Cytochrome P450 BM3 of Bacillus megaterium-A possible endosulfan biotransforming gene

Computing chemistry was applied to understand biotransforrnation mechanism of an organochlorine pesticide, endosulfan. The stereo specific metabolic activity of human CYP-2B6 （cytochrome P450） on endosulfan has been well demonstrated. Sequence and structural similarity search revealed that the bacterium Bacillus megaterium encodes CYP-BM3, which is similar to CYP-2B6. The functional similarity was studied at organism level by batch-scale studies and it was proved that B. megaterium could metabolize endosulfan to endosulfan sulfate, as CYP-2B6 does in human system. The gene expression analyses also confirmed the possible role of CYP-BM3 in endosulfan metabolism. Thus, our results show that the protein structure based in-silico approach can help us to understand and identify microbes for remediation strategy development. To the best of our k.owledge this is the first report which has extrapolated the bacterial gene for endosulfan biotransformation through in silico prediction approach for metabolic gene identification.

Objective: To study the occurrence and serotype diversity of Salmonella isolates in different species of poultry(chick.n, emu and duck. and determine their resistance pattern against various antibiotics of different ***: About 507 samples comprising 202 caecal contents and 305 fecal samples from chick.n, emu and duck.were processed for isolation of Salmonella enterica. Salmonellae were isolated and detected by standard protocol of ISO 6579 Amendment 1: Annex D. Genetic confirmation was also made by using 16 S r RNA genus specific PCR. Serotype specific PCR was also done to detect the most common serovars viz. Salmonella Enteritidis, Salmonella Typhimurium and Salmonella Gallinarum. All obtained isolates were subjected to a set of 25 antibiotics to study their antibiogram by using Baeur-k.rby disk.diffusion ***: Out of 507 samples processed, 32 isolates of Salmonella enterica(18 from caecal contents and 14 from faecal samples) were obtained, of which 24 belonged to 6 different serovars, 6 were untypeable and 2 were rough strains. Salmonella Enteritidis was the most predominant serotype(9), followed by Salmonella Typhimurium(5), Salmonella Virchow(4), Salmonella Gallinarum(3), Salmonella Reading(2) and Salmonella Altona(1). Antibiotic resistance pattern was maximum(100%) to oxacillin, penicillin and clindamycin, followed by ampicillin(68.75%), tetracycline(65.62%), nalidixic acid(56.25%) and colistin(46.87%). High sensitivity of isolates was recorded for chloramphenicol(96.87%) followed by meropenem(84.37%). Conclusions: Occurrence of high proportion of serovars in our study which can cause serious gastroenteritis in humans is a matter of concern. Salmonella Altona has been detected for the first time in India from poultry. This serotype is k.own to cause serious outbreak. of gastroenteritis in humans. Cu-Ni-Sn spinodal alloys(Spinodal bronze)are potential materials with robust applications in components associated with defence applications lik. bearings,propellers,bushes,and shafts of heavily loaded aircraft,off-road vehicles,and *** paper presents a comparative study using water,Brine solution,and SAE 40 oil as the quenching media in regular bronze(Cu-6Sn)and spinodal bronze(Cu-9Ni-6Sn)*** analysis was conducted by optical microscopy,transmission electron microscopy(TEM),and X-ray diffraction technique(XRD)on bronze and spinodal bronze samples immersed in the three different quenching media to understand the grain size and hardness values *** analysis was performed to analyze the effect of quenching media on the wear aspects of bronze and spinodal bronze *** hardness value of the brine-aged spinodal bronze samples was as high as 320 Hv,and the grain size was very low in the range of 60μm.A quantitative comparison between brine-aged regular bronze and brine-aged spinodal bronze showed that the hardness(Hv)was almost 80%higher for brine-aged spinodal ***,the grain size was approximately 30%finer for spinodal bronze when compared with regular *** the load was increased in spinodal bronze samples,there was an initial dip in wear rate followed by a marginal *** was a steady increase in friction coefficient with a rise in load for brine-aged regular bronze and spinodal bronze *** results indicate that brine solution is the most effective quenching medium for cast Cu-Ni-Sn spinodal alloys.

AIM To assess the cleansing efficacy and safety of a new Colon capsule endoscopy(CCE) bowel preparation *** This was a multicenter,prospective,randomized,controlled study comparing two CCE regimens. Subjects were asymptomatic and average risk.for colorectal cancer. The second generation CCE system(Pill Cam? COLON 2;Medtronic,Yoqneam,Israel) was utilized. Preparation regimens differed in the 1 st and 2 nd boosts with the Study regimen using oral sulfate solution(89 m L) with diatrizoate meglumine and diatrizoate sodium solution("diatrizoate solution")(boost 1 = 60 m L,boost 2 = 30 m L) and the Control regimen oral sulfate solution(89 m L) alone. The primary outcome was overall and segmental colon cleansing. Secondary outcomes included safety,polyp detection,colonic transit,CCE completion and capsule excretion ≤ 12 h. RESULTS Both regimens had similar cleansing efficacy for the whole colon(Adequate: Study = 75.9%,Control = 77.3%;P = 0.88) and individual segments. In the Study group,CCE completion was superior(Study = 90.9%,Control = 76.9%;P = 0.048) and colonic transit was more often < 40 min(Study = 21.8%,Control = 4%;P = 0.0073). More Study regimen subjects experienced adverse events(Study = 19.4%,Control = 3.4%;P = 0.0061),and this difference did not appear related to diatrizoate solution. Adverse events were primarily gastrointestinal in nature and no serious adverse events related either to the bowel preparation regimen or the capsule were observed. There was a trend toward higher polyp detection with the Study regimen,but this did not achieve statistical significance for any size category. Mean transit time through the entire gastrointestinal tract,from ingestion to excretion,was shorter with the Study regimen while mean colonic transit times were similar for both study *** A CCE bowel preparation regimen using oral sulfate solution and diatrizoate solution as a boost agent is effective,safe,and achieved superior CCE completion.

Chance fractures are usually associated with seat belt injuries. Mechanism is always related to flexion-distraction at vertebral level. Double level Chance-type fractures have rarely been reported in published literature. We presented such a fracture at D10 and L3 level in a 38-year-old patient with ank.losing spondylitis. Management was done with posterior decompression and short segment fixation separately.
