The effect of adjunct caudal block on postoperative analgesia in robot-assisted laparoscopic radical prostatectomy: A prospective randomized controlled, single blinded pilot study in a tertiary centre

Cytokine and Antioxidant Regulation in the Intestine of the Gray Mouse Lemur (Microcebus murinus) during Torpor

during food shortages, the gray mouse lemur（Microcebus murinus） of Madagascar experiences daily torpor thereby reducing energy expenditures. The present study aimed to understand the impacts of torpor on the immune system and antioxidant response in the gut of these *** interaction may be of critical importance given the trade-off between the energetically costly immune response and the need to defend against pathogen entry during hypometabolism. The protein levels of cytokines and antioxidants were measured in the small intestine（duodenum, jejunum,and ileum） and large intestine of aroused and torpid lemurs. While there was a significant decrease of some pro-inflammatory cytokines（IL-6 and TNF-a） in the duodenum and jejunum during torpor as compared to aroused animals, there was no change in anti-inflammatory cytokines. We observed decreased levels of cytokines（IL-12p70 and M-CSF）, and several chemokines（MCP-1and MIP-2） but an increase in MIP-1a in the jejunum of the torpid animals. In addition, we evaluated antioxidant response by examining the protein levels of antioxidant enzymes and total antioxidant capacity provided by metabolites such as glutathione（and others）. Our results indicatedthat levels of antioxidant enzymes did not change between torpor and aroused states, although antioxidant capacity was significantly higher in the ileum during torpor. These data suggest a suppression of the immune response, likely as an energy conservation measure, and a limited role of antioxidant defenses in supporting torpor in lemur intestine.

Effects of live yeast on differential genetic and functional attributes of rumen microbiota in beef cattle

Several studies have evaluated the effects of live yeast supplementation on rumen microbial population;however,its effect on differential microbial genes and their functional potential has not been described. Thus, this study applied shotgun metagenomic sequencing to evaluate the effects of live yeast supplementation on genetic and functional potential of the rumen microbiota in beef cattle. Eight rumen-cannulated Holstein steers were randomly assigned to two treatments in a cross-over design with two 25-day experimental periods and a 10-day wash-out between the two periods. The steers were housed in individual pens and fed 50% concentrate-mix and 50% red clover/orchard hay ad libitum. Treatments were(1) control(CON;basal diet without additive) and(2) yeast(YEA;basal diet plus 15 g/d of live yeast product). Rumen fluid samples were collected at 3, 6, and 9 h after feeding on the last d of each period. Sequencing was done on an Illumina Hi Seq 2500 platform. dietary yeast supplementation increased the relative abundance of carbohydrate-fermenting bacteria(such as Ruminococcus albus, ***, R. bromii, and R. obeum) and lactate-utilizing bacteria(such as Megasphaera elsdenii, desulfovibrio desulfuricans, and d. vulgaris). A total of 154 differentially abundant genes(dEGs) were obtained(false discovery rate dia of Genes and Genomes(KEGG) annotation analysis of the dEGs revealed that 10 pathways, including amino sugar and nucleotide sugar metabolism, oxidative phosphorylation, lipopolysaccharide biosynthesis, pantothenate and coenzyme A biosynthesis, glutathione metabolism, beta-alanine metabolism,polyketide sugar unit biosynthesis, protein export, ribosome, and bacterial secretory system, were enriched in steers fed YEA. Expression Profiling and Structural Characterization of Micro RNAs in Adipose Tissues of Hibernating Ground Squirrels

