The fungal Umsrt1 and maize ZmsUT1 sucrose tra.s.orters battle for plant sugar resources

The biotrophic fungus Ustilago maydis ca.s.s corn smut disease, inducing tumor formation in its host Zea ma.s. Upon infection, the fungal hyphae invaginate the plasma membrane of infected maize ceils, establishing an interface where pathogen and host are separated only by their plasma membranes. At this interface the fungal and maize sucrose tra.s.orters, Umsrtl and ZmsUT1, compete for extracellular sucrose in the corn smut/maize pathos- ystem. Here we biophysically characterized ZmsUT1 and Umsrtl in Xenopus oocytes with respect to their voltage-, pH- and substrate-dependence and determined affinities toward protons and sucrose. In contrast to ZmsUT% Umsrtl has a high affinity for sucrose and is relatively pH- and voltage-independent. Using these quantitative parameters, we developed a mathematical model to simulate the competition for extracellular sucrose at the contact zone between the fungus and the host plant. This approach revealed that Umsrtl exploits the apoplastic sucrose resource, which forces the plant tra.s.orter into a.s.crose export mode providing the fungus with sugar from the phloem. Importantly, the high sucrose concen- tration in the phloem appeared disadvantageous for the ZmsUT1 preventing sucrose recovery from the apoplastic space in the fungus/plant interface.

Dear Editor, The plant hormone a.s.isic acid （ABA） governs drought stress management in plants. ABa.s.gnal tra.s.uction appea.s.to be rather short. When under water stress con- ditions, the ABA level rises above a certain threshold and the phytohormone binds to its cytosolic RCAR/PYR/PYL-type receptor （Levchenko et al,, 2005; Joshi-saha et al., 2011）. Following receptor activation, protein phosphatases of the clade-A PP2C type are inactivated, allowing snRK protein kinases to phosphorylate their targets, such as bZIP tran- scription factors （Fujii et al., 2009 and references therein）. In guard cells and oocytes, we could demonstrate that the same snRK-PP2C pair OsT1-ABI1 controls the opening of the slow anion channel sLACl （Geiger et al., 2009; Lee et al., 2009）. Besides this calcium-independent activation, sLAC1 is a.s. activated by the calcium-dependent kinases CPK21 and 23, which can be inhibited by ABI1 （Geiger et al., 2010）.

Phylogenetic investigation on five genera of tintinnid ciliates (Ciliophora, Choreotrichia), based on the small subunit ribosomal RNA gene sequences

The phylogenetic relationship among tintinnid ciliates are relatively poorly studied based on molecular data. In the present work, seven species belonging to five genera of the order Tintinnida (Amphorellopsis acuta, Codonellopsis nipponica, Favella taraikaensis, steno-semella nivalis, Tintinnopsis beroidea, Tintinnopsis cylindrica and Tintinnopsis lohmanni) were analyzed using the information on their small subunit ribosomal RNA gene sequences. Phylogenetic trees were constructed using Bayesian inference (BI), maximum pa.s.mony (MP), neighbor-joining (NJ), and least-squares (Ls) methods. Generally, similar topologies were revealed with high or moderate supports, in which the main results show that (1) all tintinnids analyzed belong to a.s.ngle a.s.mblage; (2) congeners in Tintinnopsis do not cluster together, which indicates that the lorica-based definition for this genus is not consistent with the ssU rRNA phylogeny; (3) A. acuta groups with Tintinnidium mucicola but not with Eutintinnus, indicating that the traditional family Tintinnidae might be a paraphyletic group; (4) stenosemella and Codonellopsis are clearly most related and possibly even merged into one genus regarding their similar morphology and molecular analyses, and possession of a hyaline collar is the only characteristic of the genus.

The well-known ciliate, Mesodinium stein, 1863, is of great importance to marine microbial food webs and is related to the ＂red tides＂. However, it is possibly one of the most confusing ciliate taxa in terms of its systematic position： either the morphological or the molecular data excluded it from all the other known a.s.mblages or groups. In the current work, the sequences of small subunit ribosomal RNA （ssU rRNA） genes for all isolates available are analysed and an examination of the secondary structure patterns of related groups is carried out. The results indicate that （1） Mesodiniurn invariably represents a completely separated and isolated clade positioned between two subphyla of ciliates with very deep branching, which indicates that they should be a primitive or ancestral group for the subphylum Intramacronucleata; （2） the secondary structure of the ssU rRNA of Mesodinium species is unusual in that, while the secondary structure of V4 in Mesodinium sp. has the deletions common to all litostome ciliates, it has more extensive deletions in helix E23_8 and a longer helix E23_1; （3） combining the phylogenetic and morphological information, we suggest establishing Mesodiniea el. nov., including the order Mesodiniida Grain, 1994, belonging to the subphylum Intramacronucleata.

