Human papillomavirus in esophageal squamous cell carcinoma in Colombia and Chile

AIM: To examine the presence of human papillomavirus (HPV) in esophageal squamous cell carcinoma (ESCC) specimens collected from Colombia and Chile located in the northern and southern ends of the continent, respectively. METHODS: We examined 47 and 26 formalin-fixed and paraffin-embedded ESCC specimens from Colombia and Chile, respectively. HPV was detected using GP5+/GP6+ primer pair for PCR, and confirmed by Southern blot analysis. Sequencing analysis of L1 region fragment was used to identify HPV genotype. In addition, P16INK4A protein immunostaining of all the specimens was conducted. RESULTS: HPV was detected in 21 ESCC specimens (29%). Sequencing analysis of L1 region fragment identified HPV-16 genome in 6 Colombian cases(13%) and in 5 Chilean cases (19%). HPV-18 was detected in 10 cases (21%) in Colombia but not in any Chilean case. Since Chilean ESCC cases had a higher prevalence of HPV-16 (without statistical significance), but a significantly lower prevalence of HPV-18 than in Colombian cases (P = 0.011) even though the two countries have similar ESCC incidence rates, the frequency of HPV-related ESCC may not be strongly affected by risk factors affecting the incidence of ESCC. HPV-16 genome was more frequently detected in p16 positive carcinomas, although the difference was not statistically significant. HPV-18 detection rate did not show any association with p16 expression. Well-differentiated tumors tended to have either HPV-16 or HPV-18 but the association was not statistically significant. HPV genotypes other than HPV-16 or 18 were not detected in either country. CONCLUSION: HPV-16 and HPV-18 genotypes can be found in ESCC specimens collected from two South American countries. Further studies on the relationship between HPV-16 presence and p16 expression in ESCC would aid understanding of the mechanism underlying the presence of HPV in ESCC.

Recombinant streptokinase vs phenylephrine-based suppositories in acute hemorrhoids, randomized, controlled trial(THERESA-3)

AIM: To compare the efficacy and safety of recombinant streptokinase(rSK) and phenylephrine-based suppositories in acute hemorrhoidal ***: A multicenter(14 sites), randomized(1:1), open, parallel groups, active controlled trial was done. After inclusion, subjects with acute symptoms of hemorrhoids, who gave their written, informed consent to participate, were centrally randomized to receive, as outpatients, rSK(200000 IU) or 0.25% phenylephrine suppositories, which had different organoleptic characteristics. Treatment was administered by the rectal route, one unit every 6 h during 48 h for rSK, and up to a maximum of 5 d(20 suppositories) for phenylephrine. Evaluations were performed at 3, 5 and 10 d postinclusion. The main end-point was the 5th-day complete clinical response(disappearance of pain and edema, and ≥ 70% reduction of the lesion size). Time to response and need for thrombectomy were secondary efficacy variables. Adverse events were evaluated too. RESULTS: 5thday complete response rates were 83/110(75.5%) and 36/110(32.7%) with rSK and phenylephrine suppositories, respectively. This 42.7% difference(95%CI: 30.5-54.2) was highly significant(P ely; P ely; P eline thrombosis in the rSK and phenylephrine groups, respectively(P = 0.016). There were no adverse events attributable to the experimental treatment. CONCLUSION: rSK suppositories showed a significant advantage over a widely used over-the-counter phen-ylephrine preparation for the treatment

While the cervical mucosal immune response to human papillomavirus (HPV) infection is believed to be central to viral clearance, it is not well characterized. We performed this analysis to determine correlates of HPV- 16-specific mucosal antibody response in women at high risk for infection with HPV. Methods. Cervical mucosal and serum samples were obtained from participants in a case control study that measured demographic risk factors of cervical disease and HPV infection. An HPV- 16 L1-virus-like particle elISA was used to detect HPV- 16-specific IgA and IgG. Antibody level results were correlated with demographic characteristics, sexual history, cervical disease, and HPV detection. Results. Cervical anti- HPV- 16 IgA and IgG inversely correlated with HPV DNA, HPV- 16 DNA, and cervical disease. Conclusions. Few studies provide serologic and mucosal antibody results in HPV- positive patients. There is a need for more longitudinal studies of HPV infection with verification of HPV positivity in tissue, measures of sera-positivity, measure-ments of mucosal immunity, and measurements of the cellular immune response.

