Aszonapyrone A Isolated from Neosartorya spinosa IFM 47025 Inhibits the NF-κB Signaling Pathway Activated by Expression of the Ependymoma-Causing Fusion Protein ZFTA-RELA

Ependymoma is a rare and chemotherapy-resistant brain tumor, which has resulted in a delay in the development of drugs to treat it. A subclass of supratentorial ependymomas (ST-EPN), designated ST-EPN-zinc finger-translocation-associated (ZFTA, ST-EPN-ZFTA), exhibits the expression of a fusion protein comprising ZFTA and v-rel reticuloendotheliosis viral oncogene homolog A (RELA), an effector transcription factor of the nuclear factor-kappa B (NF-κB) pathway (ZFTA-RELA). The expression of ZFTA-RELA results in the hyperactivation of the oncogenic NF-κB signaling pathway, which ultimately leads to the development of ST-EPN-ZFTA. To identify inhibitors of the NF-κB signaling pathway activated by the expression of ZFTA-RELA, we used a doxycycline-inducible ZFTA-RELA-expressing NF-κB reporter cell line and found that extracts of the fungus Neosartorya spinosa IFM 47025 exhibited NF-κB inhibitory activity. We identified eight compounds [aszonapyrone A (2), sartorypyrone A (3), epiheveadride (4), acetylaszonalenin (5), (R)-benzodiazepinedione (6), aszonalenin (7), sartorypyrone E (8) and (Z, Z)-N,N’-(1,2-bis[(4-methoxyphenyl)methylene]-1,2-ethanediyl)bis-formamide (9)] from N. spinosa IFM 47025 culture extract using a variety of chromatographic techniques. The structures of these compounds were identified through the analysis of various instrumental data (1D, 2D-NMR, MS, and optical rotation). The NF-κB responsive reporter assay indicated that compounds 2, 3, 5, 7, and 9 exhibited inhibitory activity. We further evaluated the inhibitory activity of these compounds against the expression of endogenous NF-κB responsive genes (CCND1, L1CAM, ICAM1, and TNF) and found that compound 2 showed significant inhibitory activity. Further studies are required to elucidate the mechanism of action of compound 2, which may serve as a lead compound for the development of a novel therapy for ST-EPN-ZFTA.

Antifungal Activity of Compounds Isolated from Bamboo Vermicompost against <i>Rhizoctonia solani</i>AG1-IB

Vermicompost has become a promising alternative to chemical pesticide because of its disease suppression effect during these decades. However, the mechanisms by which vermicompost suppress plant disease are not well-understood. Antifungal compounds, which are released by beneficial organisms in the vermicompost, may play an important role in inhibiting plant pathogens;however, these mechanisms have not been widely examined. In this study, Sterilized Water Extracts of bamboo vermicompost (SWE), using a 0.22-μm cellulose acetate filter, significantly (P < 0.05) inhibited the mycelium growth of Rhizoctonia solani AG1-IB on a Potato Dextrose Agar (PDA) plate. This suggests that antifungal compounds are present in bamboo vermicompost. The ethanol acetate (EtOAc) crude extracts of bamboo vermicompost showing antifungal activity were further separated. Two compounds were isolated from the EtOAc fraction of bamboo vermicompost and characterized as ergosterol peroxide (1) and (22E, 24R)-5α,8α-epidioxyergosta-6,9(11),22-trien-3β-ol (2). Their chemical structures and mass spectra were determined by nuclear magnetic resonance and mass spectrometry analyses. Ergosterol peroxide tested at 150, 300, 600, and 900 μg showed 13%, 22%, 34%, and 53% mycelial growth inhibition against R. solani AG1-IB, respectively. Because EtOAc crude extracts of the initial substrate materials of bamboo vermicompost did not inhibit mycelium growth of R. solani AG1-IB, antifungal compounds in the vermicompost may be released by microbes but not from the original substrates during vermicomposting.

The Quantitative Density Measurement of Unsteady Flow around the Projectile

Three-dimensional density measurement of unsteady flow field around a sphere is carried out in the ballistic range at Institute of Fluid Science, Tohoku University. Simultaneous multi-angle measurement system using twelve digital cameras is installed in the test chamber of the ballistic range to achieve the three-dimensional density measurement. The Colored-Grid Background Oriented Schlieren (CGBOS) technique using colored-grid background is utilized for the reconstruction of density. The Mach number of the sphere is set to 1.5. The short pulse LEDs to illuminate the backgrounds are also installed in the chamber to capture the unsteady flow field around a flying sphere. Three-dimensional density distribution around a sphere is successfully reconstructed.

Two-dimensional quantum transport of multivalley （111） surface state in topological crystalline insulator SnTe thin films

The magneto-transport properties of （111）-oriented single-crystal thin films of SnTe were investigated. SnTe （111） thin films were epitaxially grown on a BaF2 substrate by molecular beam epitaxy. By optimizing the growth conditions and the thickness of the films, the bulk carrier density could be reduced, making it possible to detect the surface transport. Magneto-conductance （MC） measurements show a cusp-like feature around zero magnetic field, which is attributed to the weak-antilocalization effect of the transport in the topological surface state. Detailed analysis of this negative MC reveals a reduced number of transport channels contributing to the surface transport, suggesting strong coupling between Dirac valleys on the SnTe （111） surface, as a characteristic feature of the transport in the multivalley structure of topological crystalline insulators.

Stretching Induces the Rearrangement of the Periodontal Ligament Cells without Altering the Orientation of Oxytalan Fibers Relative to the Cell Axis in Vitro

The periodontal ligament (PDL) contains oxytalan fibers as well as collagen fibers, which helps it to withstand the mechanical stress to which it is constantly exposed. The oxytalan fibers are produced by PDL fibroblasts. However, the arrangement of PDL fibroblasts and the orientation of oxytalan fibers relative to the fibroblast cell axis have not been investigated under the condition of mechanical stress. We hypothesized that such stress would alter the arrangement and orientation of these cells and their oxytalan fibers. The aim of this study was to evaluate the effects of stretching strain on PDL fibroblasts, focusing on the cellular arrangement and orientation of oxytalan fibers relative to the long cell axis in cell/matrix layers by staining the major component of the fibers, fibrillin-1. The angle between the long cell axis and the oxytalan fibers was approximately 70 degrees under both non-stretching and stretching conditions. Moreover, stretching induced the rearrangement of the cells. This is the first study to demonstrate that stretching induces the rearrangement of the PDL fibroblasts without altering the angle between the long cell axis and the oxytalan fibers. These results may reflect the orientation of oxytalan fibers in the PDL under the condition of mechanical stress.

Prognostic factors of minimally invasive surgery for gastric cancer: Does robotic gastrectomy bring oncological benefit?

Preparation of PDOT:PSS Transparent Conductive Film Using Ink-Jet Printing

Flexible devices produced using organic materials have attracted the attention of many researchers. Important components of these flexible devices include transparent electrodes, which transmit visible light and possess conductivity. The present study improved the characteristics of a transparent conductive film that was made of poly(3, 4 ethylenedioxythiophene):poly(styrenesul-fonate) (PEDOT:PSS), an organic conductive material, and that had been prepared using ink-jet printing. To improve the resistance value and visible light transmittance of the film, the film substrate was first cleaned with ultraviolet/ozone treatment, and then the film was annealed after it was deposited on the substrate and dipped into a polar solvent. Consequently, the resistance value of the thin film decreased. However, the surface state of the film changed according to the treatment method and affected its visible light transmittance. Thus, the surface state of the film substrate, the annealing temperature after film deposition, and the dipping treatment with a polar solvent influenced the characteristics of a thin film.