Dear Editor, It has been known since the work of Francis Darwin that, in response to a reduction in atmospheric relative humidity （rh）, stomatal aperture decreases. screening for Arabidopsis mutants compromised in stomatal responses to reduced rh resulted in the identification of two genes, OsT1 and ABA2, that are involved in stomatal response to low rh conditions. Interestingly both encode proteins previously known to be involved in ABa.s.gnaling （Xie et al., 2006, and references therein）. These findings strongly suggested that, at least in part, the stomatal response to low rh is mediated by ABA and the intracellular ABa.s.gnaling pathway. Ciliates in the subcla.s.Hypotrichia have long been difficult to cla.s.fy as they are one of the most polymorphic and highly differentiated groups,leading to their systematics remaining *** relationships within the hypotrich family strongylidiidae have been ambiguous due to discordance between the morphological and genetic *** this study,a new strongylidiid genus Heterouroleptus is established,mainly based on the novel mode of origin of the ventral cirral rows:left ventral cirral row(LVR)originates from frontal-ventral-tra.s.erse cirral anlagen(FVTA)Ⅲ(anterior portion),IV(middle portion),and V(rear portion);right ventral cirral row comes from the entire FVTA VI.A new species,Hetero-uroleptus weishanensis ***.,***.,is investigated along with the morphometric and molecular data from a population of strongylidium *** new sequences and nuclear gene markers(single-gene and multi-gene)are provided to analyze the phylogenetic relationships of strongylidiids,with the COI gene utilized to uncover further genetic information at species level and *** results reveal that:(1)strongylidiidae is monophyletic and has a close relationship with Dorsomarginalia;(2)Heterouroleptus *** a clade that is sister to all the other strongylidiids;(3)Hemiamphisiella Foissner,1988 and Pseudouroleptus Hemberger,1985 should not be synonyms,and both genera.s.ould be subdivided due to their variable morphological characteristics;(4)LVR originating from three anlagen is a plesiomorphy of *** discovery of the origin of the LVR not only contributes to the establishment of the genus Heterouroleptus,but a.s. helps to improve the diagnosis of the family strongylidiidae.

Defamiliarizing Reality for Mental and Physical subjection:Rereading Harold Pinter's The Room and The Dumb Waiter as Political Comedies

While Pinter's earliest pla.s.have been recognized in the modernist history of theatre as comedies of menace and his later pla.s.as political comedies,this article argues that his earliest pla.s.are equally very liable to be interpreted as political *** of their a.s.rdist dramatization of people's helpless exposure to external,unidentifiable threa.s.a common post-WWII characteristic feature of human experience,I claim that The Room and The Dumb Waiter(both written 1957,staged 1960),two model examples of Pinter's earliest oeuvres,do not simply follow the a.s.hetic of a.s.rdist theatre to express human *** audience's experience of viewing the theatrical performances of both pla.s.in terms of discursive cyclicality or character normality is subverted into one of changeability,strangeness,and *** foreground the political implications of such revolutionary theatrical experience,Pinter's pla.s.are examined in the light of his unique use of defamiliarization,relying not on Brecht's traditional techniques of singing,dancing,image-projecting,or captioning,but on a.s.mple,dual technique of image destruction and *** consists of divesting characters of their normality and portraying them instead as individua.s.who identify only with unusual images of place,time,body,and *** this special technique of defamiliarization,both pla.s.are examined to reveal Pinter's central political theme of undermining reality for purposes of mental and physical subjections.

Developing a Model for Parkinson’s Disease Detection Using Machine Learning algorithms

Parkinson’s disease(PD)is a chronic neurological condition that progresses over *** start to have trouble speaking,writing,walking,or performing other basic skills as dopamine-generating neurons in some brain regions are injured or *** patient’s symptoms become more severe due to the worsening of their signs over *** this study,we applied state-of-the-art machine learning algorithms to diagnose Parkinson’s disease and identify related risk *** research worked on the publicly available dataset on PD,and the dataset consists of a.s.t of significant characteristics of *** aim to apply soft computing techniques and provide an effective solution for medical professiona.s.to diagnose PD *** research methodology involves developing a model using a machine learning *** the model selection,eight different machine learning techniques were adopted:Namely,Random Forest(RF),Decision Tree(DT),support Vector Machine(sVM),Naïve Bayes(NB),Light Gradient Boosting Machine(LightGBM),K-Nearest Neighbours(KNN),Extreme Gradient Boosting(XGBoost),and Logistic Regression(LR).subsequently,the concentrated models were validated through 10-fold Cross-Validation and Receiver Operating Characteristic(ROC)—Area Under the Curve(AUC).In addition,GridsearchCV was utilised to measure each algorithm’s best parameter;eventually,the models were trained through the hyperparameter tuning *** 98%accuracy,LightGBM had the highest accuracy in this ***,KNN,and sVM came in second with 96%***,the performance scores of NB and LR were recorded to be 76%and 83%,*** is to be mentioned that after applying 10-fold cross-validation,the average performance score of LightGBM accounted for 93%.At the same time,the percentage of ROC-AUC appeared at 0.92,which indicates that this LightGBM model reached a.s.tisfactory ***,we extracted meaningful insights and figured out potential ga.s.on top of *** extracting meaningful in
