We have previously demonstrated that 8-Opioid receptor(DOR) plays a crucial role in neuroprotection under both normoxia as well as hypoxic *** hypoxic/ischemic injury to the brain has been recognized as one of the causes of Parkinson’s disease andα-synuclein causes neuronal injury in the Parkinson’s brain,we hypothesized that DOR may paly a role in antiparkinson *** this work,we studied the effects of DOR activation and inhibition on cultured HEK293 cells under hypoxia(1%O and 0.5%O) and/or MPP *** injury was measured by lactate dehydrogenase release assay,Methanethiosulfonate(MTS) assay and JC-1 *** results show that 1) MPP induces cytotoxic effect by enhancingα-synuclein overexpression and the formation of toxicα-synuclein oligomers.2) 48h hypoxia at 1%O concentration and 24h hypoxia at 0.5%O concentration,were sufficient to demonstrate cytotoxicity viaα-synuclein overexpression and the formation of toxicα-synuclein *** the other hand,24h hypoxia at 1%O concentration did not alterα-synuclein expression,and nor did it induce formation of *** was rather observed to enhance the viability of these cells.3) A DOR agonist,UFP-512(5μM) treatment markedly attenuated cellular injury andα-synuclein overexpression induced by 48h hypoxia at 1%O concentration or 24h hypoxia at 0.5%O concentration;4) UFP-512 treatment did not alter mitochondrial membrane potential(△Ψm) in intact cells,although the DOR agonist had a significant cytoprotective effect on the cells exposed to hypoxia or MPP.5) UFP-512 up-regulated the phosphorylation of cAMP-response element binding protein(CREB) in the DOR-protected cells.6) The observed cytoprotective effects of DOR activation were abolished after the treatment with a DOR antagonist(Naltrindole).These findings suggest that the activation of DOR attenuatesα-synuclein mediated cell injury induced by MPP under normoxia as well as hypoxic ***,these observations open new avenue

The hepatic endoplasmic reticulum(ER)-anchored cytochromes P450(P450s)are mixedfunction oxidases engaged in the biotransformation of physiologically relevant endobiotics as well as of myriad xenobiotics of therapeutic and environmental relevance.P450 ER-content and hence function is regulated by their coordinated hemoprotein syntheses and proteolytic *** P450 proteolytic turnover occurs through a process known as ER-associated degradation(ERAD)that involves ubiquitindependent proteasomal degradation(UPD)and/or autophagic-lysosomal degradation(ALD).Herein,on the basis of available literature reports and our own recent findings of in vitro as well as in vivo experimental studies,we discuss the therapeutic and pathophysiological implications of altered P450 ERAD and its plausible clinical *** specifically(i)describe the P450 ERAD-machinery and how it may be repurposed for the generation of antigenic P450 peptides involved in P450 autoantibodypathogenesis in drug-induced acute hypersensitivity reactions and liver injury,or viral hepatitis;(ⅱ)discuss the relevance of accelerated or disrupted P450-ERAD to the pharmacological and/or toxicological effects of clinically relevant P450 drug substrates;and(ⅲ)detail the pathophysiological consequences of disrupted P450 ERAD,contributing to non-alcoholic fatty liver disease(NAFLD)/non-alcoholic steatohepatitis(NASH)under certain synergistic cellular conditions.

The Hansen solubility parameters(HSP)are frequently used for solvent selection and characterization of polymers,and are directly related to the suspension behavior of pigments in solvent *** performance of currently available group contribution(GC)methods for HSP were evaluated and found to be insufficient for computer-aided product design(CAPD)of paints and coatings.A revised and,for this purpose,improved GC method is presented for estimating HSP of organic compounds,intended for organic *** to the significant limitations of GC methods,an uncertainty analysis and parameter confidence intervals are provided in order to better quantify the estimation accuracy of the proposed *** to other applicable GC methods,the prediction error is reduced significantly with average absolute errors of 0.45 MPa^(1/2),1.35 MPa^(1/2),and 1.09 MPa^(1/2) for the partial dispersion(δD),polar(δP)and hydrogen-bonding(δH)solubility parameters respectively for a database of 1106 *** performance for organic pigments is comparable to the overall method performance,with higher average errors forδD and lower average errors forδP andδH.

AIMTo determine the effect of pH, osmolality, and buffering system on the oxygen permeability (dk) of soft contact *** hydrogel lenses (nelfilcon A and etafilcon A) and 2 silicone hydrogel lenses (lotrafilcon A and balafilcon A) were used in the study. These lenses were incubated in phosphate-buffered saline (PBS) and borate-buffered saline (BBS) solutions adjusted by 0.8 pH increments to a pH in the range of 5.8-9.0 or in hypotonic (280 mOsmol/kg), isotonic (310 mOsmol/kg) and hypertonic (380 mOsmol/kg) PBS solutions. Polarographic method was used for measuring the dk and lenses were stacked as 4 layers to correct the boundary *** values of all contact lenses measured in BBS solutions were more stable than those in PBS solutions. Especially the etafilcon A lens showed a relative big change compared with other types of contact lenses at the same conditions. When the osmolality of PBS solution increased from hypotonic to hypertonic, dk of all contact lenses decreased. Variations in dk existed depending on lens materials, etafilcon A lens was the most affected and nelfilcon A was the least affected by *** the result obtained, it is revealed that dk of contact lenses is changed by the pH, osmolality, and buffering condition of tear. Thus, dk of contact lens can be varied by the lens wearers' physiological and/or pathological conditions.
