AIM： To study the effect of H2 gas on liver injury in massive hepatectomy using the Intermittent Pringle maneuver in sw.ne. METHODS： Male Bama pigs （n = 14） treated w.th ketamine hydrochloride and Sumianxin Ⅱ as induc- tion drugs follow.d by inhalation anesthesia w.th 2% isoflurane, underw.nt 70% hepatotectomy w.th loss of bleeding less than 50 mL, and w.th hepatic pedicle occlusion for 20 min, w.re divided into tw. groups： Hydrogen-group （n = 7）, the pigs w.th inhalation of 2% hydrogen by the tracheal intubation during major hepa- totectomy; Contrast-group （n= 7）, underw.nt 70% hepatotectomy w.thout inhalation of hydrogen. Hemo- dynamic changes and plasma concentrations of alanine aminotransferase （ALT）, aspartate aminotransferase （AST）, hyaluronic acid （HA）, tumor necrosis factor-α （TNF-α）, interleukin-6 （IL-6）, and malondialdehyde （MDA） in liver tissue w.re measured at pre-operation, post-hepatotectomy （PH） 1 h and 3 h. The apoptosis and proliferating cell nuclear antigen （PCNA） expres- sion in liver remnant w.re evaluated at PH 3 h. Then w. compared the tw. groups by these marks to evalu- ate the effect of the hydrogen in the liver injury during major hepatotectomy w.th the Pringle Maneuver in the sw.ne. RESULTS： There w.re no significant differences in body w.ight, blood loss and removal liver w.ight be- tw.en the tw. groups. There w.s no significant differ- ence in changes of portal vein pressure betw.en tw. groups at pre-operation, PH 30 min, but in hydrogen gas treated-group it slightly decrease and low.r than its in Contrast-group at PH 3 h, although there w.re no significant difference （P = 0.655）. ALT and AST in Hydrogen-group w.s significantly low.r comparing to Contrast-group （P = 0.036, P = 0.011, vs P = 0.032, P = 0.013） at PH 1 h and 3 h, although the tw. groups all increased. Atorvastatin, a lipid-low.ring medication, provides neuroprotective effects, although the precise mechanisms of action remain unclear. Our previous studies confirmed activated autophagy follow.ng spinal cord injury, w.ich w.s conducive to recovery of neurological functions. w. hypothesized that atorvastatin could also activate autophagy after spinal cord injury, and subsequently improve recovery of neurological functions. A rat model of spinal cord injury w.s established based on the Allen method. Atorvastatin（5 mg/kg） w.s intraperitoneally injected at 1 and 2 days after spinal cord injury. At 7 days post-injury, w.stern blot assay, reverse transcription-polymerase chain reaction, and terminal deoxynucleotidyl transferase-mediated dU TP nick-end labeling（TUNEL） staining results show.d increased Beclin-1 and light chain 3B gene and protein expressions in the spinal cord injury ＋ atorvastatin group. Additionally, caspase-9 and caspase-3 expression w.s decreased, and the number of TUNEL-positive cells w.s reduced. Compared w.th the spinal cord injury ＋ saline group, Basso, Beattie, and Bresnahan locomotor rating scale scores significantly increased in the spinal cord injury ＋ atorvastatin group at 14–42 days post-injury. These findings suggest that atorvastatin activated autophagy after spinal cord injury, inhibited apoptosis, and promoted recovery of neurological function.

Aim:To investigate the effect of peroxisome proliferator-activated receptor gamma(PPARγ)activation on the grow.h of rat hepatic stellate cells(HSC)and trans-forming grow.h factor beta(TGF-β)-induced connective tissue grow.h factor(CTGF)***:After being treated w.th various amounts of thePPARγ natural ligand 15-deoxy-A 12,14-prostaglandin J2(15-d-PGJ2)or syntheticligand Gw.845,the status of HSC proliferation and apoptosis w.re detected byMTT assay and flow.***,HSC w.re treated w.th PPARγ-specific antagonist Gw.662 prior to the addition of 15-d-PGJ2or Gw.845 andw.re subsequently stimulated w.th TGF-β*** mRNA and protein levels ofCTGF expression w.re detected by semi-quantitative RT-PCR and w.stern blot-ting *** changes in the HSC w.re observed by ***:15-d-PGJ2and Gw.845 markedly inhibited HSC prolifera-tion and induced cell apoptosis in a dose-dependent ***,PPARγligands significantly suppressed TGF-β1-induced CTGF expression(at both tran-scriptional and post-transcriptional levels)in HSC,and the inhibitory effect w.sdramatically,if not completely,abolished by pretreatment w.th Gw.662,sug-gesting that the inhibition w.s indeed mediated by PPARγ.Moreover,morpho-logical observation revealed that PPARγ activation caused obvious changes ofHSC from activated to quiescent ***:The PPARγligand hasa potent inhibitory effect on the grow.h of HSC and TGF-β1-induced CTGFexpression,w.ich makes it a potential antifibrotic candidate for the treatment andprevention of hepatic fibrosis.

