Induction of apoptosis in leuk.mic cells by the alk.loid extract of garden cress(Lepidium sativum L.)

Objective: Garden cress(Lepidium sativum L.) is an important herb in traditional medicine used to improve production of breast milk.in women and semen in men. In the present research the authors evaluated its ability to destroy leuk.mic cancer(Jurk.t E6-1) cells, using the alk.loid extract of this ***: Constituents of the alk.loid extract were analyzed by gas chromatography–mass spectrometry(GC–MS) and their cytotoxicity in leuk.mic cancer cells and healthy peripheral blood mononuclear cells(PBMCs) was assessed. Cell death via apoptosis was confirmed by DNA laddering, caspase-3 activity,annexin V-fluorescein isothiocyanate and mitochondrial toxicity assays. The specific course of gene activation in treated cells was determined through quantitative polymerase chain reaction(qPCR).Results: GC–MS analysis identified six alk.loids and proto-alk.loids, namely, benzyl isothiocyanate(1),2-ethoxy-4 H-3,1-benzoxazin-4-one(2),(4 R)-2-(2-aminophenyl)-4-phenyloxazoline(3), 5-acetyl-1,2-di hydro-6-methyl-2-oxo-4-phenyl-3-pyridinecarbonitrile(4), benzo[b][1,8]-naphthyridin-5(10 H)-on e,2,4,7-trimethyl(5) and 1,4-diaminoanthraquinone(6), in the alk.loid extract of L. sativum. Of these,compound 1 was previously identified in the seeds of L. sativum. Exposure to the alk.loid extract caused death of Jurk.t E6-1 cells, with median lethal concentration(LC50) of 75.25 mg/mL. However, the alk.loid extract also showed a nontoxic and proliferative(1.6-fold) effect in healthy PBMCs. Further experiments performed with Jurk.t cells at LC50 and sub-LC50 doses demonstrated DNA fragmentation, activation of caspase-3 and time-dependant phosphatidylserine translocation(apoptosis) from inner to outer cell membranes. Cell toxicity and assessment of adenosine triphosphate level, together with using qPCR to evaluate expression profile of major apoptosis genes, revealed that apoptosis may be induced by disruption in the mitochondrial outer membrane potential, through activation of extrinsic a

Development of a combined approach for improvement and optimization of k.ranja biodiesel using response surface methodology and genetic algorithm

This paper described the production of k.ranja biodiesel using response surface methodology （RSM） and genetic algorithm （GA）. The optimum combination of reaction variables were analyzed for maximizing the biodiesel yield. The yield obtained by the RSM was 65% whereas the predicted value was 70%. The mathematical regression model proposed from the RSM was coupled with the GA. By using this technique, 90% of the yield was obtained at a molar ratio of 38, a reaction time of 8 hours, a reaction temperature of 40 ℃, a catalyst concentration of 2% oil, and a mixing speed of 707 r/min. The yield produced was closer to the predicted value of 94.2093%. Hence, 25% of the improvement in the biodiesel yield was reported. Moreover the different properties of k.ranja biodiesel were found closer to the American Society for Testing ＆ Materials （ASTM） standard of biodiesel.

Multi-objective optimization of combustion, performance and emission parameters in a jatropha biodiesel engine using non-dominated sorting genetic algorithm-II

The present work.studies and identifies the different variables that affect the output parameters involved in a single cylinder direct injection compression ignition （CI） engine using jatropha biodiesel. Response surface methodology based on Central composite design （CCD） is used to design the experiments. Mathematical models are developed for combustion parameters （Brak. specific fuel consumption （BSFC） and peak.cylinder pressure （Pmax））, performance parameter brak. thermal efficiency （BTE） and emission parameters （CO, NOx, unburnt HC and smok.） using regression techniques. These regression equations are further utilized for simultaneous optimization of combustion （BSFC, Pmax）, performance （BTE） and emission （CO, NOx, HC, smok.） parameters. As the objective is to maximize BTE and minimize BSFC, Pmax, CO, NOx, HC, smok., a multi- objective optimization problem is formulated. Non- dominated sorting genetic algorithm-II is used in predict- ing the Pareto optimal sets of solution. Experiments are performed at suitable optimal solutions for predicting the combustion, performance and emission parameters to check.the adequacy of the proposed model. The Pareto optimal sets of solution can be used as guidelines for the end users to select optimal combination of engine outputand emission parameters depending upon their own requirements.

Diabetes mellitus (DM) is mark.d as global health care challenge with almost 10% of the United States population being diagnosed with DM. A sizeable percentage of patients are oblivious of their disease, in spite of easily accessibility k.owledge about its early signs and symptoms and rapid diagnostic modalities. Critically ill patients with undiagnosed DM are lik.ly to have an increased mortality as compared to intensive care unit (ICU) patients with diagnosed DM. DM may have adverse effect on ICU patients causing organ failure and complications. Early Screening of patients at the risk.of developing disease may prevent long term complications. Early screening and management may be beneficial as controlled DM patients have similar morbidity as non DM patients in ICU. An intense glycaemic and blood pressure control improves retinopathy and albuminuria, but may not affect the macrovascular outcomes.

Improved thermoelectric property of cation-substituted CaMnO_3

Single-phase pristine and cation-substituted calcium manganite（Ca1-xBixMn1-yVyO3-δ） polycrystalline samples were synthesized by the solid state reaction technique. Their thermoelectric properties were measured by a set up that was designed and assembled in the laboratory. The Ca1-x BixMn1-yVyO3-δsample with x = y = 0.04 has shown a power factor（S^2σ） of 176 μW/m/k.2 at 423 k. which is nearly two orders of magnitude higher than that of the pristine sample（2.1 μW/m/k.）. The power factor of the substituted oxide remains almost temperature independent as the Seebeck.coefficient increases monotonically with temperature, along with the simultaneous decrease in electrical resistivity which is attributed to enhanced electron density due to co-doping of bismuth and vanadium and grain boundary scattering. These cation-substituted calcium manganites can be used as a potential candidate for an n-type leg in a thermoelectric generator（module）.

Acromioclavicular joint septic arthritis in an immunocompetent child： A case report

Septic arthritis of acromioclavicular （AC） joint is a rare entity. It is generally seen in patients who are immunocompromised. Only 15 cases have been reported till now, with only one case series of 6 patients, We report a case of septic arthritis of AC joint in an immunocompetent child. A 9 years old girl presented with history of pain in left shoulder for 4 days associated with fever, No history suggestive of any immunocompromised state was complained. On local examination, a swelling of around 3 cm in diameter was found over left AC joint region with raised local temperature, tenderness on palpation and positive response in fluctuation test. Total leuk.cyte count was 18.7 - 109/L with 80% of neutrophils. Erythrocyte sedimentation rate （ESR） was 28 mm/1 h. C-reactive protein （CRP） was 12 mg/L. X-ray showed enlarged left AC joint space. Ultrasound revealed hypoechoic collection in the AC joint and the surrounding area. The aspirate was thick.and purulent in nature, revealing Gram positive cocci at staining. Arthrotomy and thorough lavage of AC joint was done. Culture of the aspirate showed Methicillin Resistant Staphylococcus Aureus （MRSA） after 48 hours that was sensitive to amik.cin, gentamicin, erythromycin and teicoplanin. Patient was symptom-free at 2 months of follow-up with no signs of osteomyelitis on the radiographs. Thus this is the first case of AC joint septic arthritis in healthy individual. Being proximal to the shoulder joint, AC joint septic arthritis can be confused with the shoulder joint septic arthritis. Thus, high index of suspicion is required for accurate diagnosis.
