Identification of IL11ra and MELk amplification in gastric cancer by comprehensive genomic profiling of gastric cancer cell lines

AIM To identify common copy number alterations on gastric cancer cell *** Four gastric cancer cell lines(ACP02, ACP03, AGP01 and PG100) underwent chromosomal comparative genome hybridization and array comparative genome hybridization. We also confirmed the results by fluorescence in situ hybridization analysis using the bacterial artificial chromosome clone and quantitative real time PCR *** The amplification of 9p13.3 was detected in all cell lines by both methodologies. An increase in the copy number of 9p13.3 was also confirmed by fluorescence in situ hybridization analysis. Moreover, the interleukin 11 receptor alpha(IL11ra) and maternal embryonic leucine zipper kinase(MELk) genes, which are present in the 9p13.3 amplicon, revealed gains of the MELk gene in all the cell lines studied. Additionally, a gain in the copy number of IL11 ra and MELk was observed in 19.1%(13/68) and 55.9%(38/68) of primary gastric adenocarcinoma samples, respectively. CONCLUSION The characterization of a small gain region at 9p13.3 in gastric cancer cell lines and primary gastric adenocarcinoma samples has revealed MELk as a candidate target gene that is possibly related to the development of gastric cancer.

Acoustic Impact Studies (AIS) of Wind Farms in Uruguay: A Methodology Proposal

The fast growing development of wind power in Uruguay has encouraged research on many issues regarding environmental acoustics, especially those related to wind turbines operation. As every new power generation device of 10 MW or larger has to have an environmental license approval before building it, a methodology for Acoustic Impact Studies (AIS) was needed. This paper presents a methodology proposal to carry out AIS, taking into account the peculiarities of the Uruguayan status. Determining the area where the studies should be done, demands for the base line of sound pressure levels, predicting sound pressure levels during the operation of future wind farm and main lines for the environmental management plan are included in this proposal. Uruguayan current national guidelines to noise pollution levels are also presented.

Measuring Sound Pressure Levels during Thermoacoustic Instabilities in Large Engines: Case Study

This paper attempts to present some registers of sound pressure levels during the operation of large diesel engines (10 MW). During these registers we have found the preparation, occurrence and ending of events of thermoacoustic instability. They appear after a loosing of chaos period or a reduction in fluctuations in some frequencies. The most interesting phenomena were registered at low frequencies. However, they were accompanied by variations in sound emissions at medium and high frequencies. As there has been very little published data concerning these phenomena at real scale, it is imperative to point out that every quasi-stationary state we have measured during these episodes has lasted some minutes, significantly much more time than that of lab scale results.

In Vitro Evaluation of Two Tissue Substitutes for Gingival Augmentation

Three-dimensional collagen matrices of porcine origin are being used as substitutes for soft tissue grafts in periodontal plastic surgery in search of aesthetic and natural results. This in vitro study aimed to compare Fibro-Gide® (GeistlichBiomaterials) and Mucoderm® (BotissBiomaterials) matrices during the initial phase of soft tissue formation. For this purpose, samples of 5 × 5 mm were obtained, and then human fibroblasts were plated on them. After 24, 48 and 72 h, cell viability was assessed using an MTT assay, and the secretion of type I collagen, MMP-2, TIMP-1 and TIMP-2 was analyzed by ELISA immunoassay. The control group (C) consisted of cells plated on polystyrene without the matrices. The morphology of the surfaces was also examined using scanning electron microscopy (SEM), as was the average roughness (ra) of the samples by a profilometer. Topographic analysis revealed that roughness was significantly higher on Mucoderm® than on Fibro-Gide® (p 0.05). The synthesis of type I collagen, MMP-2 and TIMP-1 were significantly higher from cells plated on Fibro-Gide® than on Mucoderm®, in all time points (p ® than on Mucoderm® (p ® induced an increase in type I collagen, MMP-2 and TIMP-1 and TIMP-2.

Mechano-Synthesized Orthoferrite Starting from Wüstite Assisted by SPS

Stoichiometric mixtures of FeO and Y2O3 were milled and heat treated to obtain yttrium iron garnet, Y3Fe5O12. Two types of heating systems were used: one, a spark plasma sintering machine and the second, an electrical oven. The magnetic properties of the resulting specimens have been analyzed and discussed as a function of the grain size and the particles’ morphology. The partial formation of garnet and orthoferrite phases was revealed on the obtained powder through microstructural analyses after 9 h of ball milling. The milled powders were transformed into the orthoferrite phase after the SPS-treatment at 700°C and 900°C. Magnetic-saturation studies revealed magnetic responses up to 12.7 emu/g for specimens SPS-treated at 700°C, whereas 2.1 emu/g for samples SPS-treated at 900°C. Conventionally treated specimens at 700°C developed 0.36 emu/g of magnetization, while 0.93 emu/g was registered for those treated at 900°C.

Y3Fe5O12Prepared by Mechanosynthesis from Different Iron Sources

Yttrium iron garnet, Y3Fe5O12 (YIG) powders were synthesized by mechanochemical processing (MCP) from different iron sources (FeO, Fe2O3 and Fe3O4) mixed with Y2O3, followed by a heat treatment. The aim of this work is to demonstrate that MCP followed by annealing at very low temperatures (as compared with the classic solid state reaction) can induce the formation of nanostructured YIG. The effect of iron source on final structure was also studied. X-ray diffraction (XRD) and scanning electron microscopy (SEM) were used to characterize the synthesized powders. The precursors mixed in a stoichiometric ratio to obtain YIG were milled at room temperature in a shaker mixer mill with a ball:powder weight ratio of 10:1. A partial synthesis of YIG was achieved after 9 h of milling time by using the three sources of iron;however, a significant fraction of the product was the perovskite YFeO3. The largest yield of YIG was obtained by using FeO. In all cases a single garnet phase could only be completely obtained after an annealing process at 900?C, around 400?C lower than the typical temperatures to prepare the material by solid state reaction. An analysis of the microstrain and lattice parameters associated with peak displacements is discussed.

