Inflammasome links traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease

Traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease are three distinct neurological disorders that share common pathophysiological mechanisms involving neuroinflammation. One sequela of neuroinflammation includes the pathologic hyperphosphorylation of tau protein, an endogenous microtubule-associated protein that protects the integrity of neuronal cytoskeletons. Tau hyperphosphorylation results in protein misfolding and subsequent accumulation of tau tangles forming neurotoxic aggregates. These misfolded proteins are characteristic of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease and can lead to downstream neuroinflammatory processes, including assembly and activation of the inflammasome complex. Inflammasomes refer to a family of multimeric protein units that, upon activation, release a cascade of signaling molecules resulting in caspase-induced cell death and inflammation mediated by the release of interleukin-1β cytokine. One specific inflammasome, the NOD-like receptor protein 3, has been proposed to be a key regulator of tau phosphorylation where it has been shown that prolonged NOD-like receptor protein 3 activation acts as a causal factor in pathological tau accumulation and spreading. This review begins by describing the epidemiology and pathophysiology of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease. Next, we highlight neuroinflammation as an overriding theme and discuss the role of the NOD-like receptor protein 3 inflammasome in the formation of tau deposits and how such tauopathic entities spread throughout the brain. We then propose a novel framework linking traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease as inflammasomedependent pathologies that exist along a temporal continuum. Finally, we discuss potential therapeutic targets that may intercept this pathway and ultimately minimize long-term neurological decline.

Layer-layout-based heuristics for loading homogeneous items into a single container

The container loading problem (CLP) is a well-known NP-hard problem. Due to the computation complexity, heuristics is an often-sought approach. This article proposes two heuristics to pack homogeneous rectangular boxes into a single container. Both algorithms adopt the concept of building layers on one face of the container, but the first heuristic determines the layer face once for all, while the second treats the remaining container space as a reduced-sized container after one layer is loaded and, hence, selects the layer face dynamically. To handle the layout design problem at a layer's level, a block-based 2D packing procedure is also developed. Numerical studies demonstrate the efficiency of the heuristics.

Quantifying and mapping spatial variability of Shanghai household carbon footprints

Understanding the spatial variability of house- hold carbon emissions is necessary for formulating sustainable and low-carbon energy policy. However, data on household carbon emissions is limited in China, the world＇s largest greenhouse gases emitter. This study quantifies and maps household carbon emissions in Shanghai using a city-wide household survey. The findings reveal substantial spatial variability in household carbon emissions, especially in transport-related emissions. Low emission clusters are founded in Hongkou, Xuhui, Luwan, Jinshan, and Fengxian. High emission clusters are located in Jiading and Pudong. Overall, the spatial pattern of household carbon emissions in Shanghai is donut-shaped： lowest in the urban core, increasing in the surrounding suburban areas, and declining again in the urban fringe and rural regions. The household emissions are correlated with a number of housing and socioeconomic factors, including car ownership, type of dwelling, size of dwelling, age of dwelling, and income. The findings underscore the importance of a localized approach to low-carbon policy- making and implementation.

Levin-Wen models are microscopic spin models for topological phases of matter in （2＋ 1）-dimension. We introduce a generalization of such models to （3 ＋ 1）-dimension based on unitary braided fusion categories, also known as unitary premodular categories. We discuss the ground state degeneracy on 3-manifolds and statistics of excitations which include both points and defect loops. Potential con- nections with recently proposed fractional topological insulators and projective ribbon permutation statistics are described.

Modern treatment of gastric gastrointestinal stromal tumors

Gastrointestinal stromal tumors (GIST) are rare mesenchymal smooth muscle sarcomas that can arise anywhere within the gastrointestinal tract. Sporadic mutations within the tyrosine kinase receptors of the interstitial cells of Cajal have been identified as the key molecular step in GIST carcinogenesis. Although many patients are asymptomatic, the most common associated symptoms include: abdominal pain, dyspepsia, gastric outlet obstruction, and anorexia. Rarely, GIST can perforate causing life-threatening hemoperitoneum. Most are ultimately diagnosed on cross-sectional imaging studies (i.e., computed tomography and/or magnetic resonance imaging in combination with upper endoscopy. Endoscopic ultrasonographic localization of these tumors within the smooth muscle layer and acquisition of neoplastic spindle cells harboring mutations in the c-kIT gene is pathognomonic. Curative treatment requires a complete gross resection of the tumor. Both open and minimally invasive operations have been shown to reduce recurrence rates and improve long-term survival. While there is considerable debate over whether GIST can be benign neoplasms, we believe that all GIST have malignant potential, but vary in their propensity to recur after resection and metastasize to distant organ sites. Prognostic factors include location, size (i.e., > 5 cm), grade (> 5-10 mitoses per 50 high power fields and specific mutational events that are still being defined. Adjuvant therapy with tyrosine kinase inhibitors, such as imatinib mesylate, has been shown to reduce the risk of recurrence after one year of therapy. Treatment of locally-advanced or borderline resectable gastric GIST with neoadjuvant imatinib has been shown to induce regression in a minority of patients and stabilization in the majority of cases. Food safety regulatory systems in Europe and China:A study of how co-regulation can improve regulatory effectiveness

Food safety has received a great deal of attention in both developed and developing countries in recent years. In China, the numerous food scandals and scares that have struck over the past decade have spurred significant food safety regulatory reform, which has been increasingly oriented towards the public-private partnership model adopted by the Europe Union＇s （EU） food safety regulatory system. This paper analyzes the development of both the EU＇s and China＇s food safety regu- latory systems, identifies the current challenges for China and additionally considers the role of public-private partnership. The success of co-regulation in the food regulatory system would bring significant benefits and opportunities for China. Finally, this paper recommends additional measures like training and grants to improve the private＇s sector effectiveness in co-regulating China＇s food safety issues.

An Evaluation Model of Supply Chain Performances Using 5DBSC and LMBP Neural Network Algorithm

A high efficient Supply Chain （SC） would bring great benefits to an enterprise such as integrated resources, reduced logistics costs, improved logistics efficiency and high quality of overall level of services. So it is important to research various methods, performance indicator systems and technology for evaluating, monitoring, predicting and optimizing the performance of a SC. In this paper, the existing performance indicator systems and methods are discussed and evaluated. Various nature-inspired algorithms are reviewed and their applications for SC Performance Evaluation （PE） are discussed. Then, a model is proposed and developed using 5 Dimensional Balanced Scorecard （5DBSC） and LMBP （Levenberg-Marquardt Back Propagation） neural network for SC PE. A program is written using Matlab tool box to implement the model based on the practical values of the 14 indicators of 5DBSC of a given previous period. This model can be used to evaluate, predict and optimize the performance of a SC. The analysis results of a case study of a company show that the proposed model is valid, reliable and effective. The convergence speed is faster than that in the previous work.
