera ***,machine learning algorithms were operated to aid in the near-precise detection of DA in the heterogeneous mixture containing AA and *** algorithms facilitate the identification and quantification of DA amidst coexisting interferents,including AA,that are commonly present in biological matrices.

Organic solar cells(OSCs) hold great potential as a photovoltaic technology for practical ***, the traditional experimental trial-and-error method for designing and engineering OSCs can be complex, expensive, and time-consuming. Machine learning(ML) techniques enable the proficient extraction of information from datasets, allowing the development of realistic models that are capable of predicting the efficacy of materials with commendable accuracy. The PM6 donor has great potential for high-performance OSCs. However, it is crucial for the rational design of a ternary blend to accurately forecast the power conversion efficiency(PCE) of ternary OSCs(TOSCs) based on a PM6 ***, we collected the device parameters of PM6-based TOSCs and evaluated the feature importance of their molecule descriptors to develop predictive models. In this study, we used five different ML algorithms for analysis and prediction. For the analysis, the classification and regression tree provided different rules, heuristics, and patterns from the heterogeneous dataset. The random forest algorithm outperforms other prediction ML algorithms in predicting the output performance of PM6-based TOSCs. Finally, we validated the ML outcomes by fabricating PM6-based TOSCs. Our study presents a rapid strategy for assessing a high PCE while elucidating the substantial influence of diverse descriptors.

Bio-inspired helicoidal composite laminates, inspired by the intricate helical structures found in nature, present a promising frontier for enhancing the mechanical properties of structural designs. Hence, this study provides a comprehensive investigation into the nonlinear free vibration and nonlinear bending behavior of bio-inspired composite plates. The inverse hyperbolic shear deformation theory(IHSDT) of plates is employed to characterize the displacement field, with the incorporation of green-Lagrange nonlinearity. The problem is modeled using the C0finite element method(FEM), and an in-house code is developed in the MATLAB environment to solve it numerically. Various helicoidal layup configurations including helicoidal recursive(HR), helicoidal exponential(HE), helicoidal semi-circular(HS), linear helicoidal(LH), and Fibonacci helicoidal(FH)with different layup sequences and quasi-isotropic configurations are studied. The model is validated, and parametric studies are conducted. These studies investigate the effects of layup configurations, side-to-thickness ratio, modulus ratios, boundary conditions, and loading conditions at different load amplitudes on the nonlinear vibration and nonlinear bending behaviors of bio-inspired composite plates. The results show that the laminate sequence exerts a substantial impact on both nonlinear natural frequencies and nonlinear bending behaviors. Moreover, this influence varies across different side-to-thickness ratios and boundary conditions of the bio-inspired composite plate.

To describe an open surgical technique for transplanting fresh ovarion tissue to the anterior abdominal wall at the incision site and to determine whether ovarian function would be restored after transplantation. Case study. Academic medical center. A 44- year- old patient who underwent an operation for uterine fibroids. Microsurgical reconstruction of ovarian cortex and its transplantation to the anterior abdominal wall at incision site of Pfannenstiel. Follicular development evident by ultrasound examination; restoration of serum FSH and LH levels to nonmenopausal range; and disappearance of menopausal symptoms. Early postoperative FSH, LH, and E2 levels showed that menopause was confirmed. Postoperative hormone levels at months 2, 3, and 6 were as follows: FSH: 77.86, 79.50, and 13.70 mIU/mL; LH: 34.60, 33.92, and 8.78 mIU/mL; E2: 29, 46, and 48 pg/mL. The patient is still followed up for postmenopausal status. Autotransplantation of cortical strips to the anterior abdominal wall at the incision site without further inci sion can be a logical solution for the patients at early climacteric age.

A non-autonomous complex ginzburg-Landau equation （CgLE） for the finite amplitude of convection is derived, and a method is presented here to determine the amplitude of this convection with a weakly nonlinear thermal instability for an oscillatory mode under throughflow and gravity modulation. Only infinitesimal disturbances are considered. The disturbances in velocity, temperature, and solutal fields are treated by a perturbation expansion in powers of the amplitude of the applied gravity field. Throughfiow can stabilize or destabilize the system for stress free and isothermal boundary conditions. The Nusselt and Sherwood numbers are obtained numerically to present the results of heat and mass transfer. It is found that throughfiow and gravity modulation can be used alternately to heat and mass transfer. Further, oscillatory flow, rather than stationary flow, enhances heat and mass transfer.

Hedgerows with intercropping systems were established at the ICIMOD test and demonstration site at godawari to assess the effective- ness of Sloping Agricultural Land Technology （SALT） in reducing run- off water volume, controlling soil loss, increasing crop production, and improving soil fertility in the mid-hills of Nepal. Runoff water volume （1996-2002）, soil loss （1996-2002） and maize yield （1995-2001）, and soil fertility-related parameters were assessed on SALT models with three factors： the type of nitrogen-fixing plant, the farmers＇ practice, and fertilizer use. Results showed a significant effect of Alnus nepalensis and/or Indigofera dosua on runoff water volume, soil loss, crop produc- tion, soil water retention, and soil nutrients （NPK）. Farmers＇ practice and fertilization did not play a significant role in reducing runoff water and soil loss. However, farmers＇ practice significantly increased crop produc- tion. Therefore, integrating soil conservation approaches on SALT sys- tems enhances stable economic output to hills and mountain farmers.

Microphotosynthetic power cells(μPSCs)generate power through the exploitation of living photosynthetic’microorganisms by harvesting *** thermodynamic limitations of this process restrict the power output of a single ***,we demonstrateμPSCs in four different array configurations to enhance power output from these power *** this effect,sixμPSCs were arrayed in series,parallel,and combinations of series and parallel *** pPSC was injected with a 2 mL liquid culture of photosynthetic microorganisms(Chlamydomonas reinhardtii)in the anode and 2 mL of 25%(w/v)electron acceptor potassium ferricyanide(K_(3)Fe(CN)_(6))in the *** combinations ofμPSCs connected in series and parallel'generated higher power than the individual series and parallel *** combinations of sixμPSCs connected in series and in parallel produced a high power density of 1914 mWm^(-2)in the presence of white fluorescent light illumination at 20μEm^(-2)s^(-1).Furthermore,to realize the array strategy for real-time applications,a 1.7V/2mA rating light-emitting diode(LED)was powered by combinations of series and parallel array *** results indicate the reliability ofμPSCs to produce electricity from photosynthetic microorganisms for low-power *** addition,the results suggest that a combination of microlevel photosynthetic cells in array format represents a powerful optimal design strategy to enhance the power output fromμPSCs.
