＜正＞ Paeonol （Pae）, a naturally occurring agent extracted from Moutan cortex, has shown great promise in the antitumor activity. The aim of this study was to investigate the effect of Pae and its combination with cisplatin （CDDP） on cell growth and apoptosis of human hepatoma cell lines HepG2 and SMMC - 7721. The cytotoxic effect of drugs on the two cell lines was measured by MTT assay. The interaction of Pae and CDDP was evaluated by coefficient of drag interaction. Morphologic changes were observed by acridine orange fluorescence staining. Cell cycle progression and apoptotic rate were detected by flow cytometry. The results indicated that Pae and CDDP had cytotoxic effect on the two cell lines in a dose -dependent manner. Pae combined with various concentrations of CDDP showed synergistically cytotoxic and apoptosis - inducing effect on the two cell lines. And the interaction between Pae and CDDP was specific to each cell line. Additionally, a combination of Pae with CDDP resulted in a stronger G2/M arrest, compared to these agents alone in the two cell lines. Pae may be effective and useful as a new biochemical modulator in chemotherapy.

＜正＞ G proteins are partners of G - protein - coupled receptors （GPCRs）. GPCRs catalyze guanine nucleotide exchange on Gα subunits,enabling both activated Gα and Gβγ subunits to target downst ream effector. Diverse extracellular signals regulate receptors to modulate cellular physiology. GPCRs signaling via heterotrimericG proteins is attenuated rapidly by G protein - coupled receptor kinase （GRK）. GPCRs phosphorylation is to promote the binding of arrestin proteins which block interactions of receptors and G - proteins. Regulators of Gprotein signaling are GTPase - activating proteins that attenuate signaling by G proteins. G proteins-AC- cAMP signal transduction play a crucial role in pathogenesis of immune arthritis. Gαs mRNA. protein express and function were decreased, and Gai mRNA. protein express and function of were increased in synoviocytes of rats with immune arthritis. The " cross - talk" was found between MAPK signal transduction and G proteins associated signal transduction. Activation of MAPKs was regulated by Gαi and Gαs signal transduction pathway. G proteins transmembrane signal pathway became new target for treatment of arthritis arthritis.

＜正＞ Shaoqiduogan（SQDG） is a compound produced from Radix Paeonia Pall and Radix Astragali. This study was aimed to examine the effect of SODG on human albumin induced immunological liver fibrosis in rats. The hyaluronic acid（HA） and procollagenⅢ（PCⅢ） were assessed by radioimmunoassay. SQDG decreased HA. PC Ⅲ,hydroxyproline content and improved the histological appearance of the liver sections. SQDG reduced lipid peroxidation and restored activities of *** vitro,SQDG raised the matrix metalloproteinase 13（MMP- 13） level and reduced the tissue inhibitors of metallop roteinase l（TIMP-1） level in HSC - T6 cell stimulated by transforming growth facor - betal （TGF -β1）. The expression of Gαi and Gas on HSC - T6 cell membrane induced by TGF -β1 were detected by Western - blot analysis. SQDG inhibited expression of Gαi2 and elevated expression of Gαs. Moreover. SQDG promoted expression of MMP - 13. inhibited the expression of TIMP - 1 and collagen - Ⅰ. These results indicated that SQDG may facilitate the collagen degradation of HSC - T6 induced by TGF -β1 via elevating the MMP - 13 /TIMP - 1 ratio and controlling the expression of Gai and Gαs.

＜正＞ To determine the serum concentration and pharmacokinetic paramaters of igurati-mod,32 healthy volunteers were divided into four groups. The groups received three single oral doses （25,50 or 75 mg） and one multiple oral doses （50 mg） of iguratimod, respectively. Serum concentration of igurattimod were measured by HPLC. The pharmacokinetic data were fit to a one - compartment model with first - order absorption. After single doses（25. 50 and 75 mg） of iguratimod, the following pharmacokinetic parameters were calculated, respectively: Tmax3.38±0.92. 4.88±1.96 and 4.33±1.00 h;Cmax 1.24±0.22, 2.13±0.54 and 3. 59±0.67mg/L;AUC20.93±4.24, 34.89±10.02 and 56.81±8.02;t1/28. 55±3.01, 6.31±3.15 and7.30±2.94 h. After multiple oral doses （50 mg） of iguratimod, the following pharmacokinetic parameters were calculated: Tmax 3. 63±1.60h;Cmax1.88±0.31 mg/L;AUC 31.88±4.52 mg h/L;t1/2 10.25±7.17 h. Iguratimod exhibited linear kinetics across oral doses of 25, 50, and 75 mg. There were no serious adverse events, and iguratimod was well tolerated over the entire dose range.

