Expression of apoptosis-related factors in chronic cyclosporine nephrotoxicity after cyclosporine withdrawal

AIM: To examine whether the reversibility of chronic cyclosporine A (CsA) nephrotoxicity is associated with apoptotic cell death and its regulatory factors. METHODS: Chronic CsA nephrotoxicity was induced in Sprague- Dawley rats by administering CsA (15 mg/kg, sc) for 5 weeks, and then withdrawing it for 5 or 10 weeks. The effect of CsA withdrawal on apoptotic cell death was evaluated by an in situ TdT-mediated deoxyuridine triphos- phate-biotin nick end-labeling (TUneL) assay and the expression of pro-apoptotic [(transforming growth factor- beta1 (TGF-β1) and Fas] and anti-apoptotic [epidermal growth factors (EGF) and Bcl-2] factors. RESULTS: Discontinuation of CsA induced significant decreases in TUneL-positive cells in a time-dependent manner and the reduction in TUneL-positive cells was correlated with the tubulointerstitial fibrosis score (r=0.919, Pneys was decreased significantly after with- drawal of CsA. In contrast, downregulated EGF and Bcl-2 expression returned to normal or supernormal levels. CONCLUSION: CsA withdrawal is associated with a decrease in apoptotic cell death and with changes in the expression of pro-apoptotic and anti-apoptotic molecules involved in renal wound repair. This may constitute one of the mechanisms underlying the reversibility of chronic CsA nephrotoxicity.

Quantitative visualization of Taylor-Couette flow with helical protrusion using optical and ultrasound imaging method

The simplified flow of drilling process in the soil hardening or oil rig site was experimentally investigated. Two flow models were used. One is the concentric cylinders with helical protrusion at the inner cylinder. The other is the concentric plain wall cylinders with axial flow. The radius ratio and aspect ratio of both models are the same with 0. 65 and 48,respectively. The mud is the typical fluid seen in the soil hardening processes. We used the water and the mud for the working fluid. We used the optical PIV for the water flow measurement and echo PIV for the mud flow. In case of the water study,the dominant vortical structures appeared in both the protrusion and plain model with axial *** case of mud flow,the vortices shown in the water model disappeared and the push and pull-up motion by the protrusion is dominant. We believe this information can be useful to understand the flow physics of drilling process in the complex fluid flow.

Denitrification Rates and Their Controlling Factors in Streams of the Han River Basin with Different Land-Use Patterns

Land-use patterns can affect various nutrient cycles in stream ecosystems, but little information is available about the effects of urban development on denitrification processes at the watershed scale. In the presented study, we investigated the controlling factors of denitrification rates within the streams of the Han River Basin, Korea, with different land-use patterns, in order to enhance the effectiveness of water resource management strategies. Ten watersheds were classified into three land-use patterns （forest, agriculture and urban） using satellite images and geographic information system techniques, and in-situ denitrification rates were determined using an acetylene blocking method. Additionally, sediment samples were collected from each stream to analyze denitrifier communities and abundance using molecular approaches. In-situ denitrification rates were found to be in the order of agricultural streams （289.6 mg N20-N m-2 d-1）〉 urban streams （157.0 mg N20-N m-2 d-1）〉 forested streams （41.9 mg N20-N m-2 d-l）. In contrast, the average quantity of denitrifying genes was the lowest in the urban streams. Genetic diversity of denitrifying genes was not affected by watershed land-use pattern, but exhibited stream-dependent pattern. More significance factors were involved in denitrification in the sites with higher denitrification rates. Multiple linear regression analysis revealed that clay, dissolved organic carbon and water contents were the main factors controlling denitrification rate in the agricultural streams, while dissolved organic carbon was the main controlling factor in the urban streams. In contrast, temperature appeared to be the main controlling factor in the forested streams.

Utilizing gastric cancer organoids to assess tumor biology and personalize medicine

While the incidence and mortality of gastric cancer (GC) have declined due to public health programs, it remains the third deadliest cancer worldwide. For patients with early disease, innovative endoscopic and complex surgical techniques have improved survival. However, for patients with advanced disease, there are limited treatment options and survival remains poor. Therefore, there is an urgent need for more effective therapies. Since novel therapies require extensive preclinical testing prior to human trials, it is important to identify methods to expedite this process. Traditional cancer models are restricted by the inability to accurately recapitulate the primary human tumor, exorbitant costs, and the requirement for extended periods of development time. An emerging in vitro model to study human disease is the patient-derived organoid, which is a three-dimensional system created from fresh surgical or biopsy tissues of a patient’s gastric tumor. Organoids are cultured in plastic wells and suspended in a gelatinous matrix, providing a substrate for extension and growth in all dimensions. They are rapid-growing and highly representative of the molecular landscape, histology, and morphology of the various subtypes of GC. Organoids uniquely model tumor initiation and growth, including steps taken by normal stomach cells to transform into invasive, intestinal-type tumor cells. Additionally, they provide ample material for biobanking and screening novel therapies. Lastly, organoids are a promising model for personalized therapy and warrant further investigation in drug sensitivity studies for GC patients.

