Compressive Stress Enhances Invasive Phenotype of Cancer Cells via Piezo1 Activation

In this study,we hypothesized that Piezo 1 channels mediate the compression-enhanced invasive phenotype of cancer cells via a caveolae-dependent *** test this hypothesis,we examined in vitro cultured human breast cancer cells for their ability to invade and degrade extracellular matrix in the presence or absence of compressive stress,together with corresponding changes in Piezo1 as well as cytoskeletal remodeling and calcium *** we show that compressive stress enhanced invasion,matrix degradation,and invadopodia formation of breast cancer *** further identified Piezo1 as the putative mechanosensitive cellular component that transmits compression to induce calcium influx,which in turn triggers several downstream ***,for the first time we observed inv-adopodia with matrix degradation ability on the apical side of the cells, similar to those commonly observed at the cell s ventral ***,we demonstrate that Piezo1 and caveolae were both involved in mediating the compressive stress-induced cancer cell invasive phenotype as Piezo 1 and caveolae were often colocalized,and reduction of Cav-1 expression or disruption of caveolae with methyl-β-cyclodextrin led to not only reduced Piezo1 expression but also attenuation of the invasive phenotypes promoted by compressive *** together,we first observed that in breast cancer cells,simulating uncontrolled growth-induced compressive stress enhanced cancer cell invasion,matrix degradation,and invadopodia and stress fiber *** study also confirmed that Piezo1 channels are highly expressed in breast cancer cells compared to normal breast cells,and is consistent with the data that compressive stress regulates cell migration of breast cancer cells but not normal breast ***,we identified that Piezol mediated these processes and the invasive phenotypes also depended on the integrity of *** findings provide the first demonstration that compres

Productivity and energy balance of forest plantation harvesting in Uganda

The study evaluated the productivity and energy balance of the harvesting and the extraction of wood in plantation of exotic tree species, i.e., Pinus patula （Schiede ex Schlectendahl et Chamisso） and Cupressus lusitanica （Mill.）. The harvesting and extraction operations concerned clear-cut felling on two different types of terrain （gentle and steep terrain） and two levels of prun- ing. The results show that cutting is not affected by the terrain gradient whereas the pruning condition shows a significant effect on the delimbing operation. The productivity of cutting is evaluated at 28.1 m3.d-1 for unpruned trees and at 30.2 m3.d t for pruned trees. For the extraction phase, rolling was significantly affected by terrain conditions. On steep terrain, the gross productivity can be evaluated at 22.5 m3.d 1 for an extraction distance of 40 m, while for the same distance on gentle terrain the productivity decreases to 14.1 m3.d~. As a result, the steep terrain condition showed higher energy efficiency, suggesting that it is the more efficient system of extraction. The productivity model and the energy balance resulting from the study provide a better understanding of the variables af- fecting motor-manual cutting and rolling extraction in exotic plantations in Uganda.

Prolapse Incidence in Swine Breeding Herds Is a Cause for Concern

Beginning in the fall of 2014 there has been a general and widespread increase in the incidence of prolapse in the U.S. swine herd. The purpose of this manuscript is to review the incidence, causative factors and treatment of rectal, vaginal, uterine and preputial prolapses. Rectal and vaginal prolapses are most common in swine when compared to other prolapse types. The cause of prolapses supports a fixation mechanism failure overcome by pressure on or weakening of support tissue. The fundamental factors affecting the incidence for prolapses are many and include factors related to nutrition, physiology, hormones, genetics, environment and other disease factors such as chronic diarrhea, cough, and dystocia. Treatment of prolapsed swine includes surgical and therapeutic management that can lead to complete recovery. However, in most cases, euthanasia is the final result. Economic loss was calculated at approximately $5220 dollars/year/1000 sows.

Primary 4-3-oxosteroid 5β-reductase deficiency:Two cases in China

Aldo-keto reductase 1d1(AKR1d1) deficiency,a rare but life-threatening form of bile acid deficiency,has not been previously described in ***,we describe the first two primary 4-3-oxosteroid 5β-reductase deficiency patients in China's Mainland diagnosed by fast atom bombardment-mass spectroscopy of urinary bile acids and confirmed by genetic analysis.A high proportion of atypical 3-oxo-4-bile acids in the urine indicated a deficiency in 4-3-oxosteroid 5β-*** of the coding exons and adjacent intronic sequence of the AKR1d1 gene were sequenced using peripheral lymphocyte genomic dNA of two patients and one of the patient's *** patient exhibited compound heterozygous mutations:c.396C>A and c.722A>T,while the other was heterozygous for the mutation c.797G>*** on these mutations,a diagnosis of primary 4-3-oxosteroid 5β-reductase deficiency could be *** ursodeoxycholic acid treatment and fat-soluble vitamin supplements,liver function tests normalized rapidly,and the degree of hepatomegaly was markedly reduced in both patients.