To the Editor:Liver cancer is the fifth most common cancer and the second most frequent cause of cancer-related death *** carcinoma(HCC)accounts for 90%of primary liver cancers w.th the highest incidence in China(more than 50%of all cases w.rldw.de)[1].Liver transplantation(LT)is regarded as an optimal therapy for selected HCC *** Milan criteria are the benchmark for candidate selection that ensure excellent prognosis for patients w.th HCC[2].The Hangzhou criteria expand 51.5%more of Milan criteria for LT candidates w.th comparable posttransplant survivals[3].How.ver,LT recipients fulfilling Milan criteria or Hangzhou criteria are at the risk of up to 13%−18%HCC recurrence rate w.thin five years[4].Only 25%−50%of recurrent HCC patients post-LT are eligible for surgical treatment w.ich have consistently presented favored survival benefit than systemic therapy[5].

The Relationship between Personality Traits and Face Shapes ...

The Ubiquitin E3 Ligase PUB17 Positively Regulates Immunity by Targeting a Negative Regulator, KH17, for Degradation

Objective: Physiognomy has over 3000 years of history in China, w.ere the belief that personality can be discerned through physiognomy is w.despread. How.ver, it hasn’t been fully verified by scientific research. Through experiments, this paper explores the relationship betw.en face shape and corresponding personality in physiognomy, and how.face shape affects people’s judgment of personality. Method: According to the eight face shapes theory of physiognomy, 10 trained laboratory assistants have selected 64 typical faces through 3816 pieces of ID photos follow.ng a designated procedure, and tested the selected 64 persons’ scores of Cattell’s 16 Personality Factors Test. Eight more ID photos have been randomly selected, and each one has been modified by Image Processing Technology into eight face shapes, keeping other facial features same to ensure that the only variable is face shape, and ultimately obtained 64 artificial faces. 949 undergraduates, as participants, have visually judged these 128 faces in a laboratory by using E-prime 2.0 and 16 PF Rating Scale. Results: Overall, there w.s no significant difference of tested sixteen personality traits among eight typical faces. Through a post-hoc test, some face shapes are perceived to have certain significant differences in some personality traits than a certain face shape. For example, on factor Q2 of 16 PF, a heart-shaped face（M = 2.625*） is significantly low.r than a diamond-shaped face（M = 4.375）. In contrast, there are various differences among the eight face shapes on people’s visual judgmental of personality traits. For example, the heart-shaped face（M = 4.01**） is significantly low.r than all other face shapes on factor A). By comprising the tested personality traits and perceived personality traits of each face shape, there are significant differences among some personality traits（e.g. diamond face on factor B, t =-2.847**）. Coronavirus disease 2019(COVID-19)has impacted almost every part of human lifew.rldw.de,posing amassive threat to human *** lack of time for new.drug discovery and the urgent need for rapid disease control to reduce mortality have led to a search for quick and effective alternatives to novel therapeutics,for example drug *** identify potentially repurposable drugs,w. employed a systematic approach to mine candidates from ***-approved drugs and preclinical small-molecule compounds by integrating gene expression perturbation data for chemicals from the Library of Integrated Netw.rk-Based Cellular Signatures project w.th a publicly available single-cell RNA sequencing dataset from patients w.thmild and severe COVID-19(GEO:GSE145926,public data available and accessed on 22 April 2020).w. identified 281 FDA-approved drugs that have the potential to be effective against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection,16 of w.ich are currently undergoing clinical trials to evaluate their efficacy against *** experimentally tested and demonstrated the inhibitory effects of tyrphostin-AG-1478 and brefeldin-a,tw. chemical inhibitors of glycosylation(a post-translational modification)on the replication of the single-stranded ribonucleic acid(ssRNA)virus influenza A virus as w.ll as on the transcription and translation of host cell cytokines and their regulators(IFNs and ISGs).In conclusion,w. have identified and experimentally validated repurposable anti-SARS-CoV-2 and IAV drugs using a systems biology approach,w.ich may have the potential for treating these viral infections and their complications(sepsis).

The expression of Selenoprotein w.Selw.in C2C12 skeletal muscle cells w.s specifically decreased to examine its influence on the amount of glutathione(GSH)and the activity of glutathione peroxidase(GPx).Selw.knock-dow. w.s performed by RNA interference(RNAi)in cultured muscle cells and verified by Real-time PCR and w.stern *** addition,cell viability,GSH content and GPx activity w.re *** results show.d that the mRNA level and protein expression of Selw.w.re decreased successfully by 71.9%and 68.8%relative to control values,cell viability decreased by 21.5%,GSH increased by 29.76%,and GPx increased by 47.58%.w.T assay show.d that compared w.th blank control,the value of positive group dropped 21.5%;In GSH and GPx assay,compared w.th blank control the positive group increased29.76%and 47.58%*** conclusion,Selw.knock-dow. by RNAi caused significant cytotoxity in skeletal muscle cells and led to compensatory increases in GSH content and GPx *** findings are consistent w.th the suggestions from bioinformatics indicating an antioxidative role for Selw.in skeletal muscle cells.
