Eremurus species,better known as‘Foxtail Lily’or‘Desert Candle’,are important worldwide in landscaping and the cutflower *** of the centers of highest diversity of the genus Eremurus is Iran,which has seven ***,little is known about the genetic diversity within the genus *** the advent of genotyping-bysequencing(GBS),it is possible to develop and employ single nucleotide polymorphism(SNP)markers in a costefficient manner in any species,regardless of its ploidy level,genome size or availability of a reference *** structure and phylogeographic analyses of the genus Eremurus in Iran using a minimum of 3002 SNP markers identified either at the genus level or at the species level from GBS data showed longitudinal geographic structuring at the country scale for the genus and for the species *** and ***,and at the regional scale for *** analyses furthermore showed a close genetic relatedness between *** and *** to the extent that they should be considered subspecies within an ***/stenophyllus species *** close genetic relatedness may explain why crosses between these two(sub)species have been found in the wild and are exploited extensively as ***,current species identification,while robust,relies on flower morphology.A subset of seven SNPs with species-specific(private)alleles were selected that differentiate the seven Eremurus *** markers will be especially useful for cultivar protection and in hybrid production,where true hybrids could be identified at the seedling stage.

Background:Emerging epidemiological evidence suggests that compared to engaging in 1 activity mode alone,a combination of moderate-tovigorous physical activity(mVPA:brisk walking/jogging,cycling)and muscle-strengthening exercise(mSE:push-ups/sit-ups,using weight machines)has more favorable associations with optimal weight ***,few studies have examined the dose-dependent and joint associations of mVPA and mSE with ***:Based on cross-sectional analyses of the European Health Interview Survey Wave 2(2013-2014),we examined prevalence ratios(PRs)of joint and stratified associations between mVPA(4 categories:(ⅰ)0 min/week,(ⅱ)1-149 min/week,(ⅲ)150-299 min/week,and(ⅳ)≥300 min/week)and mSE(3 categories:(ⅰ)0 day/week,(ⅱ)1 day/week,and(ⅲ)>2 days/week)with body mass index-defined obesity(body mass index of>30.0 kg/m2)using Poisson regression with robust error *** were examined unadjusted and adjusted for sociodemographic and lifestyle characteristics(e.g.,sex,age,education,income,and smoking status).Results:Data were available for 280,456 adults(≥18 years),of which 46,166(15.5%)were *** interaction mVPA x mSE guideline adherence was statistically significant for obesity(p≤0.05).The joint mVPA-mSE analysis showed that compared to the reference group(i.e.,no mVPA and no mSE),the PRs followed a dose-dependent pattern,with the lowest observed among those reporting≥150 mVPA min/week and≥1 mSE days/week(PR:0.43;95%confidence interval:0.41-0.46).When stratified across each mVPA strata,the PRs were mostly lower among those engaging in mSE 1 day/week,as compared to those doing mSE≥2 days/***:"There was evidence for a dose-dependent association between joint mVPA-mSE with a reduced prevalence of *** health strategies for the prevention and management of obesity should recommend both mVPA and mSE.

IL-34 is a novel biomarker for rheumatoid arthritis(RA),as its serum levels are linked to C-reactive protein,erythrocyte sedimentation rate,rheumatoid factor(RF),IL-6 and radiographic progression[1,2].IL-34 reprograms naïve circulating m0 cells into glycolytic(CD14^(+)CD86^(+)GLUT1^(high))m34 macrophages(mΦs)by binding to its pathogenic receptor,SDC-1,which controls the activation of its coreceptor/m-CSFR[3].Glycolytic m34 mΦs are primed to differentiate into osteoclasts through SDC-1 ligation[3].