Helicobacter pylori eradication therapy for functional dyspepsia: Systematic review and meta-analysis

AIM: To evaluate whether Helicobacter pylori (H. pylori) eradication therapy benefits patients with functional dyspepsia (FD).METHODS: Randomized controlled trials (RCTs) investigating the efficacy and safety of H. pylori eradication therapy for patients with functional dyspepsia published in English (up to May 2015) were identified by searching PubMed, EMBASE, and The Cochrane Library. Pooled estimates were measured using the fixed or random effect model. Overall effect was expressed as a pooled risk ratio (RR) or a standard mean difference (SMD). All data were analyzed with Review Manager 5.3 and Stata ***: This systematic review included 25 RCTs with a total of 5555 patients with FD. Twenty-three of these studies were used to evaluate the benefits of H. pylori eradication therapy for symptom improvement; the pooled RR was 1.23 (95%CI: 1.12-1.36, P nefit of H. pylori eradication therapy on quality of life with an SMD of -0.01 (95%CI: -0.11 to 0.08, P = 0.80). Six studies demonstrated that H. pylori eradication therapy reduced the development of peptic ulcer disease compared to no eradication therapy (RR = 0.35; 95%CI: 0.18-0.68, P = 0.002). Eight studies showed that H. pylori eradication therapy increased the likelihood of treatment-related side effects compared to no eradication therapy (RR = 2.02; 95%CI: 1.12-3.65, P = 0.02). Ten studies demonstrated that patients who received H. pylori eradication therapy were more likely to obtain histologic resolution of chronic gastritis compared to those who did not receive eradication therapy (RR = 7.13; 95%CI: 3.68-13.81, P < 0.00001).CONCLUSION: The decision to eradicate H. pylori in patients with functional

A Parametric Study for High-Efficiency Gas-Liquid Separator Design

A gas liquid centrifugal separator is widely used in industry on account of its simple geometry and little maintenance. These separators have considerable advantages over filters, scrubbers or precipitators in term of compact design, lower pressure drop and higher capacity. A gas liquid centrifugal separator is a device that utilizes centrifugal forces and low pressure caused by rotational motion to separate liquid from gas by density differences. Efficient and reliable separation is required for the optimum operation. These separators are often operated at less than peak efficiency due to the entrainment of separated liquid through an outlet pipe which is closely associated with the very complicated flow phenomena involved. Design parameters such as length of the separation space, vane exit angle, inlet to outlet diameter ratio, models for separation efficiency and pressure drop as a function of physical dimensions are not available in literature. This leaves the designer with very little to go on except known designs and experimentation. The aim of present study is to perform a parametric study to get higher efficiency for gas-liquid separator. A parametric study has been carded out with the help of CFD tools to analyze a separation performance of a centrifugal separator by varying the length of separator space. The best design parameters are analyzed based upon obtained results, tangential velocities, vortices, total pressure losses. From the present study several attempts are made to improve the performance of conventional centrifugal separators.

Three metabolites from an entomopathogenic bacterium, Xenorhabdus nematophila, inhibit larval development of Spodoptera exigua （Lepidoptera： Noctuidae） by inhibiting a digestive enzyme, phospholipase A2

An entomopathogenic bacterium, Xenorhabdus nematophila, has been known to induce significant immunosuppression of target insects by inhibiting immune-associated phospholipase A2 （PLA2）, which subsequently shuts down biosynthesis of eicosanoids that are critical in immune mediation in insects. Some metabolites originated from the bacterial culture broth have been identified and include benzylideneacetone, proline-tyrosine and acetylated phenylalanine-glycine-valine, which are known to inhibit enzyme activity of PLA2 extracted from hemocyte and fat body. This study tested their effects on digestive PLA2 of the beet armyworm, Spodoptera exigua. Young larvae fed different concentrations of the three metabolites resulted in significant adverse effects on larval development even at doses below 100 #g/mL. In particular, they induced significant reduction in digestive efficiency of ingested food. All three metabolites significantly inhibited catalytic activity of digestive PLA2 extracted from midgut lumen of the fifth instar larvae at a low micromolar range. These results suggest that the inhibitory activities of the three bacterial metabolites on digestive PLA2 of S. exigua midgut may explain some of their oral toxic effects.

Enhanced thermoelectric performance of spark plasma sintered p-type Ca_(3-x)Y_(x)Co_(4)O_(9+δ)systems

Highly textured dense Ca3-xYxCo4 O9+δ(0≤x≤0.3)samples were fabricated by combining sol-gel process with spark plasma sintering(SPS).Y^3+substitution for Ca^2+simultaneously increased the Seebeck coefficient and reduced the thermal *** latter was attributed to the increase in lattice anharmonicity,structural distortion,and grain boundary area,which enhanced the phonon ***2.7Y0.3Co4 O9+δshowed the largest dimensionless figure-of-merit(ZT=0.194)at 1073 K because it had the largest Seebeck coefficient and the lowest thermal *** ZT value was 55%larger than that of undoped Ca3 Co4 O9(0.125 at 1073 K).Therefore,Y^3+substitution,sol-gel powder synthesis,and SPS are highly effective for enhancing the thermoelectric prope rties of Ca_(3)Co_(4)O_(9).