Identification of optimal contemporary antiemetic prophylaxis for doxorubicin-cyclophosphamide chemotherapy in Chinese cancer patients: post-hoc analysis of 3 prospective studies

Reduced Locomotor Activity Correlates with Increased Severity of Arthritis in a Mouse Model of Antibody-Induced Arthritis

Introduction: Rheumatoid arthritis (ra) is a chronic autoimmune disease characterized by synovial hyperplasia and progressive cartilage and bone destruction that leads to a substantial loss of general functions and/or a decline in physical activities such as walking speed in humans. The k/BxN serum transfer arthritis in mice shares many immunological and pathological features with human ra. Very few studies are available in mice that investigate the changes in physical activity in relation to arthritis development. In this study we investigate the effect of arthritis on the locomotor activity of mice during k/BxN sera transfer arthritis. Methods: Arthritis was induced in Balb/c mice by injecting intraperitoneally with 200 μl of k/BxN sera;Balb/c mice injected with phosphate buffered saline (PBS) served as control. Progress of arthritis was estimated by daily measurements of joint thickness. Each mouse’s locomotor activity (travel distance and travel time) was assessed every day for duration of 20 minute period using the SmartCageTM platform. Data were analyzed using the SmartCageTM analysis software (CageScoreTM). Results: Arthritic Balb/c mice showed a reduction in distance covered and travel speed when compared to arthritis-free, control Balb/c mice. Maximum decline in locomotor activity was observed during the peak period of the disease and correlated to the increase in joint thickness in the arthritic mice. Conclusion: This report demonstrates that measuring locomotor activity of mice during progression of k/BxN sera-induced arthritis using the SmartCageTM platform offers a quantitative method to assess physical activity in mice during arthritis.

TNF-αAntagonist and Infection in Rheumatoid Arthritis

Background: Anti-TNF treatment may increase infection risk, although this has been difficult to study because the timing of anti-TNF treatment is driven by disease activity, which may influence infection susceptibility leading to confounding that varies over time. We evaluated the association between anti-TNF initiation in rheumatoid arthritis (ra) patients on disease modifying anti-rheumatic drugs (DMARD) and infection using multiple approaches adjusting for time-varying confounding. Methods: 383 anti-TNF-na?ve ra patients on ≥ 1 non-biologic-DMARD at enrollment from the Brigham and Women’s Rheumatoid Arthritis Sequential Study (BraSS) were followed up to two years. Pooled logistic regressions estimated the association between anti-TNF and infection by including time-varying covariates in the adjusted models and inverse probability treatment weighting (IPTW). Results: Adjustment for time-varying disease activity and other suspected confounders yielded non-statistically significant positive associations between anti-TNF start and infection regardless of analytic approach (RRmvar_adj = 2.1, 95% CI: 0.8 - 5.8). Conclusions: Incorporating changing clinical status, and treatment indications and consequences, yielded consistently (though not significantly) elevated relative risks of infection associated with anti-TNF initiation. Due to limited statistical power, we cannot draw firm conclusions. However, we have illustrated multiple approaches adjusting for potential time-varying confounding in longitudinal studies and hope to replicate the approaches in larger studies.

[背景]颗粒物(PM)与人类健康之间的关联随着季节变化而不同,这种现象可能与颗粒物的成分以及毒性相关的季节性交互作用有关。[目的]检测PM成分的季节性变化,并使用多个PM样本评估PM的体外促炎症可能性。[方法]以5个污染源不同的城市地区为采集地点,每周采集样本,在多雨温暖季节和干燥寒冷季节累积采集90份PM_(10)和PM_(2.5)样本。对其中检测到的元素、多环芳烃(PAHs)和内毒素进行主成分分析(PCA)。在人单核细胞(THP-1)中检测PM诱导肿瘤坏死因子α(TNFα)和白介素6(IL-6)分泌的可能性,并用组分评分建立促炎症反应的模型。[结果]PM的成分随其大小和季节而变化。PCA确定了PM中两种随季节变化的主要组分。燃烧相关的成分(如钒、苯并(a)芘、苯并(a)蒽)组成组分1(C_1),土壤相关的成分(如内毒素、硅、铝)组成组分2(C_2)。多雨温暖季节采集的PM中C_2含量较高,而干燥寒冷季节采集的PM(尤其是PM_(2.5))中C_1含量较高。细胞因子生成水平与PM_(10)和C_2(多雨温暖季节)正相关,但与PM_(2.5)和C_1(干燥寒冷季节)负相关。C_2含量较高的PM暴露水平越高,TNFα分泌越多;样本含有≥0.1%的C_1相关PAHs时,无论样本中C_2含量高低,暴露于该样本后TNFα对PM的反应性分泌都会降低。IL-6检测结果提示PM成分和颗粒大小之间存在复杂的交互作用。[结论]TNFα分泌具有季节性并与PM大小相关,PM中的土壤含量及PAH含量的变化是其根本原因。这些结果提示,流行病学研究中一些健康结局与PM之间关联的季节性差异能用PM组分的混合物来解释。
