An improved LC–MS/MS method for the quantification of alverine and para hydroxy alverine in human plasma for a bioequivalence study

A highly sensitive and selective high performance liquid chromatography–tandem mass spectrometry method was developed and validated for the quantification of alverine（ALV） and its active metabolite, para hydroxy alverine（PHA）, in human plasma. For sample preparation, solid phase extraction of analytes was performed on Phenomenex Strata-X cartridges using alverine-d5 as the internal standard. The analytes were separated on Symmetry Shield RP18（150 mm×3.9 mm, 5 μm） column with a mobile phase consisting of acetonitrile and10 mM ammonium formate（65：35, v/v）. detection and quantitation was done by electrospray ionization mass spectrometry in the positive mode using multiple reaction monitoring. The assay method was fully validated over the concentration range of 15.0–15,000 pg/mL for ALV and 30.0–15,000 pg/mL for PHA. The intra-day and inter-day accuracy and precision（% CV） ranged from 94.00% to 96.00% and 0.48% to 4.15% for both the analytes. The mean recovery obtained for ALV and PHA was 80.59% and 81.26%, respectively. Matrix effect,expressed as IS-normalized matrix factor ranged from 0.982 to 1.009 for both the analytes. The application of the method was demonstrated for the specific analysis of ALV and PHA for a bioequivalence study in 52 healthy subjects using 120 mg ALV capsules. The assay reproducibility was also verified by reanalysis of 175 incurred subject samples.

Managing Knowledge in Shared Spaces

Information has always been an integral component of business strategy. However, with the rise of information technology (IT) and globalization of businesses, information has taken a far more important role in explaining business performance. IT can be used to enhance research activities of the firm;and its product knowledge can be used to improve business productivity. In fact, scholars and researchers have indicated that knowledge can be a source of sustainable advantages for the firm. However, not much research has been done that explains what kind of knowledge is important for sustainable advantages. In this paper, we present three kinds of knowledge: core, complementary, and peripheral. We also develop an idea of shared space along three dimensions: physically shared space, technically shared space, and cognitively shared space. The main theme of this paper is to show the relevance and importance of these different kinds of knowledge along the shared spaces that create sustainable advantages for the firm.

Optimization of Threshold Ranges for Rapid Flood Inundation Mapping by Evaluating Backscatter Profiles of High Incidence Angle SAR Images

Rapid satellite-based flood inundation mapping and delivery of flood inundation maps during a flood event can provide crucial information for planners and decision makers to prioritize relief and rescue operations. The present study is undertaken to optimize the threshold ranges for the classification of flood water in Synthetic Aperture Radar(SAR) images(of 20° to 49° incidence angles) for quick flood inundation mapping and response during flood disasters. This is done through assessing the signature of flood water in Horizontal transmit and Horizontal received(HH), Horizontal transmit and Vertical received(HV), Vertical transmit and Horizontal received(VH), and Vertical transmit and Vertical received(VV) polarization radar data. The mean backscattering signature profiles of various water bodies were analyzed to discriminate flood water from other water bodies. The study shows that there is better demarcation of land-water surface in HH polarization. VV polarization has the potential to identify partially submerged features, which can be useful in flood damage assessments. The backscatter of flood water in HV and VH is the same and both HV and VH polarizations are adequate for the mapping of flood water. At near range to far range,-8 to-12 d B,-15 to-24 d B, and-6 to-15 d B can be used as optimum ranges for the classification of flood water in HH, HV, and VV polarizations. These optimum threshold ranges can be applied to the automation of flood mapping using SAR images in near-real time, where much time was often spent on finding the thresholds in order to produce flood inundation maps in a short time from the onset of flood disasters and deliver such maps to the concerned agencies.

A simple,rapid and sensitive ultra performance liquid chromatography-tandem mass spectrometry（UPLC-MS/MS） method has been developed for the simultaneous determination of cilostazol and its pharmacologically active metabolite 3,4-dehydro cilostazol in human plasma using deuterated analogs as internal standards（ISs）.Plasma samples were prepared using solid phase extraction and chromatographic separation was performed on UPLC BEH C18（50 mm × 2.1 mm.1.7 μm） *** method was established over a concentration range of 0.5-1000 ng/mL for cilostazol and 0.5-500 ng/mL for 3.4-dehydro ***- and inter-batch precision（%CV） and accuracy for the analytes were found within 0.93-1.88 and 98.8-101.7% for cilostazol and 0.91-2.79 and 98.0-102.7% for the metabolite *** assay recovery was within 95-97% for both the analytes and internal *** method was successfully applied to support a bioequivalence study of 100 mg cilostazol in30 healthy subjects.

The antioxidative,antimicrobial,and anti-inflammatory properties of fermented sheep milk with *** (KGL4),as well as the generation of antioxidative and antimicrobial peptides,are evaluated in the *** and antibacterial activities in sheep milk fermented with KGL4 increased with incubation hours,along with varied antioxidative properties (ABTS assay:35.12%,hydroxyl free radical scavenging assay:29.12%,superoxide free radical scavenging activity:36.38%).Antimicrobial activity of fermented sheep milk against *** (19 mm),*** (15.67 mm),*** (14 mm),and *** (13 mm) was also ***,after 48 h,the KGL4 showed maximum proteolysis (10.40 mg/ml) at 2.5% rate of addition sheep *** antioxidative and antimicrobial activities of fermented sheep milk fractions (3 kda and 10 kda permeates and retentates) were also *** highest ABTS activity (26.90%) was found in 10 kda permeate,whereas the 3 kda retentate had higher hydroxyl free radical scavenging activity (69.20%) and 3 kda permeate showed maximum superoxide free radical scavenging activity (32.85%).The 10 kda retentate had shown maximum antimicrobial activity against *** (13.67 mm) and *** (17.00 mm).Proteins spotted on 2d gel electrophoresis of KGL4 were varied from 10 to 70 ***-LC/MS was used to identify 5 novel peptide sequences from 2-d gel *** in the BIOPEP database confirmed the antioxidative and antimicrobial effects of the novel fermented sheep milk *** sheep milk with KGL4 (SMKGL4) significantly reduced excessive TNF-α,IL-6,& IL-1β production in LPS-activated RAW 264.7 cells.