Micro RNAs（mi RNAs） are small non-coding RNAs that are important in regulating metabolic stress. In this study, we determined the expression and structural characteristics of 20 mi RNAs in brown（BAT） and white adipose tissue（WAT） during torpor in thirteen-lined ground squirrels. Using a modified stem-loop technique, we found that during torpor, expression of six mi RNAs including let-7a, let-7b, mi R-107, mi R-150, mi R-222 and mi R-31 was significantly downregulated in WAT（P 〈 0.05）, which was 16%–54% of euthermic non-torpid control squirrels,whereas expression of three mi RNAs including mi R-143, mi R-200 a and mi R-519 d was found to be upregulated by 1.32–2.34-fold. Similarly, expression of more mi RNAs was downregulated in BAT during torpor. We detected reduced expression of 6 mi RNAs including mi R-103 a, mi R-107, mi R-125 b, mi R-21, mi R-221 and mi R-31（48%–70% of control）, while only expression of mi R-138 was significantly upregulated（2.91 ± 0.8-fold of the control, P 〈 0.05）. Interestingly,mi RNAs found to be downregulated in WAT during torpor were similar to those dysregulated in obese humans for increased adipogenesis, whereas mi RNAs with altered expression in BAT during torpor were linked to mitochondrial b-oxidation. mi RPath target prediction analysis showed that mi RNAs downregulated in both WAT and BAT were associated with the regulation of mitogen-activated protein kinase（MAPK） signaling, while the mi RNAs upregulated in WAT were linked to transforming growth factor b（TGFb） signaling. Compared to mouse sequences, no unique nucleotide substitutions within the stem-loop region were discovered for the associated pre-mi RNAs for the mi RNAs used in this study, suggesting no structure-influenced changes in pre-mi RNA processing efficiency in the squirrel. This study aims to enhance the adhesion strength and anti-corrosion performance of the cold galvanizing coating(CGC)applied on the hot-dip galvanized steel(HdG).Polydopamine(PdA)is deposited on the HdG surface with different time ranges and as an interlayer between CGC and HdG through covalent *** surface morphology and the covalent interaction between PdA/HdG are exhibited by scanning electron microscope(SEM),atomic force microscopy(AFM)and X-ray photoelectron spectroscopy(XPS).The pull-off adhesion tests before and after neutral slat spry tests show an enhanced dry adhesion strength and less adhesion loss of the hybrid CGC/PdA coated HdG compared with the direct CGC coated *** addition,open circuit potential(OCP)reveals that the corrosion protection performance of the hybrid CGC/PdA coated HdG increases by 200%(up to 201 d)and the corrosion density icorrattaining about 4.45×10^(-7)A/cm^(2).Electrochemical impedance spectroscopy(EIS)measurements and X-ray diffraction(XRd)analysis confirm that the precipitate of the stable chelation formed by PdA and Zn^(2+)between CGC and HdG substrate can also improve the corrosion protection *** a strategy of strengthening adhesion and forming the chelate compound at the HdG surface promises a new route to corrosion protection of CGC on HdG.

Efficacy Improvement of a Bioherbicidal Fungus Using a Formulation-Based Approach

Greenhouse experiments were conducted to determine the effects of an invert (water-in-oil) emulsion (IE) on dew period duration and dew delay of Colletotrichum coccodes for biocontrol of the problematic weed, eastern black nightshade (Solanum ptycanthum). dew periods of 4, 8, or 12 h provided 10%, 25%, and 40% control of eastern black nightshade plants, respectively, when C. coccodes (Strain NRRL 15,547) spores were applied in water + Tween 80 surfactant 12 days after inoculation, but a minimum of 16 h of dew was required to achieve ~95% plant mortality. In contrast, at these same intervals of dew, 95%, 100% and 100% mortality occurred, respectively, when fungal spores were formulated in the IE. Even in the absence of dew, 60% mortality and 70% dry weight reductions of plants was achieved with the fungus/IE formulation delaying dew by 2 h after inoculation did not significantly reduce weed control or plant dry weight reductions when plants were inoculated with the fungus either in the aqueous or in the IE formulation. However, when dew was delayed for 4, 8, or 12 h, only 60%, 50%, and 25% mortality, respectively, of plants receiving the aqueous spore treatment occurred. In contrast, 95%, 90%, and 90% weed mortality occurred after the same dew delays of plants receiving the fungus/ IE treatments. These results demonstrate that formulating C. coccodes spores in an invert emulsion greatly improves the bioherbicidal potential of this fungus. Furthermore, results suggest that this formulation may render pathogens previously rejected for development as bioherbicides due to restrictive dew requirements more efficacious for use in controlling their target weeds.

Heart regeneration occurs by dedifferentiation and proliferation of pre-existing cardiomyocytes(CMs).However,the signaling mechanisms by which injury induces CM renewal remain incompletely ***,we find that cardiac injury in zebrafish induces expression of the secreted Wnt inhibitors,including dickkopf 1(dkkl),dkk3,secreted Frizzled-related protein 1(sFrpl),and sFrp2,in cardiac tissue adjacent to injury *** blocking of Wnt activity via dkkl overexpression enhances CM proliferation and heart regeneration,whereas ectopic activation of Wnt8 signaling blunts injury-induced CM dedifferentiation and *** Wnt signaling is dampened upon injury,the cytoplasmic β-catenin is unexpectedly increased at disarrayed CM sarcomeres in myocardial wound *** analyses indicated that p21-activated kinase 2(Pak2)is induced at regenerating CMs,where it phosphorylates cytoplasmic β-catenin at Ser 675 and increases its stability at disassembled ***-specific induction of the phospho-mimeticβ-catenin(S675E)enhances CM dedifferentiation and sarcomere disassembly in response to ***,inactivation of Pak2 kinase activity reduces the Ser 675-phosphorylatedβ-catenin(pS675-β-catenin)and attenuates CM sarcomere disorganization and dedifferentiation・Taken together,these findings demonstrate that coordination of Wnt signaling inhibition and Pak2/pS675-βYatenin signaling enhances zebrafish heart regeneration by supporting CM dedifferentiation and proliferation.
