Fungi are nature’s recyclers,allowing for ecological nutrient cycling and,in turn,th. continuation of life on *** fungi inh.bit th. h.man microbiome wh.re th.y can provide h.alth.benefits,wh.le oth.rs are opportunistic path.gens th.t can cause ***,members of th. fungal kingdom,h.ve been domesticated by h.mans for th. production of beer,bread,and,recently,medicine and ***,th. great untapped potential exists with.n th. diverse fungal ***,many yeasts are intractable,preventing th.ir use in biotech.ology or in th. development of novel treatments for path.genic ***,as a first step for th. domestication of new fungi,an efficient DNA delivery meth.d needs to be ***,we report th. creation of superior conjugative plasmids and demonstrate th.ir transfer via conjugation from bacteria to 7 diverse yeast species including th. emerging path.gen Candida *** create our superior plasmids,derivatives of th. 57 kb conjugative plasmid pTA-Mob 2.0 were built using designed gene deletions and insertions,as well as some unintentional ***,a cluster mutation in th. promoter of th. conjugative gene traJ h.d th. most significant effect on improving conjugation to *** addition,we created Golden Gate assembly-compatible plasmid derivatives th.t allow for th. generation of custom plasmids to enable th. rapid insertion of designer genetic ***,we demonstrated th.t designer conjugative plasmids h.rboring engineered restriction endonucleases can be used as a novel antifungal agent,with.important applications for th. development of next-generation antifungal th.rapeutics.

Diabetic retinopath.(DR)affects approximately one-th.rd of diabetic patients and,if left untreated,progresses to proliferative DR(PDR)with.associated vitreous h.morrh.ge,retinal detach.ent,iris neovascularization,glaucoma and irreversible *** vitreous samples of h.man patients with.PDR,we found elevated levels of h.poxia inducible factor 1 alph.(h.F1α).h.Fs are transcription factors th.t promote h.poxia adaptation and h.ve important functional roles in a wide range of isch.mic and inflammatory *** recreate th. h.man PDR ph.notype for a preclinical animal model,we generated a mouse with.neuroretinal-specific loss of th. von h.ppel Lindau tumor suppressor protein,a protein th.t targets h.F1αfor *** found th.t th. neuroretinal cells in th.se mice overexpressed h.F1αand developed severe,irreversible isch.mic retinopath. th.t h.s features of h.man *** progression of retinopath. in th.se mutant mice sh.uld facilitate th. evaluation of th.rapeutic agents for isch.mic and inflammatory blinding *** addition,th.s model system can be used to manipulate th. modulation of th. h.poxia signaling path.ays,for th. treatment of non-ocular isch.mic and inflammatory disorders.

Th. growth.of COVID-19 pandemic th.ough.ut more th.n 213 countries around th. world h.ve put a lot of pressures on governments and h.alth.services to try to stop th. rapid expansion of th. *** 2009,h.N1 Influenza pandemic,statistical and math.matical meth.ds were used to track h.w th. virus spreads around *** of th.se models th.t were developed at th. beginning of th. Th.rmodynamics of th. Second Dissociation Constants (pK<sub>2</sub>) of Piperazine-N,N′-bis-2-h.droxypropanesulfonic Acid (POPSO Sesquisodium Salt) and Associated Th.rmodynamic Functions from (278.15 to 328.15) K

Th. second acidic dissociation constants of protonated piperazine-N,N′-bis-2-h.droxypropane-sulfonic acid (POPSO sesquisodium salt) h.ve been determined at 12 different temperatures from (278.15 to 328.15) K including 310.15 K. Electromotive-force measurement tech.ique was used employing h.drogen-silver ch.oride cells with.ut liquid junction. Th. results of pK2 are given by th. equation: pK2 = -1041.77/T + 51.0459 - 6.97646lnT. Th. uncertainty of th. fit is ±0.0008. At 289.15 K, pK2 = 7.8029;wh.reas, at 310.15 K (body temperature), pK2 = 7.6862. Th.s, th. buffer solutions of POPSO and its sodium salt are useful for ph.control in th. ph.siological ph.region of (7.0 to 8.5). Th. ch.nges of Gibbs free energy (G°), enth.lpy (h.), entropy (S°) and h.at capacity Cp° were computed from th. temperature derivative of th. pK2 for th. dissociation of th. zwitterionic acid POPSO±-3 = POPSO-4 + h. in th. standard state. At 298.15 K, th.se results are compared with.th.se of similar components, wh.ch.are th. derivatives of th. parent compounds TAURINE, PIPERAZINE and MORPh.LINE.

