Transient and stable expression of transgenes is central to many investigations in plant biology research. Chemical regulation of expression can circumvent problems of plant lethality caused by constitutive overexpression or allow inducible knock （out/down） approaches. Several chemically inducible or repressible systems have been described and successfully applied. However, cloning and application-specific modification of most available inducible expression systems have been limited and remained complicated due to restricted cloning options. Here we describe a new set of 57 vectors that enable transgene expression in transiently or stably transformed cells. All vectors harbor a synthetically optimized XVE expression cassette, allowing I^-estradiol mediated protein expression. Plasmids are equipped with the reporter genes GUS, GFP, mCherry, or with HA and Strepll epitope tags and harbor an optimized multiple cloning site for flexible and simple clon- ing strategies. Moreover, the vector design allows simple substitution of the driving promoter to achieve tissue-specificity or to modulate expression ranges of inducible transgene expression. We report details of the kinetics and dose-dependence of expression induction in Arabidopsis leaf mesophyll protoplasts, transiently transformed Nicotiana benthamiana leaves, and stably transformed Arabidopsis plants. Using these vectors, we investigated the influence of CBL （Calcineurin B-like） protein expression on the subcellular localization of CIPKs （Calcineurin B-like interacting protein kinases）. These analyses uncovered that induced co-expression of CBL3 is fully sufficient for dynamic translocation of CIPK5 from the cytoplasm to the tonoplast. Thus, the vector system presented here facilitates a broad range of research applications.

Photorespiration is an essential metabolic process in leaves that facilitates recovery of carbon lost by the oxygenase reaction of Rubisco and avoids the accumulation of the toxic product, 2-phosphoglycolate （2PG） of this reaction （Bauwe et al., 2012）. However, there is also evidence to suggest that photorespiration has a more complex role during normal growth than the mere detoxification of 2PG and the recovery of 3-phosphoglycerate （3PGA）（Bauwe et al., 2012）.

Novel Screen-Printed All-Solid-State Copper(II)-Selective Electrode for Mobile Environmental Analysis

Based on poly(vinyl chloride) membranes, a novel miniaturized screen-printed all-solid-state copper(II)-selective electrode has been developed for applications in environmental monitoring. Performance and applicability of the ion-selective electrode (ISE) have been proved by potentiometric investigations. Conducting polymers were used as intermediate layers and as solid contacts between the ion-selective membrane and the graphite transducer. The ion-complexing reagent 2-mercapto-benzoxazole was incorporated into poly(vinyl chloride) membranes. In the concentration range 10-6 - 10-2 mol/L, the ISE exhibited a linear Nernstian potential response to copper(II) with an average slope value of 28 mV/decade. The detection limit was 3 × 10-7 mol/L. The electrode exhibits a short response time (<10 s) and can be used in the range of pH = 3 - 7. Selectivity coefficents against certain interfering ions are investigated. The life time of the electrode under laboratory conditions was approximately 12-month. The electrode was applied in the investigation of different aqueous environmental samples and the electrode characteristics were described. The copper(II) ASS electrode has also successfully been used in potentiometric, complexometric titrations with ethylenediaminetetraacetic acid.

PPCPs - A human and veterinary fingerprint in the Pearl River delta and northern south China sea

Solid-Contact Ion-Selective Electrodes Based on Graphite Paste for Potentiometric Nitrate and Ammonium Determinations

Robust and easy-to-handle solid-contact ion-selective electrodes (SC-ISEs) based on graphite paste have been developed for the potentiometric detection of NO-3 and NH+4 in environmental samples. Polypyrrole (PPy) has been used as intermediate layer and solid contact between the electron-conducting graphite paste substrate and the ion-conducting polyvinylchloride (PVC)-containing membrane with the immobilized ionophore. For the nitrate- and ammonium-selective electrodes, tridodecylmethylammonium nitrate (TDMA-NO3) and nonactin have been used as ion-complexing compounds, respectively. Both ISEs show nearly Nernstian response in the linear range from 10-5 mol/L to 10-1 mol/L with average slopes of >58 mV/decade. The ISEs possess short response times (<20 s).

All-Solid-State Screen-Printed Sensors for Potentiometric Calcium(II) Determinations in Environmental Samples

This paper describes preparation, characterization and electrochemical performance of novel planar miniaturized all-solid-state (ASS) screen-printed potentiometric sensors for the detection of Ca2+ ions in environmental samples. Screen-printed graphite-based ion-selective electrodes (ISEs) and screen-printed reference electrodes based on silver-containing pastes have been applied in a space saving manner on common ceramic substrates with small dimensions. Applications to environmental samples are shown by direct potentiometry and potentiometric titrations in real water samples. Conducting polymers (CPs) have been used as solid-contact materials and as intermediate layer between the polyvinyl chloride (PVC)-containing ion-selective membrane and the graphite-containing substrate. Different diamides have been incorporated into the PVC membrane. In the range from 10-4 mol/L to 10-1 mol/L, the ISEs show linear slopes of 27 mV/decade, which is close to the Nernstian response. Moreover, the ISEs have response times of 6 months. The novel potentiometric ASS sensors enable simple and exact Ca2+ determinations in real samples.

Attachment in Children with ADHD

Objective: Parallels in findings of attachment and ADHD research suggest a connection between both constructs. However, the few articles dealing with that tie investigate children with all conceivable expressions of ADHD and have not found sufficient evidence regarding the connection between ADHD and attachment. This study sought to collect evidence of a possible connection between the two aspects among children with the predominantly inattentive ADHD type. Method: The sample consisted of 93 children aged 5 - 9 years, 48 of whom had an ADHD diagnosis of the predominantly inattentive type. Attachment was coded using the German adaptation of the Attachment Story Completion Task, externalizing behavior problems were rated using the Child Behavior Checklist 4 - 18. ADHD was assessed using both clinical diagnoses as well as the German ADHD Rating scale. Results: Results revealed a large difference in the distribution of attachment classification between children with ADHD and those without ADHD. Regression analysis, however, showed no independent effect on ADHD when externalizing behavior problems were controlled for. Conclusions: Both constructs seem to be connected, however, in the investigation of the influence of attachment on the disorder, ADHD as a global construct is too unspecific. As opposed to previous studies, considering only the inattentive type, the relation might completely be moderated by externalizing behavior problems. Subsequent studies should first focus on specific symptom patterns which at best should be analyzed within longitudinal studies.
