Hemolysis of PM_(10) on RbCs in vitro:An indoor air study in a coal-burning lung cancer epidemic area

Epidemiological studies have suggested that inhalation exposure to indoor ambient air from coal-burning environments is causally associated with respiratory health *** order to explore the toxicological mechanisms behind the adverse health effects,the hemolytic activity of PM_(10)(particulate matter with an aerodynamic diameter of 10 um or less)samples collected from homes burning coal in the recognized China"cancer village"Xuanwei were evaluated and matched against their trace elemental *** results demonstrated that the hemolytic activity of indoor PM_(10) in coal-burning environments ranged from 4.28%to 5.24%,with a clear positive dose-response *** low dose samples exhibited a reduced hemolytic activity,PM_(10) could have a toxic effect upon people in a coal-burning indoor environment for extended time *** concentrations of analyzed trace elements in PM_(10) samples ranged from 6966 to 12,958 *** the analyzed elements,Zn,Ti,Ni,Cu,Pb,ba,Mn,Cr and V were found at higher concentrations and accounted for over 95%of the total *** concentrations of total analyzed elements in the PM_(10) samples revealed a significant positive correlation with PM_(10) hemolytic *** the analyzed elements,Zn,Pb and Cs positively correlated with hemolysis,while Li,U and V negatively correlated with the hemolysis of human red blood cells(RbCs).Therefore,the heavy metal elements could be one of the main factors responsible for the hemolytic capacity of indoor PM_(10) in coal-burning environments.

Effects of high glucose and severe hypoxia on the biological behavior of mesenchymal stem cells at various passages

bACKGROUND Mesenchymal stem cells(MSCs)have been extensively studied for therapeutic potential,due to their regenerative and immunomodulatory *** passage and stress factors may affect the biological characteristics of MSCs,but the details of these effects have not been recognized *** To investigate the effects of stress factors(high glucose and severe hypoxia)on the biological characteristics of MSCs at different passages,in order to optimize the therapeutic applications of *** In this study,we investigated the impact of two stress conditions;severe hypoxia and high glucose on human adipose-tissue derived MSCs(hAD-MSCs)at passages 6(P6),P8,and ***,senescence and apoptosis were evaluated measuring WST-1,senescence-associated beta-galactosidase,and annexin V,*** Cells at P6 showed decreased proliferation and increased apoptosis under conditions of high glucose and hypoxia compared to control,while the extent of senescence did not change significantly under stress *** P8 hAD-MSCs cultured in stress conditions had a significant decrease in proliferation and apoptosis and a significant increase in senescence compared to counterpart cells at *** cultured in high glucose at P10 had lower proliferation and higher senescence than their counterparts in the previous passage,while no change in apoptosis was *** the other hand,MSCs cultured under hypoxia showed decreased senescence,increased apoptosis and no significant change in proliferation when compared to the same conditions at *** These results indicate that stress factors had distinct effects on the biological processes of MSCs at different passages,and suggest that senescence may be a protective mechanism for MSCs to survive under stress conditions at higher passage numbers.

Maize(Zea mays)endosperm is an important food source for humans and *** addition,it is considered an excellent model for research on seed development because of its relatively large size for both embryo and endosperm compared to those of other crop species,such as rice(Oryza sativa)and wheat(Triticum aestivum).After double fertilization,early maize endosperm development includes three stages:(i)a multinucleate coenocyte stage;(i)a cellularization stage,in which the cellular endosperm is formed;and(ii)a stage of differentiation into distinct *** early differentiation,four major cell types with specific functions differentiate within the endosperm:starchy endosperm,aleurone layer,basal endosperm transfer layer(bETL),and embryo surrounding *** subaleurone,conducting zone,basal intermediate zone,and endosperm adjacent to scutellum arise in late differentiation(Sabelli and Larkins 2009).After the main cell types are established,maize endosperm undergoes intensive cell proliferation,which marks the transition to the endosperm-filling *** the endosperm-flling stage,the maternal sucrose products are transported to seeds through the bETL region,where they will act as the main carbon substrate for storage products such as starch and proteins(Chen et al.,2023).In fact,the endosperm of maize seeds accounts for approximately 90%of the total grain weight,serving as an important nutrient reservoir(Flint-Garcia et al.,2009).