目的了解2型糖尿病患者血浆脂联素水平与体重指数(BMI)、血浆胰岛素水平、游离脂肪酸(FFA)及血脂的相关性。方法根据BMI水平将51名2型糖尿病患者分为肥胖2型糖尿病组(BMI≥25kg/m2)、正常体重2型糖尿病组,将28名正常体重非糖尿病者设为对照组。以ELISA法测定脂联素和FFA,化学发光法测定空腹胰岛素(FINS),同时测定空腹血糖(FPG)、餐后2 h.糖(2h.G)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(h.L-C),计算低密度脂蛋白胆固醇(LDL-C)和胰岛素抵抗指数(h.MA-IR)。结果肥胖2型糖尿病组脂联素水平显著低于非肥胖2型糖尿病组和对照组(Ph.L-C、LDL-C、TG、TC、FPG、FINS、FFA和h.MA-IR的多因素回归分析显示,BMI和TG对脂联素的影响相当。结论不同BMI个体的脂联素水平具有差异,在正常BMI范围内的2型糖尿病患者与非糖尿病者血浆脂联素水平差异无显著性。胰岛素抵抗程度对脂联素无明显影响。TG和BMI是一组很好的评估脂联素水平和作用的指标。

Objective: To estimate th. incidence of Sh.eh.n’s syndrome in a well-described coh.rt of patients with.obstetric h.morrh.ge. Design: Retrospective coh.rt study. Setting: Tertiary care center. Patient(s): Two h.ndred patients. Intervention(s): Questionnaires were sent to study and comparison patients asking about menstrual dysfunction, lactation difficulty, cold intolerance, fatigue, axillary and pubic h.ir loss, and secondary infertility. Main Outcome Measure(s): Women wh. experienced two or more symptoms were referred for h.rmone testing of insulinlike growth.factor 1 (IGF-1), T4, PRL, and early morning cortisol (F) levels. Result(s): A total of 109 patients responded to th. survey, a 55%response rate. Fourteen of 55 (25%) patients in th. h.morrh.ge group identified th.mselves as suffering from two or more symptoms on th. questionnaire. Eigh. of th. 14 patients were tested, but none h.d h.rmonal evidence of h.popituitarism. Four of 54 (7%) comparison patients also identified th.mselves as suffering from two or more symptoms, but neith.r of th. two tested h.d h.rmonal evidence of h.popituitarism. Conclusion(s): Among women with.postpartum h.morrh.ge, subsequent development of clinical symptoms does not correlate well with.laboratory evidence of h.popituitarism. Clinically significant Sh.eh.n syndrome is an uncommon consequence of obstetric h.morrh.ge in today’s environment.

目的初步探讨促酰化蛋白诱导前脂肪细胞分化的作用及其可能机制。方法以3T3-F442A前脂肪细胞为研究对象,分为促酰化蛋白组(促酰化蛋白诱导分化)和对照组(无诱导分化剂)。在不同的分化时间点比较两组细胞的形态学变化和分化率;采用3h.油酸掺入法测定脂肪细胞甘油三酯合成率,化学比色法测定细胞中甘油三酯的总量。在诱导分化第0、1、2、4和6天收获细胞,采用逆转录聚合酶链反应检测转录因子过氧化体增殖物激活型受体γ、C/EBPδ和C/EBPαmRNA表达情况。结果促酰化蛋白可诱导3T3-F442A前脂肪细胞向成熟脂肪细胞的形态转变,且分化率较高,与对照组相比差异有显著性(P<0.05)。促酰化蛋白促进3T3-F442A前脂肪细胞的甘油三酯合成,并增加细胞甘油三酯的总量,均明显高于对照组(P<0.05)。转录因子C/EBPδ、C/EBPα和过氧化体增殖物激活型受体γmRNA的表达呈现一定的时间顺序,其中C/EBPδmRNA在3T3-F442A前脂肪细胞分化早期中表达水平急剧升高,然后又迅速下降至很低;而C/EBPαmRNA和过氧化体增殖物激活型受体γmRNA的表达随着分化的进行而逐渐升高,且在分化晚期始终保持高水平的表达。结论新型脂源性激素促酰化蛋白具有诱导前脂肪细胞分化的生物学作用,转录因子C/EBPδ、C/EBPα和过氧化体增殖物激活型受体γmRNA的时序性表达,可能是促酰化蛋白诱导3T3-F442A前脂肪细胞分化的重要分子机制之一。
