An extended treatment protocol with pegylated interferon and ribavirin for hepatitis C recurrence after liver transplantation

AIM: To evaluate the efflicacy and tolerability of an extended treatment protocol and to determine the predictors of sustained virological response (SVR) after liver transplantation (lT).METHODS: Between August 2005 and November 2008, patients with recurrent hepatitis C virus (HCV) after lT were selected for treatment if liver biopsy showed at least grade 2 inflammation and/or stage 2 flibrosis. All patients were to receive pegylated interferon (PEG)/regimens combining ribavirin (RBV) for an additional 48 wk after HCV ***: Extended protocol treatment was initiated in thirty patients. Overall, 73% had end of treatmentresponse and 60% had SVR. Nineteen patients completed treatment per protocol, of them, sixteen (84%) had end of treatment response, and fourteen (74%) achieved SVR. Both early virological response and 24-week virological response were individually associated with SVR but this association was not signif icant on multivariate analysis. Eleven patients (37%) discontinued therapy due to adverse effects. Cytopenias were the most common and most severe adverse effect, and required frquent growth factor use, dose adjustments and treatment cessations. The risk of rejection was not ***: Recurrent HCV after lT can be safely treated with extended virological responseguided therpy using PEG/RBV, but requires close monitoring for treatment-related adverse effects, particularly cytopenias.

The recommended therapy for chronic hepatitis C (CHC) infection is the combination of a Pegylated interferon and Ribavirin. Almost all such patients on combination therapy experience one or more adverse events during the course of treatment. Significant neurological side effects are rare. A few cases of Bell's Palsy, chronic inflammatory demyelinating polyneuropathy and even one case of acute demyelinating polyneuropathy with atypical features for Guillain-Barre syndrome (GBS) associated with Interferon therapy have been reported but no report of GBS with typical features has been published. We present a case report of typical GBS associated with Peginterferon alfa-2a and Ribavirin used for treatment of CHC infection.

Validation of an Optimized Spectrophotometric Method for the Selective Determination of labetalol Hydrochloride

A simple sensitive and economical method for the determination of labetalol hydrochloride has been proposed, based on the reaction of labetalol with sodium nitroprusside and hydroxylamine hydrochloride in sodium dihydrogen phosphate-sodium hydroxide buffer solution of pH 12. The green-blue color produced due to the formation of a nitroso derivative has been measured at 695 nm. The Beer＇s range was obeyed in the concentration range of 2-51 μg·ml^-1 with molar absorptivity of 0.48 × 10^4 l·mol^-1·cm^-1. Rigorous statistical analyses were performed for the validation of the method. A detailed investigation of the selectivity of the method has been done to find it to be highly selective for the determination of labetalol hydrochloride in the presence of its acidic degradation product and common excipients of formulations. The proposed method was successfully applied to the determination of labetalol hydrochloride in the laboratory prepared dosage forms. Comparison of the means of the proposed procedure with a reference method using point as well as interval hypotheses showed no statistically significant difference. The developed method was extended to investigate its applicability to biological samples.

Designing single-atom nanozymes with densely exposed metal atom active sites and enhancing catalytic activity to detect pollutants remain a serious ***,we reported a single-atom nanozyme with layered stacked Fe/Cu dual active sites(Fe/Cu-NC SAzyme)synthesized via hydrothermal and hightemperature pyrolysis using folic acid as a *** with Fe-NC and Cu-NC SAzyme,Fe/Cu-NC SAzyme has higher peroxidase-like activity,which indicates that the doping of synthesized Fe/Cu bimetals can improve the catalytic activity and that the atomic loading of Fe and Cu in Fe/Cu-NC is 5.5 wt%and 2.27 wt%,*** S^(2-)is added to the Fe/Cu-NC catalytic system,a high-sensitivity and high-selectivity S^(2-)colorimetric sensing platform can be established,with a wide linear range(0.09-6μmol/l)and a low detection limit(30 nmol/l),which can be used to detect S^(2-)in environmental water ***’s more,the Fe/Cu-NC SAzyme can activate peroxymonosulfate(PMS)to degrade 99.9%of rhodamine B(Rh B)within 10 min with a degradation kinetics of 0.5943 min^(-1).This work details attractive applications in Fe/Cu-NC SAzyme colorimetric sensing and dye degradation.

Adsorption of phosphate on magnetite-enriched particles (MEP) separated from the mill scale

Phosphate is a major pollutant in water, causing serious environmental and health consequences. In present study, the phosphate adsorption on novel magnetite-enriched particles (MEP) was comprehensively investigated. A new method and device were introduced for the separation of MEP from the mill scale at low magnetic intensity. Particles were characterized with different techniques such as XRD, XRF, SEM and EDS. The XRD and XRF analysis of MEP identified the dominant existence of crystalline magnetite. Furthermore, the morphological analysis of MEP confirmed the agglomerate porous morphology of magnetite. Oxygen and iron, the main constituents of magnetite were acknowledged during the elemental analysis using EDS. The phosphate adsorption on MEP is well explained using various isotherm and kinetic models, exhibiting the monolayer adsorption of phosphate on the surface of MEP. The maximum adsorption capacity was determined 6.41 mg/g. Based on particle size (45–75 and 75–150 μm) and empty bed contact time (1 and 2 h), four columns were operated for 54 days. MEP were appeared successful to remove all phosphate concentration from the column influent having 2 mg/l concentration. The operated column reactors were successfully regenerated with alkaline solution. The results indicated potential for practical application of the MEP for phosphate removal.

