Rapid LC-ESI-MS-MS Method for the Simultaneous Determination of Sitagliptin and Pioglitazone in Rat Plasma and Its Application to Pharmacokinetic Study

A liquid chromatography tandem mass spectrometry (LC-MS/MS) based method was developed for the simultaneous monitoring plasma levels of Sitagliptin (STG) and Pioglitazone (PIO) for applicability to pharmacokinetic studies. The method was based on HPLC separation on the reversed phase Phenomenex Synergy C18 column (30 mm length, 4.6 mm internal diameter, and 4.0 μm particle size) at a temperature of 40?C using a binary gradient mobile phase consisting of methanol and 2 mM ammonium acetate buffer pH adjusted to 4.5 with acetic acid, at a flow rate of 1 mL?min?1. Tolbutamide was used as an internal standard. Detection of analytes was achieved with LC-MS/MS system in Multiple Reaction Monitoring (MRM) mode. The method was validated over concentration range of 10.98 - 2091.77 ng?mL?1 for SIT and 8.25 - 1571.63 ng?mL?1 for PIO and lower limit of quantification was 10.98 ng?mL?1 and 8.25 ng?mL?1 for STG and PIO respectively. Recoveries from spiked controls were within acceptance criteria for all the analytes and internal standard at all QC levels. Within batch and between batch accuracy for STG was found within 96.9% - 100.3% and for PIO was found within 100.0% - 104.3%. Within batch and between batch precision for STG was less than 3.1% CV (coefficient of variation) and for PIO was less than 5.3% CV at all concentrations levels. This method was successfully applied to monitor pharmacokinetics profile of both STG and PIO on simultaneous oral administration to rats. This method can be applicable for pharmacokinetic drug-drug interaction studies.

在我们如今的生活当中,临时建筑已屡见不鲜除却通俗意义上的工地简易临时建筑以外,人们还常常能在商场、展会或大型公共活动等场景中见到具有观赏或实用价值的限时建筑、快闪空间、临时景观装置等。这些临时或限时建筑往往依据特定主题或功能应运而生,然而在不久之后就会面临被拆除的命运。看似“昙花一现”的临时建筑生命周期短暂,并会带来不少废弃建材,因而如何以新的建造思考方式来达成可持续的发展,成为了值得大家关注的议题。本期建筑栏目收录的两个临时建筑项目,功能截然不同,但却相当一致地指向可持续再生的理念,Samira Rathod Design Atelier为2023科钦一穆吉里斯双年展打造的“容器”,采用当地材料及工艺,以低成本的预算为人们提供一处公共空间,在承载可持续发展理念的同时,也展现出建筑的诗意;裸筑更新建筑设计事务所的“撇搽之间,穿斗陌阡”则是为城市购物空间打造的周年庆限时建筑,设计师将建筑以汉字会形,而其竹结构在拆除之后,还能作为户外竹凳被再次使用临时建筑不应只呈现短暂之美,它作为城市景观或公共空间的一部分,更应助力城市可持续发展,激发人们的互动参与.

Complication rates following percutaneous interventions have decreased over the past decade due to advancement in both interventional equipment and procedure technique. Despite these advances, the risk of iatrogenic perforations still exists with associated high morbidity and mortality. We are presenting three cases of guidewire-induced perforations including coronary artery and renal artery perforation with their complications and successful management in these case-series.

Pulmonary Resection for Metachronous Lung Cancer Following Contralateral Pneumonectomy

Patients who undergo pneumonectomy for lung cancer are at risk of recurrent disease and metachronous tumours in the remaining lung. Establishing a diagnosis can be difficult as biopsy may induce a pneumothorax, whilst resection is limited by remaining lung function. However, pneumonectomy should not be a contraindication to further lung resection. We report two cases of lung cancer resection in the residual lung 6 and 11 years following pneumonectomy respectively. A limited resection can be safely performed for a new pulmonary parenchymal lesion on a background of a previous pneumonectomy.

Simple extraction and membrane purification process in isolation of steviosides with improved organoleptic activity

In recent years there has been increasing demand for natural non-nutritive high intensity sweeteners with low-calorie value as an alternative to sucrose. Extracts of the leaves of Stevia rebaudiana (Bertoni), have been known for their sweet taste. Steviosides and rebaudioside-A are the two major diterpenoid glucosides components present in the leaf extracts of the stevia, these glycosides are 300 times sweeter than sugar and also exhibits wide therapeutic activity. The conventional methods of isolation of steviosides involve long extraction and purification procedures;therefore optimization of product yields is a challenging problem. The present study, establishes a new improvised process of extraction of steviosides from the stevia leaves in which the dry treated leaves were grounded, defatted, and extracted through pressurized hot water extractor (PHWE), followed by purification and concentration of the sweet glycosides through ultra (UF) and nano (NF) membrane filtration in obtaining high (98.2%) purity steviosides. This process established “green” method for isolation of high quality steviol glycosides, with improved final yield is 10.1% from 11% of crude leaf extract and observed the improved organoleptic and biological activity (antioxidant). Thus the method confirms a simple, inexpensive and eco-friendly process in obtaining pure steviosides.

The Effect of Torsion Deformity and Medial knee Osteoarthritis on Lower Limb Extensor Moments during Gait

The aim of this study was to quantify the effect of torsion deformity on the lower limb kinetics during the loading response phase of gait. A total of 24 subjects: 6 end-staged medial knee OA with torsion deformity (TkO), 8 without torsion deformity (kOA), and 10 controls (CON) were imaged using computed tomography (CT). Internal moment of support and sagittal hip, knee and ankle joint moments were assessed using gait analysis. TkO showed greater external rotations of the proximal tibia and the distal femur compared to subjects with medial knee OA without torsion deformity and controls. TkO showed greater moment of support and a greater knee extensor moment when compared to controls when supporting the weight of the body during gait. The TkO intorsion deformity occurred as a result of a proximal malrotation of the tibia. In the presence of torsion deformity, the kinetic synergy of the lower limb showed increased total moment of support for subjects with medial knee OA. The greater extensor output from TkO may be the result of an increased muscular response to overcome an interrupted inter-segmental exchange of accelerations during the loading response phase of gait.

Molecular Mapping of QTLs for Drought Related Traits at Seedling Stage under PEG Induced Stress Conditions in Rice

Differential response of seedling characteristics under water stress conditions is known to be associated with drought resistance in rice and elucidation of its genetics could be of help in breeding for tolerance to the stress. A recombinant inbred population derived from the cross between a semi-dwarf variety IR64 and landrace INRC10192 was grown in hydroponic culture and phenotyped for varied responses of seedlings to water deficit imposed by poly ethylene glycol (PEG). The ratio between mean value of seedling trait under stress and control conditions was used for assessing drought tolerance. In all 19 putative QTL relating to five seedling traits viz., shoot length, maximum root length, shoot dry weight, root dry weight and root to shoot dry weight ratio under PEG induced stress conditions were identified confirms that the traditional tall landraces as the one chosen for the study posses hitherto unexploited drought tolerant genes and utilization of them as potential donors in breeding for yield enhancement would be rewarding. They might be useful for improving drought resistance of rice by marker assisted selection/breeding.
