AIM: To elucidate the sequential transfer of iron amongst ferritin, transferrin and transferrin receptor under various iron status conditions. METHODS: Incorporation of 59Fe into mucosal and luminal proteins was carried out in control WkY rats. The sequential transfer of iron amongst ferritin, transferrin and transferrin receptor was carried out in iron deficient, control and iron overloaded rats. The duodenal proteins were subjected to immunoprecipitation and quantitation by specific ELISA and in situ localization by microautoradiography and immunohistochemistry in tandem duodenal sections. Human duodenal biopsy (n = 36) collected from subjects with differing iron status were also stained for these proteins. RESULTS: Ferritin was identified as the major protein that incorporated iron in a time-dependent manner in the duodenal mucosa. The concentration of mucosal ferritin was significantly higher in the iron excess group compared to control, iron deficient groups (731.5 ± 191.96 vs 308.3 ± 123.36, 731.5 ± 191.96 vs 256.0 ± 1.19, P kes up iron through a sequential transfer involving interaction of luminal transferrin, transferrin-transferrin receptor and ferritin.

AIM： To understand the digestive stability and mechanism of release and intestinal uptake of pea ferritin iron in caco-2 cell line model.METHODS： Pea seed ferritin was purified using salt fractionation followed by gel filtration *** bioavailability of ferritin iron was assessed using coupled in vitro digestion/Caco-2 cell model in the presence or absence of ascorbic acid and phytic ***-2 cell ferritin formation was used as a surrogate marker of iron uptake. Structural changes of pea ferritin under simulated gastric pH were characterized using electrophoresis, gel filtration and circular dichroism spectroscopy.RESULTS： The caco-2 cell ferritin formation was significantly increased （P 〈 0.001） with FeSO4 （19.3±9.8 ng/mg protein） and pea ferritin （13.9 ± 6.19 ng/mg protein） compared to the blank digest （3.7 ± 1.8 ng/mg protein）. Ascorbic acid enhanced while phytic acid decreased the pea ferritin iron bioavailability. However,either in the presence or absence of ascorbic acid, the ferritin content of caco-2 cells was significantly less with pea ferritin than with FeSO4. At gastric pH, no band corresponding to ferritin was observed in the presence of pepsin either on native PAGE or SDS-PAGE. Gel filtration chromatography and circular dichroism spectroscopy revealed a pH dependent loss of quaternary and secondary structure.CONCLUSION： Under gastric conditions, the iron core of pea ferritin is released into the digestive medium due to acid induced structural alterations and dissociation of protein. The released iron interacts with dietary factors leading to modulation of pea ferritin iron bioavailability,resembling the typical characteristics of non-heme iron.

AIM： To understand the interactions between iron and zinc during absorption in iron- and zinc-deficient rats, and their consequences on intestinal oxidant-antioxidant balance. METHODS： Twenty-four weanling Wistar-kyoto rats fed an iron- and zinc-deficient diet （〈 6.5 mg Fe and 4.0 mg Zn/kg diet） for 4 wk were randomly divided into three groups （n = 8, each） and orally gavaged with 4 mg iron, 3.3 mg zinc, or 4 mg iron ＋ 3.3 mg zinc for 2 wk. At the last day of repletion, 3 h before the animals were sacrificed, they received either 37 mBq of SSFe or ^65Zn, to study their localization in the intestine, using microautoradiography. Hemoglobin, iron and zinc content in plasma and liver were measured as indicators of iron and zinc status. Duodenal sections were used for immunochemical staining of ferritin and metallothionein. Duodenal homogenates （mitochondrial and cytosolic fractions）, were used to assess aconitase activity, oxidative stress, functional integrity and the response of antioxidant enzymes. RESULTS： Concurrent repletion of iron- and zinc-deficient rats showed reduced localization of these minerals compared to rats that were teated with iron or zinc alone; these data provide evidence for antagonistic interactions. This resulted in reduced formation of lipid and protein oxidation products and better functional integrity of the intestinal mucosa. Further, combined repletion lowered iron-associated aconitase activity and ferritin expression, but significantly elevated metallothionein and glutathione levels in the intestinal mucosa. The mechanism of interactions during combined supplementation and its subsequent effects appeared to be due to through modulation of cytosolic aconitase, which in turn influenced the labile iron pool and metallothionein levels, and hence reduced intestinal oxidative ***： Concurrent administration of iron and zinc corrects iron and zinc deficiency, and also reduces the intestinal oxidative damage associated with iron suppl

In this work, we demonstrated a simple and efficacious two-step method for the synthesis of Ag@Au core–shell nanoparticles(Ag@Au NPs) and the Ag/Au hollow nanocages(Ag/Au NCs) with Ag nanoparticles(Ag NPs) as seeds by adjusting p H, and the preparation of hybrid Ag@Au NPs- or Ag/Au NCs-graphene oxide nanocomposites(Ag@Au NPs-GO or Ag/Au NCs-GO)based on the self-assembly. It was noticed from the electrostatic assembly experiment that the loading amount of Ag/Au NCs on GO nanosheet was more than that of Ag@Au NPs. The as-synthesized hybrid materials were characterized by transmission electron microscopy, atomic force microscopy, f-potential, high-angle annular darkfield scanning transmission electron microscopy, thermogravimetric analyzer and X-ray diffraction. Catalytic activities of Ag@Au NPs, Ag/Au NCs and Ag/Au NCs-GO nanostructures were investigated in the reduction of 4-, 3-or 2-nitrophenol to 4-, 3- or 2-aminophenol, and on the basis of comparative kinetic studies the following trend was obtained for the related catalytic activity: Ag/Au NCsGO [ Ag/Au NCs [ Ag@Au NPs. These observations were attributed to the simultaneous effects of surface area available for catalytic reaction and composition of the hybrid nanostructures.

Re-irradiation(Re-RT)in liver tumours is rarely reported owing to poor tolerance of liver and high incidence of radiation induced liver disease *** based robotic radiosurgery allows to deliver high dose radiation to the liver tumour and restricts the dose to healthy uninvolved liver,thereby increasing the potential for *** to radiation is low for entire liver and hence re-radiation is a *** the other hand,as regenerative potential of hepatocytes is rapid,replacement of necrotic liver tissue occurs with regenerated *** regenerated hepatocytes are radiation na?ve,do not have"memory"of radation therapy treatment and hence have potential of *** are reporting a series of two breast cancer patients presented with liver oligometastasis treated with fiducial based Cyberknife system(Ck).Both the patients were treated multiple times with Ck and had long-term survival(>2 years)without any clinical features of radiation induced liver *** selected patients are suitable for multiple sessions of Ck for liver lesions with long-term outcome.