Flares of joint inflammation and resistance to currently available biologic therapeutics in rheumatoid arthritis(RA)patients could reflect activation of innate immune ***,we show that a TLR7 GU-rich endogenous ligand,miR-Let7b,potentiates synovitis by amplifying RA monocyte and fibroblast(FLS)***-Let7b ligation to TLR7 in macrophages(mΦs)and FLSs expanded the synovial inflammatory ***,secretion of m1 monokines triggered by miR-Let7b enhanced Th1/Th17 cell *** showed that IRAK4 inhibitor(i)therapy attenuated RA disease activity by blocking TLR7-induced m1 mΦor FLS activation,as well as monokine-modulated Th1/Th17 cell ***4i therapy also disrupted RA osteoclastogenesis,which was amplified by miR-Let7b ligation to joint myeloid ***,the effectiveness of IRAK4i was compared with that of a TNF inhibitor(i)or anti-IL-6R treatment in collagen-induced arthritis(CIA)and miR-Let7b-mediated *** found that TNF or IL-6R blocking therapies mitigated CIA by reducing the infiltration of joint F480+iNOS+mΦs,the expression of certain monokines,and Th1 cell ***,these biologic therapies were unable to alleviate miR-Let7b-induced *** superior efficacy of IRAK4i over anti-TNF or anti-IL-6R therapy in miR-Let7b-induced arthritis or CIA was due to the ability of IRAK4i therapy to restrain the migration of joint F480+iNOS+mΦs,vimentin+fibroblasts,and CD3+T cells,in addition to negating the expression of a wide range of monokines,including IL-12,mIP2,and IRF5 and Th1/Th17 *** conclusion,IRAK4i therapy may provide a promising strategy for RA therapy by disconnecting critical links between inflammatory joint cells.

Synovial macrophages are crucial in the development of joint inflammation and bone damage;however, the pathways that controlmacrophage remodeling in inflammatory m1 cells or bone-eroding osteoclasts are not fully understood. We determined thatelevated IL-7R/CD127 expression is the hallmark of rheumatoid arthritis (RA) m1 macrophages and that these cells are highlyresponsive to interleukin-7 (IL-7)-driven osteoclastogenesis. We established that lipopolysaccharide (LPS), interferon-γ (IFNγ), andtumor necrosis factor-α (TNFα), the classic m1 macrophage mediators, enhance IL-7R expression in RA and murine *** local expression of IL-7 provokes arthritis, predominantly through escalating the number of F480^(+)iNOS^(+) cells rather than CD3^(+)T cells. Ectopic LPS injection stabilizes IL-7-induced arthritis by increasing myeloid IL-7R expression, in part via IFNγ ***, in RAG−/− mice, IL-7-mediated arthritis is suppressed because of the reduction in myeloid IL-7R expression due to the lackof IFNγ. moreover, the amelioration of IL-7-induced arthritis by anti-TNF therapy is due to a decrease in the number of cells in theunique F480^(+)iNOS^(+)IL-7R^(+)CCL5^(+) subset, with no impact on the F480^(+)Arginase^(+) cell or CD3^(+) T cell frequency. Consistent with thepreclinical findings, the findings of a phase 4 study performed with RA patients following 6 months of anti-TNF therapy revealedthat IL-7R expression was reduced without affecting the levels of IL-7. This study shifts the paradigm by discovering that IL-7-induced arthritis is dependent on F480^(+)iNOS^(+)IL-7R^(+)CCL5^(+) cell function, which activates TH-1 cells to amplify myeloid IL-7Rexpression and disease severity.

Predictors for massive Haemorrhage during Caesarean Delivery Due to Placenta Praevia

Objectives: To describe the incidence and predictive factors for massive haemorrhage during a caesarean delivery in women with placenta praevia. methods: We performed a retrospective cohort study among consecutive women with a placenta praevia planned for a caesarean delivery between 2001-2011 in one academic centre and two teaching hospitals in the Netherlands. massive haemorrhage was defined as ≥1000 mL blood loss during caesarean section. We used logistic regression analysis to assess which maternal and pregnancy characteristics were predictors for massive haemorrhage during caesarean delivery. Results: Of 54,794 deliveries, we identified 215 (0.39%) women with placenta praevia who underwent a Caesarean delivery, of whom 94 (44%) had massive haemorrhage at or after surgery. After univariable preselection, multivariable logistic regression indicated that higher maternal age (OR 2.09;95% CI 1.17 - 3.74), no Caucasian ethnicity (OR 1.73;95% CI 0.92 - 3.27), multiple pregnancy (OR 3.92;95% CI 0.72 - 21.28), lower systolic blood pressure during pregnancy (OR 1.03;95% CI 1.00 - 1.07) and the placenta located at the anterior wall (OR 2.21;95% CI 1.20 - 4.04) were independent predictors for massive haemorrhage during a caesarean delivery in women with placenta praevia. Predicted probabilities varied between 22% and 71%. The model has a reasonable discriminative ability and acceptable calibration. Conclusions: Women with placenta praevia are at high risk for massive haemorrhage during caesarean delivery. The risk profile can be used to identify extreme high-risk women in whom extensive preventive measures are justified.
