AIM： To investigate genetic differences between Crohn＇s disease （CD） patients with a sustained remission vs relapsers after discontinuing infliximab while in cortico- steroid-free remission. METHODS： Forty-eight CD patients received infliximab and were in full corticosteroid-free clinical remission but then discontinued infliximab for reasons other than a loss of response, were identified by review of an electronic database and charts. Infliximab-associated remis- sion was defined as corticosteroid-free plus normaliza- tion of clinical disease activity [CD activity index （CDAI）〈 150] during follow-up visits based on physician global assessments. A CD relapse （loss of infliximab-induced remission） was clinically defined as a physician visit for symptoms of disease activity （CDAI 〉 220） and a thera- peutic intervention with CD medication（s）, or a hospital- ization with complications related to active CD. Genetic analyses were performed on samples from 14 patients （n = 6 who had a sustained long term remission after stopping infliximab, n -- 8 who rapidly relapsed after stopping infliximab）. Nucleotide-binding oligomerization domain 2 （NOD2）/caspase activation recruitment do- main 15 （CARD15） polymorphisms （R702W, G908R and l1007fs） and the inflammatory bowel disease 5 （IBDS） polymorphisms （IGR2060a1 and IGR3081a1） were ana- lyzed in each group. RESUlTS： Five single nucleotide polymorphisms of IBD5 and NOD2/CARD15 genes were successfully analyzed for all 14 subjects. There was no signifcant increase in frequency of the NOD2/CARD15 polymor- phisms （R702W, G908R and l1007fs） and the IBD5 polymorphisms （IGR2060al and IGR3081a1） in either group of patients; those whose disease relapsed rap- idly or those who remained in sustained long term remission following the discontinuation of infliximab. Institutional performance and participatory paradigms: Comparing two groups of watersheds in semi-arid region of India

Watershed development programmes carried out in different agroclimatic conditions in India resulted in beneficial impact in terms of productivity enhancement and natural resources conservation, but less attention paid to institutional and participatory aspects. This paper explored the performance of various institutions regarding execution of watershed development programmes in semi-arid region of India. Recorded observations from documents maintained at watershed level and information collected through primary survey as well as focus group discussion with different types of stakeholders were used for analysis. The results indicated lacunae in participatory aspects during programme implementation process like monitoring activities, management of common property resources and equity. Gap in linkages and differential level of performance of various watershed level functionaries indicates the necessity for corrections in the structures and linkages pattern for sustainability of the infrastructure and institutions. The study also showed unequal priorities by the implementing agencies towards in-stitutions, land and water resources development, production enhancement activities and fodder re-sources development as well as rationalities of technical, economic, financial, political and social aspects among the watersheds.

Clonal Fidelity of Micropropagated Psidium guajava l. Plants Using Microsatellite Markers

Micropropagation of Psidium guajava l. (guava) is a viable alternative to currently adopted techniques for large-scale plant propagation of commercial cultivars. Assessment of clonal fidelity in micropropagated plants is the first step towards ensuring genetic uniformity in mass production of planting material. In the present study, 31 plants of guava cultivar “lucknow 49” regenerated by micropropagation were tested for genetic fidelity by comparing them to the mother plant from which explant material was obtained. Efficient rooting of in vitro proliferated shoots was obtained by culture on 1/2 strength MS medium supplemented with either 9.8 μM indole butyric acid (IBA) or 11.4 μM indole acetic acid (IAA). leaf samples of 31 regenerated plants were compared to the mother plant using 17 simple sequence repeat (SSR) markers. While 16 SSRs detected the same allele, locus mPgCIR07 detected slight differences, where six micropropagated plants were 1 bp smaller (152 bp) than the parental genotype (153 bp). Differences in leaf tissues for anthocyanin pigmentation were also noted among micropropagated plants. Results of the study indicated efficient rooting of “lucknow-49” cultivar for rapid propagation of planting material, and revealed that micropropagated plants were identical for 16 of the 17 loci examined. Although most mutations induced by tissue culture may not have an effect on phenotype, the possibility that novel phenotypes can be generated in a commercial setting exists.

Background:Persistent patent ductus arteriosus(pPDA)is a common condition in preterm *** metaanalysis aimed to assess the safety and efficacy of transcatheter closure(TC)when compared to surgical ligation(Sl)in preterm infants with ***:A literature search of Ovid Cochrane library,Medline,Embase,Epub,Scopus,PMC Preprints,and *** was conducted from inception to May 06,*** studies reported infants diagnosed with pPDA born at≤2000 g birth weight or at≤37 weeks’who underwent TC or Sl as *** review was registered in PROSPERO(CRD42022325944).Results:From 97 studies screened,8 studies met the eligibility criteria,with a total of 756 preterm infants undergoing either TC(n=366)or Sl(n=390).Compared to TC,Sl had higher mortality rates(OR=0.32,95%CI:0.16,0.66,I^(2)=0%).No difference was seen in post-procedural complication rate(OR=0.90,95%CI:0.18,4.44,I^(2)=79%),mean duration of post-procedural mechanical ventilation(MD=−2.21 days,95%CI:−4.88,0.47,I^(2)=60%),hospital stay length(MD=−8.30 days,95%CI:−17.03,0.44,I^(2)=0%)or neonatal intensive care unit stay length(MD=−3.50 days,95%CI:−10.27,3.27,I^(2)=0%).Conclusion:Our meta-analysis demonstrated TC as a viable alternative option in managing preterm infants with pPDA in the context of *** the promising trends demonstrated in this meta-analysis,further studies with larger sample size and controlled baseline characteristics are needed to evaluate the safety and efficacy of TC and Sl for preterm infants with pPDA.

