Glucagon-like peptide-1 analogue prevents nonalcoholic steatohepatitis in non-obese mice

AIM: To investigate whether a glucagon-like peptide-1(GLP-1) analogue inhibits nonalcoholic steatohepatitis(NASH), which is being increasingly recognized in Asia, in non-obese mice. METHODS: A methionine-choline-deficient diet(MCD) along with exendin-4(20 μg/kg per day, ip), a GLP-1 analogue, or saline was administered to male db/db mice(non-obese NASH model). Four or eight weeks after commencement of the diet, the mice were sacrificed and their livers were excised. The excised livers were examined by histochemistry for evidence of hepatic steatosis and inflammation. Hepatic triglyceride(TG) and free fatty acid(FFA) content was measured, and the expression of hepatic fat metabolism- and inflammation-related genes was evaluated. Oxidative stress-related parameters and macrophage recruitment were also examined using ***: Four weeks of MCD feeding induced hepatic steatosis and inflammation and increased the hepatic TG and FFA content. The expression of fattyacid transport protein 4(FATP4), a hepatic FFA influxrelated gene; macrophage recruitment; and the level of malondialdehyde(MDA), an oxidative stress marker, were significantly augmented by a 4-wk MCD. The levels of hepatic sterol regulatory element-binding protein-1c(SREBP-1c) m RNA(lipogenesis-related gene) and acyl-coenzyme A oxidase 1(ACOX1) m RNA(β-oxidation-related gene) had decreased at 4 wk and further decreased at 8 wk. However, the level of microsomal triglyceride transfer protein m RNA(a lipid excretion-related gene) remained unchanged. The administration of exendin-4 significantly attenuated the MCD-induced increase in hepatic steatosis, hepatic TG and FFA content, and FATP4 expression as well as the MCD-induced augmentation of hepatic inflammation, macrophage recruitment, and MDA levels. Vanadium alloy is proposed as an attractive candidate for first wall and blanket structural material of fusion reactors. The retention and release behaviors of hydrogen and helium in vanadium alloy may be an important issue. In the present work, 1.7 keV deuterium and 5 keV helium ions are respectively implanted into V-4Cr-4Ti and V-4Ti at room temperature. The retention and release of deuterium and helium are measured with thermal desorption spectroscopy (TDS). When the helium ion fluence is larger than 3 × 1017 He/cm2, the retained helium saturates with a value of approximately 2.5 ×1017 He/cm2. However, when the ion fluence is 1×1019 D/cm2, the hydrogen saturation in vanadium alloy does not take place. Experimental results indicates that hydrogen and helium retention in vanadium alloy may lead to serious problems and special attention should be paid when it is applied to fusion reactors.

Low activation materials have to be developed toward fusion demonstrationreactors. Ferritic steel, vanadium alloy and SiC/SiC composite are candidate materials of the firstwall, vacuum vessel and blanket components, respectively. Although changes of mechanical-thermalproperties owing to neutron irradiation have been investigated so far, there is little data for theplasma material interactions, such as fuel hydrogen retention and erosion. In the present study,deuterium retention and physical sputtering of low activation ferritic steel, F82H, wereinvestigated by using deuterium ion irradiation apparatus. After a ferritic steel sample wasirradiated by 1.7 keV D^+ ions, the weight loss was measured to obtain the physical sputteringyield. The sputtering yield was 0.04, comparable to that of stainless steel. In order to obtain theretained amount of deuterium, technique of thermal desorption spectroscopy (TDS) was employed to theirradiated sample. The retained deuterium desorbed at temperature ranging from 450 K to 700 K, inthe forms of DHO, D_2, D_2O and hydrocarbons. Hence, the deuterium retained can be reduced by bakingwith a relatively low temperature. The fluence dependence of retained amount of deuterium wasmeasured by changing the ion fluence. In the ferritic steel without mechanical polish, the retainedamount was large even when the fluence was low. In such a case, a large amount of deuterium wastrapped in the surface oxide layer containing O and C. When the fluence was large, the thickness ofsurface oxide layer was reduced by the ion sputtering, and then the retained amount in the oxidelayer decreased. In the case of a high fluence, the retained amount of deuterium became comparableto that of ferritic steel with mechanical polish or SS 316 L, and one order of magnitude smallerthan that of graphite. To determine whether dried fermented ginger (DFG), fermented with Japanese mugwort silage juice, could be replaced by fermented corncob powder (FCP) as a of feed ingredient source without significant body weight decrease or damage to visceral organs (using gross anatomical observation), to intestinal villi (using light microscopy), or to the epithelial cells on the villus apical surface (using scanning electron microscopy) the following investigation was performed. Sixty-four male broilers were allotted to 4 groups: a basal diet group (control group), and basal diet groups with DFG at a level of 50 ppm;with DFG at 50 ppm and FCP at 250 ppm (50 ppm DFG + 250 ppm FCP group);and with FCP at a level of 500 ppm (500 ppm FCP group). Feed intake, body weight gain, feed efficiency, carcass quality, small intestinal length and weight, and visceral organ weight were not different among groups. Furthermore, regarding intestinal villus height, villus area and crypt depth, a significant difference was not found among the groups. When these values of the control were expressed as an index of 100, the duodenal villus height of the 50 ppm DFG + 250 ppm FCP group and the 500 ppm FCP group were 114 and 119, respectively. The duodenal villus area of the 50 ppm DFG + 250 ppm FCP group and the 500 ppm FCP group were 125 and 158, respectively. These villus heights and areas are thought to be activated. On the epithelial cells on the villus apical surface in the duodenum and jejunum, the 50 ppm DFG + 250 ppm FCP group had protuberated cells into the intestinal lumen and deeper cells at the sites of recently exfoliated cells, suggesting that these cells are activated. The present results indicate that small amounts of fermented corncob powder can be used as a feed supplement when mixed with fermented ginger powder, due to the synergy between the two ingredients, resulting in a 6% increase in body weight gain。

