Mature osteoclasts degrade bone matrix by exocytosis of active proteases from secretory lysosomes through a ruffled ***,the molecular mechanisms underlying lysosomal trafficking and secretion in osteoclasts remain largely ***,we show with GeneChip analysis that RUN and FYVE domain-containing protein 4(RUFY4)is strongly upregulated during *** lacking Rufy4 exhibited a high trabecular bone mass phenotype with abnormalities in osteoclast function in ***,deleting Rufy4 did not affect osteoclast differentiation,but inhibited bone-resorbing activity due to disruption in the acidic maturation of secondary lysosomes,their trafficking to the membrane,and their secretion of cathepsin K into the extracellular ***,RUFY4 promotes late endosome-lysosome fusion by acting as an adaptor protein between Rab7 on late endosomes and LAMP2 on primary ***,Rufy4-deficient mice were highly protected from lipopolysaccharide-and ovariectomy-induced bone ***,RUFY4 plays as a new regulator in osteoclast activity by mediating endo-lysosomal trafficking and have a potential to be specific target for therapies against bone-loss diseases such as osteoporosis.

Sleep Posture Classification Using RGB and Thermal Cameras Based on Deep Learning Model

Sleep posture surveillance is crucial for patient comfort,yet current systems face difficulties in providing compre-hensive studies due to the obstruction caused by *** posture assessment remains challenging because of the complex nature of the human body and variations in sleep ***,this study introduces an innovative method utilizing RGB and thermal cameras for comprehensive posture classification,thereby enhancing the analysis of body position and *** method begins by capturing a dataset of sleep postures in the form of videos using RGB and thermal cameras,which depict six commonly adopted postures:supine,left log,right log,prone head,prone left,and prone *** study involves 10 participants under two conditions:with and without ***,the database is normalized into a video *** subsequent step entails training a fine-tuned,pretrained Visual Geometry Group(VGG16)and ResNet50 *** the third phase,the extracted features are utilized for *** fourth step of the proposed approach employs a serial fusion technique based on the normal distribution to merge the vectors derived from both the RGB and thermal ***,the fused vectors are passed to machine learning classifiers for final *** dataset,which includes human sleep postures used in this study’s experiments,achieved a 96.7%accuracy rate using the Quadratic Support Vector Machine(QSVM)without the ***,the Linear SVM,when utilized with a blanket,attained an accuracy of 96%.When normal distribution serial fusion was applied to the blanket features,it resulted in a remarkable average accuracy of 99%.

Tracking of horizontal alignment of the long and large diameter uncharged hole in NATM tunnel

The long and large diameter uncharged hole boring(LLB)method is a cut blasting method that minimizes blast-induced vibrations by creating long and large diameter uncharged holes at the excavation face of tunnels prior to tunnel *** in this method typically uses a 50 m long with a 382 mm diameter hammer bit in the horizontal direction at the tunnel ***,the significant weight and uni-directional rotation of the rod head,as well as variables such as geological characteristics,machine conditions,and inexperienced operators result in significant deviation from the target borehole alignment that hinders the vibration-dampening effect of the uncharged ***,since there is no method to verify the alignment of the boreholes until main tunnel construction,borehole misalignment is often not discovered until weeks after construction,which requires tunnel construction to cease until the equipment can be remobilized and an additional borehole be created,causing significant delays and increased costs for the entire tunnel *** this study,the borehole alignment tracking and ground exploration system(BGS)is developed to predict and monitor the quality and alignment of boreholes for cut blasting methods such as the LLB methods immediately after *** BGS was subsequently tested at a subway construction site to evaluate its performance in the *** measurements yielded by the BGS were compared with manually measured boring positions at every 5 m along the *** the BGS showed a maximum deviation of approximately 12%at a local point where the hole surface was relatively rough,the accuracy for the final boring position was approximately 97%,demonstrating excellent precision of the alignment tracking *** BGS demonstrates excellent performance in predicting ground conditions and the boring quality of a cut hole immediately after drilling,and shows promise in various other applications for monitoring borehole alignment.

This review article provides overall understanding of stainless,environment-friendly,and nonflammable Mg alloys(SEN alloys)recently developed at the Korea Institute of Materials *** alloys are produced by adding small amounts of Ca and Y(eachilar to those of commercial AZ91 alloys.(5)Simultaneous introduction of Ca and Y suppresses the formation of Mg17Al12 discontinuous precipitates during aging,leading to the enhanced elongation of aged SEN alloys.(6)Adding mischmetal into the SEN9 alloy leads to a six-fold enhancement in ***,the studies conducted on SEN alloys demonstrate their excellent ignition and corrosion resistances and mechanical properties,which broaden the industrial applications of Mg alloys by addressing their inherent weaknesses.

Increasing concentrations of dietary threonine,tryptophan,and glycine improve growth performance and intestinal health with decreasing stress responses in broiler chickens raised under multiple stress conditions

The current study aimed to compare the effects of increasing concentrations of dietary threonine(Thr),tryptophan(Trp),and glycine(Gly)on growth performance,stress biomarkers,and intestinal function in broiler chickens under multiple stress *** hundred sixty broiler chickens at 21 d old were randomly allotted to 5 treatments with 8 *** in a positive control(PC)treatment were raised under low stock density(16.9 birds/m2 per cage)with recommended environmental conditions,whereas birds in 4 treatments were subjected to multiple stress conditions:a cyclic heat stress of 30±0.3°C for 10 h and 23±0.2°C for 14 h per day with high stock density(25.3 birds/m2 per cage).A basal diet was assigned to both PC and negative control(NC)*** additional diets were individually formulated to contain double concentrations of digestible Thr,Trp,or Gly+Ser compared with their concentrations in the basal *** experiment lasted for 14 *** showed that NC treatment had less growth performance(Piler chickens with decreasing stress response under multiple stress conditions.

A Transfer Function Design for Medical Volume Data Using a Knowledge Database Based on Deep Image and Primitive Intensity Profile Features Retrieval

Direct volume rendering(DVR)is a technique that emphasizes structures of interest(SOIs)within a volume visually,while simultaneously depicting adjacent regional information,e.g.,the spatial location of a structure concerning its *** DVR,transfer function(TF)plays a key role by enabling accurate identification of SOIs interactively as well as ensuring appropriate visibility of *** generation typically involves non-intuitive trial-and-error optimization of rendering parameters,which is time-consuming and *** at mitigating this manual process have led to approaches that make use of a knowledge database consisting of pre-designed TFs by domain *** these approaches,a user navigates the knowledge database to find the most suitable pre-designed TF for their input volume to visualize the *** these approaches potentially reduce the workload to generate the TFs,they,however,require manual TF navigation of the knowledge database,as well as the likely fine tuning of the selected TF to suit the *** this work,we propose a TF design approach,CBR-TF,where we introduce a new content-based retrieval(CBR)method to automatically navigate the knowledge *** of pre-designed TFs,our knowledge database contains volumes with SOI *** an input volume,our CBR-TF approach retrieves relevant volumes(with SOI labels)from the knowledge database;the retrieved labels are then used to generate and optimize TFs of the *** approach largely reduces manual TF navigation and fine *** our CBR-TF approach,we introduce a novel volumetric image feature which includes both a local primitive intensity profile along the SOIs and regional spatial semantics available from the co-planar images to the *** the regional spatial semantics,we adopt a convolutional neural network to obtain high-level image feature *** the intensity profile,we extend the dynamic time warping technique to address subtle alignment