建筑物震后损伤评估是地震灾害损失评估、震后恢复和重建工作的重要内容之一。建筑物震后损伤评估要现场找出结构损伤或破坏位置并对其进行分类统计和分析,它不仅提供了有关任何结构中损坏构件的数量和类别的重要信息,还帮助我们发现导致损坏的可能的结构缺陷。传统的损伤评估技术包括由工程师或专家对每个受损结构进行目视检查,这种检查方式不仅危险、耗时,而且需要大量熟练人力。基于计算机视觉的结构损伤识别技术已经成为结构监测和检测领域中一个极为先进和创新的研究方向,并逐渐受到了土木工程研究人员的关注。计算机视觉技术的主要优点是非接触、远距离、高精度、更大的覆盖范围以及可通过移动设备和无人机进行识别。本论文主要研究更易受地震影响的砌体结构的损伤评估。首先,本论文对卷积神经网络（CNN）理论进行了回顾和讨论,分析了迁移学习,超参数调试,卷积可视化等算法。同时,对砌体结构墙体在地震中可能发生的多类损伤（如对角裂缝、拐角分离、分层和平面外破坏等）进行了分类。其次,本文提出了一种通过在Image Net数据集上预先训练的VGG16模型上应用微调和转移学习而构建的深度CNN模型架构。梯度加权类激活映射（GradCAM）技术用于解决损坏的定位问题。最后,将本论文的方法应用于砌体结构墙体的损伤评估,对具有不同网络参数的多个模型在训练、测试和验证集结果进行了对比分析,并给出了基于GradCAM注意力机制的损伤定位图。结果表明,所提出模型的整体性能在表征和定位损伤方面表现出良好的一致性和鲁棒性。论文的结果证明了该模型在实际应用中的可行性。

Zinc supplementation as an adjunct to standard therapy in childhood nephrotic syndrome-a systematic review

AIM To evaluate the role of zinc as add on treatment to the "recommended treatment" of nephrotic syndrome(NS)in *** All the published literature through the major databases including IVledline/Pubmed,Embase,and Google Scholar were searched till 31 st december *** lists from the articles were reviewed to identify additional pertinent *** papers concerning the role of zinc in childhood NS were reviewed by the authors,and the data were extracted using a standardized data collection *** trials(RCTs) comparing zinc vs placebo was *** of zinc was studied in both steroid sensitive and steroid dependent/frequent relapsing *** primary outcome measure was the risk of relapse in 12 *** secondary outcome measures were mean relapse rate per patient in 12 mo,mean relapse rate per patient in 6 mo,risk of infection associated relapse in 12 mo,cumulative dose of steroids in two groups,mean length of time to next relapse,adverse effects of therapy,and change in serum zinc *** Of 54 citations retrieved,a total of 6 RCTs were *** was used at a dose of 10-20 mg/d,for the duration that varied from 6-12 *** to placebo,zinc reduced the frequency of relapses,induced sustained remission/no relapse,reduced the proportion of infection episodes associated with relapse with a mild adverse event in the form of metallic *** GRAdE evidence generated was of "very low-quality".CONCLUSION Zinc may be a useful additive in the treatment of childhood *** evidence generated mostly was of "very low-quality".We need more good quality RCTs in different country setting as well different subgroups of children before any firm recommendation can be made.

Genomic analysis of a novel strain of Bacillus nealsonii, isolated from Surti buffalo rumen

Aim: Whole genome sequencing and functional annotation of Bacillus nealsonii strain AAU1, an amylolytic anaerobic spore forming isolate from ruminal contents of buffalo. Methods and Results: Morphologically, the strain was observed as slender Grampositive rods, occurring in pairs. Optimal growth was observed at 40°C (range: 30°C to 45°C) and pH 6.5 (range: 5.5 to 7.5) when cultivated in Hungate’s medium supplemented with starch. The microorganism showed extracellular constitutive amylolytic activity, proving to be capable of utilizing glucose, maltose, mannose, trehalose, dextrin and starch under anaerobic conditions. Sequence analysis revealed a GC content of 35.1 mol%. Comparison of housekeeping gene sequences for RNA polymerase subunit B (rpoB) and gyrase A (gyrA) identified sequence similarity within the Bacillus genus, confirmed by 16S rRNA gene sequence similarity which identified Bacillus nealsonii dSM 15077 as the closest publically available relative. Chemotaxonomic analysis provided conflicting results with straight-chain saturated C16: 0/C16:0 aldehyde, C16:0 dMA, C14:0 and monounsaturated 16:1w7c and 16:1w9c the major fatty acids in contrast to those reported for B. nealsonii dSM15077. Further characterization using AN-Bilog and physiological parameters provided genotypic and phenotypic support for taxonomic classification of isolate AAU1 with published Bacillus species including B. licheniformis, B. subtilis, B. circulans and B. nealsonii. Conclusion: Based on the data presented, isolate is likely to represent a new strain/subspecies, for which the identifier B. nealsonii AAU1 is proposed. Significance and Impact of Study: The strict anaerobic conditions prevailing in the bovine rumen from where AAU1 was isolated may have resulted in genetic polymorphism influencing its metabolic characteristics.
