The treatment of advanced pancreatic cancer has not moved much beyond single agent gemcitabine until recently when protocols such as FOLFIRINOX(fluorouracil,leucovorin,irinotecan and oxaliplatin)and nab-paclitaxelgemcitabine have demonstrated some improved *** in technology especially in massively parallel genome sequencing has progressed our understanding of the biology of pancreatic cancer especially the candidate signalling pathways that are involved in tumourogenesis and disease *** has allowed identification of potentially actionable mutations that may be targeted by new biological *** heterogeneity of pancreatic cancer makes tumour tissue collection important with the aim of being able to personalise therapies for the individual as opposed to a one size fits all approach to treatment of the *** paper reviews the developments in this area of translational research and the ongoing clinical studies that will attempt to move this into the everyday oncology practice.

Efficiency of sample-based indices for spatial pattern recognition of wild pistachio(Pistacia atlantica) trees in semi-arid woodlands

The efficiency of sample-based indices proposed to quantify the spatial distribution of trees is influenced by the structure of tree stands, environmental heterogeneity and degree of aggregation. We evaluated 10 commonly used distance-based and 10 density-based indices using two structurally different stands of wild pistachio trees in the Zagros woodlands, Iran, to assess the reliability of each in revealing stand structure in woodlands. All trees were completely stem-mapped in a nearly pure（40 ha） and a mixed（45 ha） stand. First, the inhomogeneous pair correlation function [g（r）] and the Clark-Evans index（CEI） were used as references to reveal the true spatial arrangement of all trees in these stands. The sampled data were then evaluated using the 20 *** was undertaken in a grid based on a square lattice using square plots（30 m 9 30 m） and nearest neighbor distances at the sample points. The g（r） and CEI statistics showed that the wild pistachio trees were aggregated in both stands, although the degree of aggregation was markedly higher in the pure stand. Three distance- and six density-based indices statistically verified that the wild pistachio trees were aggregated in both stands. The distance-based Hines and Hines statistic（ht） and the densitybased standardised morisita（Ip）, patchiness（IP） and Cassie（CA） indices revealed aggregation of the trees in the two structurally different stands in the Zagros woodlands and the higher clumping in the pure stand, whereas the other indices were not sensitive enough.

Reduced expression ofβ-catenin inhibitor Chibby in colon carcinoma cell lines

AIm： To analyse the Chibby expression and its function in colon carcinoma cell lines and colorectal carcinoma （CRC）. mETHODS： Chibby expression levels were investigated by quantitative RT-PCR in a panel of seven different colon carcinoma cell lines. By sequencing, we analysed mutational status of Chibby. To test whether Chibby exhibited effects on β-catenin signalling in colon carcinoma cells, we transfected SW480 cells with Chibby expression plasmid and, subsequently, analysed activity of β-catenin and tested for alterations in cellular phenotype. In addition, we examined Chibby mRNA levels in samples of colorectal carcinomas and adjacent normal tissues by using quantitative RT-PCR and hybridised gene chips with samples from CRC and normal tissues. RESULTS： Chibby mRNA expression was strongly downregulated in colon carcinoma cell lines in comparison to normal colon epithelial cells and no mutation in any of the examined colon carcinoma cell lines was found. Further, we could show that Chibby inhibited β-catenin activity in TOPflash assays when over-expressed in SW480 cells. Proliferation and invasion assays with Chibby transfected SW480 cells did not reveal profound differences compared to control cells. In contrast to these in vitro data, quantitative RT-PCR analyses of Chibby mRNA levels in CRC tumor samples did not show significant differences to specimens in adjacent non-cancerous tissue. Consistent with these findings, gene chips analysing tissue samples of tumors and corresponding normal tissue did not show altered Chibby expressionCONCLUSION： Altered Chibby expression might be observed in vitro in different colon carcinoma cell lines. However, this finding could not be confirmed in vitro in CRC tumors, indicating that Chibby is not likely to promote CRC tumor development or progression. Ferredoxin-NADP＋ oxidoreductase （FNR）, functioning in the last step of the photosynthetic electron transfer chain, exists both as a soluble protein in the chloroplast stroma and tightly attached to chloroplast membranes. Surface plasmon resonance assays showed that the two FNR isoforms, LFNR1 and LFNR2, are bound to the thylakoid membrane via the C-terminal domains of Tic62 and TROL proteins in a pH-dependent manner. The tic62 trol double mutants contained a reduced level of FNR, exclusively found in the soluble stroma. Although the mutant plants showed no visual phenotype or defects in the function of photosystems under any conditions studied, a low ratio of NADPH/NADP~ was detected. Since the CO2 fixation capacity did not differ between the tic62 trol plants and wild-type, it seems that the plants are able to funnel reducing power to most crucial reactions to ensure survival and fitness of the plants. However, the activity of malate dehydrogenase was down-regulated in the mutant plants. Apparently, the plastid metabolism is able to cope with substantial changes in directing the electrons from the light reactions to stromal metabolism and thus only few differences are visible in steady-state metabolite pool sizes of the tic62 trol plants.

Background Percutaneous left atrial appendage occlusion(LAAO)requires puncture of the interatrial *** immediate hemodynamic effects of iatrogenic atrial septal defects(iASD)after LAAO have not been examined so *** aimed at evaluat-ing these effects through invasive measurements of pressure and oxygen ***,we assessed the incidence of per-sistent iASD at three *** Forty-eight patients scheduled for percutaneous LAAO were prospectively included in the *** and oxygen saturation were measured(1)in the right atrium(RA)before transseptal puncture,(2)in the left atrium(LA)through the transseptal sheath after transseptal puncture,(3)in the LA after removal of introducer sheath,and(4)in the RA after removal of introducer *** echocardiography was performed at three months to detect *** Pressure in the RA increased significantly after removing the introducer sheath(P=0.034),whereas no difference was found in oxygen saturation in the RA(P=0.623).Pressure measurement in the LA showed no significant difference after re-moving the introducer sheath(P=0.718).Oxygen saturation in the LA also showed no significant difference(P=0.129).Follow-up transesophageal echocardiogram at 3 months revealed a persistent iASD in 4 patients(8.5%).CONCLUSIONS Our study suggests that iASD after percutaneous LAAO does not result in significant shunts directly after the procedure,although a significant increase of mean right atrial pressure can be *** iASDs after percutaneous LAAO seem to be relatively rare at three months.
