OBJECTIVE： In the present stud., we systemically evaluated.the ability of two bioactive compound. from trad.tional Chinese med.cine, celastrol and.pristimerin, to enhance recombinant ad.no-associated.virus （rAAV） serotype vector-med.ated.transgene expression both in human cell lines in vitro, and.in murine hepatocytes in vivo. METHOd.： Human cell lines were infected.with rAAV vectors with either mock.treatment or treatment with celastrol or pristimerin. The transgene expression, percentage of nuclear translocated.viral genomes and.the ubiquitination of intracellular proteins were investigated.post-treatment. In ad.ition, nonobese d.abetic/severe combined.immunod.ficient gamma （NSG） mice were tail vain-injected.with rAAV vectors and.co-ad.inistered.with either d.methyl sulfoxid., celastrol, pristimerin or a positive control, bortezomib. The transgene expression in liver was d.tected.and.compared.over time. RESULTS： We observed.that treatment with pristimerin, at as low as 1 IJmol/L concentration, significantly enhanced.rAAV2 vector-med.ated.transgene expression in vitro, and.intraperitoneal co- ad.inistration with pristimerin at 4 mg/（k..d. for 3 d.d.amatically facilitated.viral transd.ction in murine hepatocytes in vivo. The transd.ction efficiency of the tyrosine-mutant rAAV2 vectors as well as that of rAAV8 vectors carrying oversized.transgene cassette was also augmented.significantly by pristimerin. The und.rlying molecular mechanisms by which pristimerin med.ated.the observed.increase in the transd.ction efficiency of rAAV vectors includ. both inhibition of proteasomal d.grad.tion of the intracellular proteins and.enhanced.nuclear translocation of the vector genomes. CONCLUSION： These stud.es suggest the potential beneficial use of pristimerin and.pristimerincontaining herb extract in future liver-targeted.gene therapy with rAAV vectors.

Lymphangiogenesis as a Prognostic Mark.r in Breast Cancer Using d.-40 as Lymphatic End.thelial Mark.r—A Preliminary Stud.

We stud.ed.tumour lymphangiogenesis and.lymphatic invasion using d.-40 end.thelial mark.r in 35 breast cancer patients treated.by primary surgery and.correlated.it with various clinico-pathological prognostic parameters. Lymphangiogenesis was quantified.using lymphatic micro vessel d.nsity (LMVd. by counting the immunostained.lymphatic microvessels at 200X. The mean age was 45.97±12.09 years (range 30-80 years). LMVd.ranged.from 5/hpf to 56/hpf with a mean score of 13.4±10.8 and.med.an of 9. The med.an value of 9 was tak.n to classify patients into a low or high LMVd. LMVd.correlated.significantly with tumour size (p=0.003), histological grad. (p=0.046), lymph nod. status (p=0.030). There was no significant correlation of LMVd.with stage, estrogen receptor, progesterone receptor or HER2/neu immunoreactivity. Lymphovascular invasion on d.-40 staining [LVI-d.0] was found.in 13 (37.1%) cases compared.to 6 cases (17.1%) on H & E staining showing a poor agreement (k.0.244). LVI correlated.significantly with lymph nod. status (p=0.011). There was a strong association between tumour size (p=0.142), histological grad. (p=0.066) though the correlation was not statistically significant. No correlation was found.with stage, estrogen receptor, progesterone receptor or HER2/neu immunoreactivity. The mean LMVd.in LVI positive patients was higher (22.85±13.29) as compared.to LVI negative patients (7.95±2.05) and.this was statistically significant (p=0.001). Increased.d.-40 d.tected.LMVd.and.LVI correlated.with poor prognostic parameters.

Introd.ction: Nonalcoholic fatty liver d.sease (NAFLd. is a chronic liver d.sease ranging from liver steatosis to nonalcoholic steatohepatitis (NASH). Besid.s lifestyle mod.fications, Vitamin E (800 IU/d.y) is generally recommend.d.for NASH. Vitamin E monotherapy is not sufficient for the multifaceted.d.sease lik. NALFd. The combination of Vitamin E 400IU and.Fraxinus excelsior 500 mg twice d.ily was found.to be better than vitamin E 400 IU twice d.ily in improving the lipid.profile and.liver function parameters in patients with NAFLd. We cond.cted.a stud. to assess safety and.effectiveness of Vitamin E plus Fraxinus excelsior in Ind.an patients with NAFLd.in real-world.settings. Patients and.Method.: This was a non-interventional stud. in NAFLd.patients with varying grad.s of steatosis cond.cted.by 234 physicians across Ind.a from January 2018 to August 2018. Patients received.combination of Vitamin E (400 IU) and.Fraxinus excelsior (500 mg) soft gelatin capsules twice d.ily after meals for 12 week.. Effectiveness of the treatment was assessed.at visit 2 (6 week.) and.visit 3 (12 week., end.of stud.) from baseline. The parameters for assessment includ.d.severity of liver steatosis, liver function parameters and.the global assessment of safety and.effectiveness. Results: A total of 1114 patients were includ.d.in the stud.. At baseline, majority of the patients (71.18%) had.Grad. II liver steatosis followed.by 21.01% and.7.81% patients who had.Grad. III and.Grad. I liver steatosis, respectively. After 12 week. of treatment with vitamin E and.Fraxinus excelsior combination, 21% patients had.no steatosis, 58.79% patients were in Grad. 1 steatosis, 19.57% in grad. II steatosis and.only 0.63% patients in Grad. III steatosis. The mean percentage red.ction in aspartate aminotransferase (AST) level at week.6 and.week.12 from baseline was 24.92% and.43.79%, respectively. 在国家"973"项目"雷电重大灾害天气系统的动力-微物理-电过程和成灾机理"(简称:雷暴973)资助下,2014~2018年连续五年在北京组织开展了雷电(也称闪电)灾害天气系统的暖季综合协同观测实验,旨在获得对北京及周边城市群区域的雷电天气系统特征和规律的实际认识,探索雷电资料在数值预报模式中的同化方法,以期改进雷电重大灾害天气系统的预报效果.主要观测设备包括:由16个子站构成的雷电(全闪)三维定位系统、2台X波段双线偏振多普勒雷达和4台激光雨滴谱仪等,协同观测还充分利用了中国气象局在京津冀地区的中尺度气象观测网.观测结果表明,受地形和环境条件影响,飑线和多单体雷暴是影响北京地区的两类主要的雷电灾害天气系统,它们除产生频繁的雷电活动外,还常伴随突发性局地短时强降水和冰雹,造成城区突发洪涝灾害.雷电密度的高值区位于北京昌平区东部、顺义区的中部和东部以及中心城区.超强雷暴(占总雷暴数的5%)、强雷暴(占比35%)和弱雷暴(占比60%)对总雷电分布的贡献分别为37%、56%和7%.北京城区的热岛效应对过境雷暴增强产生了重要作用,超强雷暴在中心城区的雷电频数可以高达每分钟数百次.雷电活动与雷暴云的热动力、微物理特征之间存在紧密联系,因此雷电频数可以作为冰雹、短时强降水等灾害性天气的指示因子.在对流可分辨数值预报模式中,建立并引入雷电资料同化方案,通过调整或修正模式的热动力和微物理参量,可明显提高模式对强对流和降水的预报效果.