Background:Anthropogenic disturbance can negatively affect an animal's energy budget by evoking movement *** research focuses mainly on immediate displacement as a disturbance effect,since this can be easily observed in the field. Methods:We combined GPS tracking data with observational data to quantify the effects of anthropogenic (air force and walkers) and non-anthropogenic disturbances on distances travelled by roosting Eurasian Oystercatchers (Haematopus ostralegus) during the non-breeding season. We compared immediate displacement after a disturbance with distance travelled during the entire high tide period (longer-term response), while accounting for environmental ***,we calculated energy expenditure due to disturbance based on observed disturbance ***:Disturbance resulted in an immediate displacement response of ~200 m (median).Air force disturbances tended to yield larger immediate responses than walker and,especially,than non-anthropogenic ***-term responses and immediate responses were approximately similar,suggesting that,over longer timescales,spatial disturbance effects in the study area remain confined to immediate ***,disturbances were infrequent (0.17 disturbances per bird per hour) and most disturbances were of natural origin (62%).Consequently, anthropogenic disturbance of roosting oystercatchers in the study area on average costs 0.08% of the daily energy expenditure. Phosphoribulokinase （PRK）, a nuclear-encoded plastid-localized enzyme unique to the photosynthetic carbon reduction （Calvin） cycle, was cloned and characterized from the stramenopile alga Vaucheria litorea. This alga is the source of plastids for the mollusc （sea slug） Elysia chlorotica which enable the animal to survive for months solely by photoautotrophic CO2 fixation. The 1633-bp V. litorea prk gene was cloned and the coding region, found to be interrupted by four introns, encodes a 405-amino acid protein. This protein contains the typical bipartite target sequence expected of nuclearencoded proteins that are directed to complex （i.e. four membrane-bound） algal plastids. De novo synthesis of PRK and enzyme activity were detected in E. chlorotica in spite of having been starved of V. litorea for several months. Unlike the algal enzyme, PRK in the sea slug did not exhibit redox regulation. Two copies of partial PRK-encoding genes were isolated from both sea slug and aposymbiotic sea slug egg DNA using PCR. Each copy contains the nucleotide region spanning exon 1 and part of exon 2 of V litorea prk, including the bipartite targeting peptide. However, the larger prk fragment also includes intron 1. The exon and intron sequences of prk in E. chlorotica and V/itorea are nearly identical. These data suggest that PRK is differentially regulated in V. litorea and E. chlorotica and at least a portion of the V. litorea nuclear PRK gene is present in sea slugs that have been starved for several months.

Reprogramming mouse ear mesenchymal stem cells (EMSC) expressing the Dlk1-Dio3 imprinted gene cluster

The identification of a single, early marker for full developmental potential of induced pluripotent stem (iPS) cells has proven elusive. Recently, however, activation of the imprinted gene cluster, Dlk1-Dio3 has emerged as a viable candidate in the mouse. To explore the relationship between Dlk1-Dio3 expression and developmental potential more fully, we used murine ear mesenchymal stem cells (mEMSC) for iPS cell induction. Mouse EMSC are easily obtained and share functional characteristics with embryonic stem (ES) cells and therefore, may be a reliable non-embryonic source for iPS cell production. We report that mEMSC express high levels of Gtl2, a maternally expressed gene within the Dlk1-Dio3 imprinted cluster. Moreover, mEMSC produce Gtl2 expressing (Gtl2on) iPSC clones that share functional characteristics with ES cell clones. The production of Gtl2on iPS cell clones from mEMSC provides a new model with which to investigate the regulation of Dlk1-Dio3 cluster activity during direct cell reprogramming.

针对基于光电非接触式及图像处理式的断纱检测方式对车间生产环境、安装位置、安装角度要求高且设备昂贵等问题,根据静电感应式检测基本原理,设计了静电荷感应式断纱传感器,对其总体结构及关键模块进行理论分析及实验测试,并搭建了静电荷感应式断纱传感器试验台对该传感器的功能进行测试。结果表明:虽然运动纱线速度及其材料是影响静电荷感应式断纱传感器的主要影响因素,但这两个因素所导致的电压波动在静电荷感应式断纱传感器关键模块后端输出点的数值远小于运动纱线在断裂和未断裂状态之间切换时所产生的电压波动。因此,检测静电荷感应式断纱传感器关键模块后端输出点的电压值,可实现对运动纱线断纱情况的准确判断。