＜正＞ Total glucosides of paeony（TGP）is an active compound extracted from roots of paeonia lactiflora Pall. Our previous study showed that TGP has anti - hepatic injury and anti - hepatofibrotic effects. In the present study, the effects of TGP on the changes of the expressions of G - protein and G - protein - AC - cAMP signal transduction pathways stimulated by platelet - derived growth factor BB （PDGF- BB） in hepatic stellate cells （HSC） were investigated. The changes of the expression of G - protein on HSC - T6 cell membrane induced by PDGFBB （10μg·L-1）were detected by Western- blot analysis. The results showed that PDGF - BB remarkably increased the expression of Gαi2,but had no effect on expression of Gαi1、Gαi3 and Gαs. PDGF - BB decreased the level of cAMP with concentration - dependently. Furthermore, the tendency of cAMP was closely related with the proliferation of HSC - T6. The expression of Gαi2 was remarkably inhibited by TGP,which also increased the level of cAMP,and inhibited the proliferation of HSC - T6. The results indicate that TGP may inhibit the proliferation of HSC - T6 induced by PDGF - BB via regulating G - protein - AC - cAMP pathway.

Interleukin-1 receptor antagonist intervenes in signaling between different types of synoviocytes in rats with adjuvant arthritis

Aim:To investigate the mechanisms of interleukin-1 receptor antagonist （IL-1ra）in the treatment of adjuvant arthritis （AA）.Methods:AA was induced in rats bytreatment with Freund's complete adjuvant （FCA）.Rats were given an intracuta-neous injection of IL-1ra （2.5,10,40mg/kg,3 times per day） from d 14 to d 21 *** proliferation and the activity of IL-1 were determinedby using MTT *** necrosis factor alpha （TNF-α） and prostaglandin E2（PGE2） concentrations were measured by *** ultrastructureof synoviocytes was observed by using a transmission electron *** of c-Jun N-terminal kinase （JNK）,extracellular regulating kinase（ERK） and p38 kinase were detected by Western blot ***:IL-1ra （10and 40 mg/kg,ic,d 14-21） modulated the secondary inflammatory reaction（P<0.01）,ultrastructure of synoviocytes and mitogen-activated protein kinase（MAPK） phosphorylation in AA *** administration of IL-1ra （10 and 40mg/kg,ic,d 14-21） in AA rats significantly decreased the production of IL-1,PGE2and TNF-α by macrophage-like synoviocytes （MLS）（P<0.01）.IL-1ra （2.5 mg/kg）also decreased the production of PGE2（P<0.01） and TNF-α（P<0.05） by MLS inAA *** increased phosphorylation of MAPK and cell proliferation in fibro-blast-like synoviocytes （FLS） stimulated by supernatants of MLS in AA rats wasalso inhibited by IL-1ra （10 and 40 mg/kg,ic.d 14-21）.Conclusion:IL-1ra hasanti-inflammatory effects because it modulates the ultrastructure of synoviocytes,decreases the production of pro-inflammatory mediators by MLS,and inhibits thephosphorylation of MAPK in FLS.

Objective To investigate the partial mechanisms of Paeoniflorin(PF) on the function of synoviocytes stimulated by recombinant interleukin-1(rIL-1α).Methods CIA was induced in male Sprague-Dawley rats immunized with chicken typeⅡcollagen in complete Freund’ s *** levels of prostaglandin E(PGE) and cyclic adenosine monophosphate(cAMP) were measured by *** proliferation of synoviocytes was determined by the 3-(4,5-2dimethylthiazal-2yl) 2,5-diphenyltetrazoliumbromide *** of Eprostanoid (EP) receptor was detected by Western blotting *** PF(12.5,62.5 mg·L) inhibited abnormal proliferation of synoviocytes stimulated by rIL-1α,rIL-1αtime dependently increased PGE production and decreased cAMP production of cynoviocytes in rats of ***,PF(12.5,62.5 mg·L) decreased the PGE production and increased cAMP level of synoviocytes pretreated by rIL-1α.The expression of Eprostanoid (EP4) decreased in synovium of CIA rats at day 7,21 and *** administration of PF(12.5,62.5 mg·L) increased the decreased EP4 expression in synoviocytes pretreated by rIL-1αin *** PF might exert its anti-inflammatory effects through regulating the proliferative and secretory function of synoviocytes by rIL-1α,which might be associated with its ability to up-regulate the EP4 receptor protein expression and cAMP level.

Objective Opioid drugs are widelv used for treating moderate to severe pain,but accumutalting evidence suggests that administration of opioids such as morphine,fentanynl and remifentanyl can leads not only to analgesia, but also to an enhancement in pain sensitivity induced by central *** phenomenon is re- ferred to as opioid induced hyperalgesia(OIH).The present study assessed the effect of melatonin on fentanyl and morphine induced hyperalgesia using a tail-flick *** In the first set of experiments,to induce hyperalgesia, fentanynl was injected four times resulting in a total dose of 60,180,240 ug/kg *** 1 *** with MT in various doses(25,50,100 mg/kg) 30 min before fentanynl and after *** second set experiment, morphine induced tolerance and hyperalgesia were induced in rats by repeated injection of morphine(10 mg/ kg,s.c.) twice daily ***(25,50,100 mg/kg) were used orally in combination with morphine. Results Moderate and high dose(180,240 ug/kg) but not low fentanynl could induce hyperalgesia after drug *** present observations suggest that melatonin(50,100 mg/kg) could prevent fentanynl and morphine induced hyperalgesia and partially reversed morphine *** Melatonin can combined used with opioids to prevent OIH.