Mass budgets of the Lambert, Mellor and Fisher Glaciers and basal fluxes beneath their flowbands on Amery Ice Shelf

We used in situ measurements and remote-sensing data sets to evaluate the mass budgets of the Lambert, Mellor and Fisher Glaciers and the basal melting and freezing rates beneath their flowbands on the Amery Ice Shelf. Our findings show the Lambert and Mellor Glaciers upstream of the ANARE Lambert Glacier Basin (LGB) traverse may have positive imbalances of 3.9±2.1 Gt a-1 and 2.1±2.4 Gt a-1, respectively, while the Fisher Glacier is approximately in balance. The upstream region as a whole has a positive imbalance of 5.9±4.9 Gt a-1. The three same glaciers downstream of the ANARE LGB traverse line are in negative imbalance, where the whole downstream region has a negative imbalance of -8.5±5.8 Gt a-1. Overall the mass budgets of the Lambert, Mellor, and Fisher Glaciers are close to bal-ance, and the collective three-glacier system is also nearly in balance with a mass budget of -2.6±6.5 Gt a-1. The significant positive imbalances for the interior basin upstream of the ice-movement stations established in the early 1970s (GL line) reported previously are possibly due to an overestimate of the total accumulation and an underestimate of the ice flux through the GL line. The mean melting rate is -23.0±3.5 m ice a-1 near the southern grounding line, which decreases rapidly downstream, and transitions to refreezing at around 300 km from the southern extremity of the Amery Ice Shelf. Freezing rates along the flowbands are around 0.5±0.1 to 1.5±0.2 m ice a-1. The per-centage of ice lost from the interior by basal melting beneath the flowbands is about 80%±5%. The total basal melting and refreezing beneath the three flowbands is 50.3±7.5 Gt ice a-1 and 7.0±1.1 Gt ice a-1, respectively. We find a much larger total basal melting and net melting than the results for the whole Amery Ice Shelf derived from previous modeling and oceanographic measurements.

Overall rate, location, and predictive factors for positive surgical margins after robot-assisted laparoscopic radical prostatectomy for high-risk prostate cancer

We report the overall rate, locations and predictive factors of positive surgical margins （PSMs） in 271 patients with high-risk prostate cancer. Between April 2008 and October 2011, we prospectively collected data from patients classified as d＇Amico high-risk who underwent robot-assisted laparoscopic radical prostatectomy. Overall rate and location of PSMs were reported. Stepwise logistic regression models were fitted to assess predictive factors of PSM. The overall rate of PSMs was 25.1% （68 of 271 patients）. Of these PSM, 38.2% （26 of 68） were posterolateral （PL）, 26.5% （18 of 68） multifocal, 16.2% （11 of 68） in the apex, 14.7% （10 of 68） in the bladder neck, and 4.4% （3/68） in other locations. The PSM rate of patients with pathological stage pT2 was 8.6% （12 of 140）, 26.6% （17 of 64） of pT3a, 53.3% （32/60） of pT3b, and 100% （7 of 7） of pT4. In a logistic regression model including pre-, intra-, and post-operative parameters, body mass index （odds ratio [OR]. 1.09; 95% confidence interval [CI]： 1.01-1.19, P = 0.029）, pathological stage （pT3b or higher vs pT2; OR： 5.14; 95% Ch 1.92-13.78; P = 0.001） and percentage of the tumor （OR： 46.71; 95% CI： 6.37-342.57; P 〈 0.001） were independent predictive factors for PSMs. The most common location of PSMs in patients at high-risk was the PL aspect, which reflects the reported tumor aggressiveness. The only significant predictive factors of PSMs were pathological outcomes, such as percentage of the tumor in the specimen and pathological stage.

The early Albian Oceanic Anoxic Event 1 b(OAE 1 b) is well documented in the Tethys, Pacific and North Atlantic, but few studies have evaluated whether or not terrestrial records of OAE 1 b exist. In order to identify terrestrial records of the early Albian OAE 1 b and to infer possible driving mechanisms, an integrated multi-proxy study from the late Aptian to Albian in the Fuxin lacustrine basin was conducted,including thick, organic-rich black mudstones, total organic carbon(TOC), organic carbon isotopes(d13 Corg), mercury concentration(Hg) and results from pyrolysis analyses(S2, Tmaxand HI). Results show three distinct short-term negative d13 Corgexcursions corresponding with relatively high TOC values,which could be counterparts of the Kilian, Paquier and Leenhardt sub-events of the early Albian OAE1 b. Atmospheric CO2 concentration(p CO2) recovered from C3 plant d13 Corgcompositions indicates an increasing trend in Unit C during the early Albian, and there are three short-term increases of p CO2 corresponding to the three sub-events of OAE 1 b at this time interval. We infer that a trend of increasing p CO2 during the Kilian sub-event in the study area is closely related to volcanism. Continental weathering calculated using chemical weathering indices(CIA, WIP and MIA(O)) show an increasing trend during the OAE 1 b interval, likely resulting from warmer and more humid conditions. Mixed sources of terrestrial plants and lacustrine plankton demonstrated by pyrolysis analyses(HI vs. Tmaxand S2 vs. TOC), indicate a terrestrial contribution to the organic-rich sediments of the Kilian, Paquier and Leenhardt sub-events of OAE 1 b. We suggest that a CO2-forced greenhouse effect during the early Albian might have triggered the relatively warm and humid palaeoclimatic conditions, and intensified chemical weathering that combined to create high nutrient and organic matter levels that were flushed into lakes contributing to eutrophication and anoxia in lacustrine and in c