Morphological Assessment of the Small Intestine of Broilers Fed Dietary Natural Zeolite Including Plant Extract

Many kinds of feed additives （e.g., probiotics, prebiotics, antibiotics, etc.） can be used in chicken diets to stimulate intestinal morphological maturation and, consequently, intestinal function. The aim of this trial was to investigate the possibility that natural zeolite including plant extract （ZEM） could encourage the hypertrophy of intestinal villi and the absorptive function of epithelial cells in broiler chickens. At 7 days old, 64 male Marshall Chunky broilers were divided into four groups, each with four replicates of four chickens. The birds were fed a basal mash diet supplemented with ZEM at 0% （control）, 0.05%, 0.1%, and 0.2% for 42 days. At 49 days old, four chickens from each treatment were killed to obtain intestinal samples. Morphological intestinal assessment was conducted using both a light and a scanning electron microscope. A tendency to increased villus height and cell area could be observed throughout the intestinal segments of the broilers fed ZEM. Particularly, the villus height in the duodenum was significantly increased （P 〈 0.05） in the 0.05% ZEM group when compared with the control group. The area of the villus cells tended to be higher in the ZEM groups in all intestinal regions and showed a significant increase （P 〈 0.05） in the ileal part of the 0.2% ZEM group. Also, the assessment of cells mitosis in the intestinal crypts showed significantly higher numbers （P 〈 0.05） in all levels of ZEM groups in the duodenum and a tendency to increase in value in the jejunum and ileum, as compared with the control group. More protuberated cells and multi-cell clusters were found in the ZEM groups than in the control group, resulting in a rough cellular surface. In conclusion, the present study might suggest that the intestinal villi and epithelial cells on the villus apical surface are hypertrophied in the ZEM groups, and that intestinal villi adapt to enhance their absorption capacity by increasing the absorptive surface area. The phenc, lic compounds found in mango fruit are antioxidants, and contribute to a reduction in the risk of cardiovascular diseases. Mango carotenoids are synthesized in mango fruit during ripening. A major by-product of mango fruit is the skin, which is available after the fruits is consumed or used, particularly in the preparation of jams and fruit juices by the fruit canning industry. The objective of this study was to investigate the effects of dietary mango skin （MS） or mango skin combined with paprika extracts （PE） on production performance, egg quality, and egg yolk polyphenols. Total of 60 44-week-old Boris Brown hens was assigned, based on egg production rate and body weight, to 6 groups （10 birds in each group）. The laying hens were fed a basal diet （control group, 18 CP, 2,800 kcal/kg ME）, a basal diet supplemented with 0.1% PE, 1% or 5% MS, or with a mixture of 0.1% PE with 1% or 5% MS respectively. During the three-week experimental feeding period, the birds had free access to feed and water. Feed consumption was measured weekly and egg production was recorded daily. The results for initial body weight, feed consumption, hen-day production, egg mass, mortality, and final body weight did not indicate any effects of the different treatments （P 〉 0.05）. There were no significant differences （P 〉 0.05） in shell-breaking strength, shell thickness, shell ratio, yolk ratio or Haugh units, except in the case of egg yolk color. Roche yolk color fan scores were better in all experimental groups than in the control （P 〈 0.0001）. The yolk color, yellow index, and ratio of redness to yellowness were greater （P 〈 0.0001） in the 0.1% PE, the 0.1% PE ＋ 1% MS, and the 0.1% PE ＋ 5% MS group than in the 1%, 5% MS, and control groups. Compared with the control, lightness was decreased significantly in the 0.1% PE, the 0.1% PE ＋ 1% MS, and the 0.1% PE ＋ 5% MS groups （P 〈 0.05）, whereas redness was increased significantly in the 0.1% PE, 0.1% PE ＋ 1% MS, and 0.1% PE ＋ 5% MS
