随着光动力学疗法(photodynamic therapy,PDT)基础研究的不断深入和临床应用的广泛开展,如何精确量化光动力剂量,并根据患者的个体差异进行剂量的实时调整和优化已成为亟待解决的挑战性难题,属PDT研究的前沿热点.综述了现有PDT剂量学研究方法及其相应检测技术的研究进展,其中包括:a.测定光通量密度、光敏剂浓度和氧分压;b.测量光敏剂的光漂白速率和光致产物;c.监测PDT前后组织的光生物学响应;d.检测单态氧在1270nm的近红外发光.同时,还分析了这些PDT剂量学方法的优点和局限性.最后,讨论了PDT剂量学研究中所面临的挑战.

Craniomaxillofacial reconstruction implants,which are extensively used in head and neck surgery,are conventionally made in standardized *** surgery,the implant must be bended manually to match the anatomy of the individual *** bending process is time-consuming,especially for inexperienced ***,repetitive bending may induce undesirable internal stress concentration,resulting in fatigue under masticatory loading in v iv o and causing various complications such as implant fracture,screw loosening,and bone *** have been reports on the use of patient-specific 3D-printed implants for craniomaxillofacial reconstruction,although few reports have considered implant *** this paper,we present a systematic approach for making 3D-printed patientspecific surgical implants for craniomaxillofacial *** approach consists of three parts:First,an easy-to-use design module is developed using Solidworks®software,which helps surgeons to design the implants and the axillary fixtures for *** engineers can then carry out the detailed design and use finite-element modeling(FeM)to optimize the ***,the fabrication process is carried out in three steps:0 testing the quality of the powder;(2)setting up the appropriate process parameters and running the 3D printing process;and (3)conducting post-processing treatments(i.e.,heat and surface treatments)to ensure the quality and performance of the ***,the operation begins after the final checking of the implant and *** the surgery,postoperative rehabilitation follow-up can be carried out using our patient tracking *** this systematic approach,we have successfully conducted a total of 41 surgical cases.3D-printed patient-specific implants have a number of advantages;in particular,their use reduces surgery time and shortens patient recovery ***,the presented approach helps to ensure implant quality.

Several modes of eukaryotic of DNA double strand break repair (DSBR) depend on synapsis of complementary DNA. The Rad51 ATPase, the S. cerevisiae homolog of e. coli RecA, plays a key role in this process by catalyzing homology searching and strand exchange between an invading DNA strand and a repair template (e.g. sister chromatid or homologous chromosome). Synthesis dependent strand annealing (SDSA), a mode of DSBR, requires Rad51. Another repair enzyme, the Rad1-Rad10 endonuclease, acts in the final stages of SDSA, hydrolyzing 3￠ overhanging single-stranded DNA. Here we show in vivo by fluo-rescence microscopy that the ATP binding function of yeast Rad51 is required to recruit Rad10 SDSA sites indicating that Rad51 pre-synaptic filament formation must occur prior to the recruitment of Rad1-Rad10. Our data also show that Rad51 ATPase activity, an important step in Rad51 filament disassembly, is not absolutely required in order to recruit Rad1- Rad10 to DSB sites.

Reactive Arrays of Colorimetric Sensors for Metabolite and Steroid Identification

The work described herein examines a rapid mix-and-measure method called DeTeCHIP suitable for screening of steroids and metabolites. The addition of steroids and metabolites to reactive arrays of colorimetric sensors generated characteristic color “fingerprints” that were used to identify the analyte. A color analysis tool was used to identify the analyte pool that now includes biologically relevant analytes. The mix-and-measure arrays allowed the detection of disease metabolites, orotic acid and argininosuccinic acid;and the steroids androsterone, 1,4-androstadiene, testosterone, stanozolol, and estrone. The steroid 1,4-androstadiene was also detected by this method while dissolved in synthetic urine. Some of the steroids, such as androstadiene, stanozolol, and androsterone were co-dissolved with (2-hydroxypropyl)-β-cyclodextrin in order to increase solubility in aqueous buffered solutions. The colorimetric arrays do not intend to eliminate eLISA or mass spectroscopy based screening, but to possibly provide an alternative analytical detection method for steroids and metabolites.

Because of their ability to sustain extremely high-amplitude electromagnetic fields and transient density and field profiles,plasma optical components are being developed to amplify,compress,and condition high-power laser *** recently demonstrated the potential to use a relativistic plasma aperture—produced during the interaction of a high-power laser pulse with an ultrathin foil target—to tailor the spatiotemporal properties of the intense fundamental and second-harmonic light generated[Duff et al.,***.10,105(2020)].Herein,we explore numerically the interaction of an intense laser pulse with a preformed aperture target to generate second-harmonic laser light with higher-order spatial *** maximum generation efficiency is found for an aperture diameter close to the full width at half maximum of the laser focus and for a micrometer-scale target *** spatial mode generated is shown to depend strongly on the polarization of the drive laser pulse,which enables changing between a linearly polarized TeM01 mode and a circularly polarized Laguerre–Gaussian LG01 *** demonstrates the use of a plasma aperture to generate intense higher-frequency light with selectable spatial mode structure.

The Food and Drug Administration(FDA) has approved two mechanismbased treatments for tuberous sclero-sis complex(TSC)-everolimus and vigabatrin. However, these treatments have not been systematically studied in individuals with TSC and severe autism. The aim of this review is to identify the clinical features of severe autism in TSC, applicable preclinical models, and potential barriers that may warrant strategic planning in the design phase of clinical trial development. A comprehensive search strategy was formed and searched across Pub Med, embase and SCOPUS from their inception to 2/21/12, 3/16/12, and 3/12/12 respectively. After the final search date, relevant, updated articles were selected from Pub Med abstracts generated electronically and emailed daily from Pub Med. The references of selected articles were searched, and relevant articles were selected. A search of *** was completed using the search term "TSC" and "tuberous sclerosis complex". Autism has been reported in as many as 60% of individuals with TSC; however, review of the literature revealed few data to support clear classification of the severity of autism in TSC. Variability was identified in the diagnostic approach, assessment of cognition, and functional outcome among the reviewed studies and case reports. Objective outcome measures were not used in many early studies; however, diffusion tensor imaging of white matter, neurophysiologic variability in infantile spasms, and cortical tuber subcategories were examined in recent studies and may be useful for objective classification of TSC in future studies. Mechanism-based treatments for TSC are currently available. However, this literature review revealed two potential barriers to successful design and implementation of clinical trials in individuals with severe autism-an unclear definition of the population and lack of validated outcome measures. Advance Directives in the Context of Uncertain Prognosis for Residents of Nursing Homes

This report documents the findings of a mixed-methods study focused on the advanced directives of 182 residents of three LTC facilities in southern Ontario, Canada. Although almost all had a completed advance directive within 3 months of death, most did not have a palliative designation or directive until a few days before they died. each facility’s written Progress Notes revealed staff members usually sought additional confirmation of care preferences from residents’ substitute decision-makers within a few days of the death. It was thus common for advance directives to change from a more interventionist approach to the least interventionist approach near death. This change indicates that the meaning and significance of advance care planning and resulting advance directives must be considered in light of the processes and temporal factors involved in their completion and use within this distinct population. The relational nature of advance care planning and concern about ageism as a factor for withholding or withdrawing life support for LTC residents are considered as possible explanatory factors. These findings and their implications are described in relation to end-of-life care policies and practices in LTC facilities.