Impacts of Date Palm Seeds （Phoenix dactyliferous L.） on Growth Indices and Nutrient Utilization of Common Carp Cyprinus carpio L,

This study was carried out to study the effect of adding different levels of date palm seed (Phoentix dactyliferous) powder in the diet of common carp (Cyprinus carpio) at fish laboratory of Animal Production Department, College of Agricultural Sciences, Sulaimani University, Sulaimani, Iraq. The actual experimental feeding trials were conducted for 70 d and the adaptation period for 21 d. A total of 81 common carp fingerlings weighted 62 ± 2 g were divided into three experimental groups. The control treatment T1 was with 0 g date palm seed, T2 with 2.5 g date palm seed per kg diet and T3 with 5 g date palm seed per kg diet. Each treatment was in three replicates in which three common carp fingerlings were stocked in each aquarium which fed the experimental diets twice daily. The results indicated that weight gain (WG) and specific growth rate (SGR) in the third treatment (T3) group were significantly higher than those of fish fed control diets. T2 consumed more diet than other treatments with 306.940 g with higher feed conversion ratio (FCR); T3 was higher significantly in food efficiency. No significant (P < 0.05) differences obtained from consumed protein while the protein efficiency ratio was significantly higher in T3 with 319.323. Control was higher significantly in flesh weight without viscera with the third treatment; in flesh weight without head and viscera each the control and the second treatment was significantly higher than the third treatment. As a conclusion, the date palm seeds powder have a positive effect on growth performance of common carp.

Particle Encapsulation in Crosslinked Hydrogel Networks： Particle Distribution Optimization

Particle encapsulation in hydrogel membranes was performed to examine which physical factors are involved in particle distribution within membranes. This investigation focused on the examination of the physicochemical interactions between particles and crosslinked hydrogel networks for the creation of homogenously dispersed particles in a membrane. For this purpose factors such as particle charge, concentration, and membrane charge were examined to ascertain their effect on particle dispersion within the matrix. Anionic hydrogels composed of methacrylic acid (MAA), cationic hydrogels composed of N, N-dimethyl amino ethyl methacrylate (DMAEM), and neutral hydrogels composed of 2-hydroxyethyl methacrylate (HEMA) were utilized to encapsulate functionalized silica particles. Particle distribution was analyzed by Scanning electron Microscopy and Near Infrared Chemical Imaging (NIR-CI). A new method was developed to evaluate the distribution of particles throughout the membranes. NIR-CI results indicated that neutral membranes with low particle concentration showed better particle distribution.

All participating patients gave their written,informed consent. After inclusion, patients with acute symptoms of hemorrhoids were centrally randomized to receive, as outpatients, by the rectal route, suppositories of rSK 200000 IU of one unit every 8 h(first 3 units)and afterwards every 12 h until 8 administrations were completed(schedule A), one unit every 8 h until 6 units were completed(schedule B), or 25 mg hydrocortisone acetate once every 8 h up to a maximum of 24 administrations. Evaluations were performed at 3, 5,and 10 d post-inclusion. The main end-point was the 5thday response(disappearance of pain and bleeding, and≥ 70% reduction of the lesion size). Time to response and need for thrombectomy were secondary efficacy variables. Adverse events were also ***: Groups were homogeneous with regards to demographic and baseline characteristics. Fifth day complete response rates were 156/170(91.8%; 95%CI:87.3-96.2), 155/170(91.2%; 95%CI: 86.6%-95.7%),and 46/170(27.1%; 95%CI: 20.1%-34.0%) with rSK(schedule A and B) and hydrocortisone acetate suppositories, respectively. These 64.6% and 63.9%differences(95%CI: 56.7%-72.2% and 55.7%-72.0%)were highly significant(P ely; P ely;P < 0.001).