目的研究单核细胞增生李斯特菌(listeria monocytogenes, lM)感染对树突状细胞(dendritic cell, DC)免疫功能的影响,探讨细菌型(WT)和l型(l-form)lM对DC的免疫活化能力差异。方法将C57Bl/6小鼠随机分为对照组、WT组、l-form组,3组小鼠分别经尾静脉感染PBS液、细菌型lM和l型lM,电镜观察脾脏DC超微结构特征;流式细胞术检测小鼠脾脏DC的数量、共刺激分子表达、胞内细胞因子分泌、脾脏T细胞亚群及其活化程度。结果透射电镜可观察到对照组小鼠脾脏DC表面有大量丝状伪足,胞浆均匀,细胞核大而偏于一侧;吞噬细菌后,表面丝状伪足减少,胞质内空泡增多;小鼠感染后第1天脾脏中DC绝对值无明显升高或降低(P>0.05),但成熟表型特征性分子表达上调(Pl-form感染组DC表面CD80和CD86分子均高于WT组(PlM后TNF-α^+DC比例明显升高,l-form感染组分泌TNF-α的DC高于WT组(PlM后第7天,3组小鼠脾脏CD4^+T细胞和CD8^+T细胞在淋巴细胞中所占比例差异均无统计学意义(P>0.05),但l-form组CD69^+ T细胞比例高于WT组(Pl型lM可介导相对高水平的TNF-α,促进DC成熟以增强其抗原递呈的能力。

甘蔗杆状病毒(Sugarcane bacilliform viruses,SCBVs)属于花椰菜花叶病毒科Caulimoviridae杆状DNA病毒属Badnavirus,是危害甘蔗的主要病毒之一,在世界各主要甘蔗产区均有发现,严重影响甘蔗的产量和品质。因此,急需建立一种快速、灵敏、特异的检测方法,为阻止SCBV病毒跨区域传播,为该病害隔离检疫和健康脱毒种苗质量监控提供关键技术支撑。本研究针对2012年国际病毒分类委员会ICTV公布的两个SCBV病毒种Sugarcane bacilliform IM virus(SCBIMV,NC03031)和Sugarcane bacilliform MO virus(SCBMOV,NC08017)的反转录酶/RNA 酶 H(RT/RNase H)序列分别设计了一套特异性引物和探针,通过优化反应体系和条件,建立了灵敏度高、特异性强、重现性好的TaqMan实时荧光定量PCR(qPCR)检测方法。该方法以重组质粒PMD18T-IM(SCBIMV)和pSCBV20(SCBMOV)重组质粒DNA为模板(109-10拷贝/u l)进行荧光定量PCR检测,建立标准曲线和回归方程,其扩增效率分别为99.26%和101.73%,最低均能检测100拷贝/μ l,比常规PCR灵敏度高100-1000倍。通过对不同SCBV株系/基因型的荧光定量PCR检测,两套qPCR检测方法均呈现良好的特异性。对采集自福州试验地国家区试甘蔗品(系)114份和云南蔗区甘蔗品种(系)62份叶片样品进行常规PCR和qPCR平行检测,常规PCR共检测到52份阳性样品,检出率为29.5%,其中,福州样品阳性检出率为41.23%,而云南样品阳性检出率为8.06%。利用本文建立的SCBIMV和SCBMOV的qPCR检测方法,在176份叶片样品中,阳性样品检出率分别为50.0%和46.6%。其中,福州、云南样品SCBIMV-qPCR方法阳性检出率分别为71.05%和11.29%,而福州、云南样品SCBMOV-qPCR方法阳性检出率分别为57.01%和27.41%,这些结果显示qPCR方法阳性样品检出率明显高于常规PCR,表明qPCR检测方法具有更高的灵敏度。对52份阳性样品的常规PCR产物进行克隆和测序,共获得80条SCBV病毒RT/RNaseH片段序列,连同已报道的38条代表18个不同SCBV基因型/株系为参考序列,利用邻接法构建系统进化树,结果表明所有供试的SCBV分离物划分成19个亚组群,即SCBV-A至SCBV-S,其中SCBV-S为本研究新发现的基因型/株系。本研究获得的福州SCBV分离物划分在E、F、l、N和Q组群,而云南SCBV分离物划分在Q、R和S组群。综上所述,本研究建立的SCBIMV、SCBMOV荧光定量PCR方法为SCBV的检验检疫和病害防控提供准确可靠、灵敏、快速的检测技术。发现福州试验地国家区试甘蔗品(系)和云南蔗区甘蔗品(系)存在多种SCBV株系或基因型,加强我国SCBV病毒的隔离检疫和病害流行监测势在必行。

Argemone mexicana extract alleviates gastrointestinal disorders by stimulating muscarinic receptors and blocking voltage-gated l-type calcium channels
