Preliminary analysis of two NAC transcription factor expression patterns in Larix olgensis

The NAC transcription factor family is plant-specific with various biological functions.However,there are few studies on the NAC gene involving coniferous species.Bioinformatics research and expression analysis of NAC genes in Larix olgensis can be used to analyse the function of the NAC gene in the future.Screening of excellent genetic materials and molecular breeding have been utilized to cultivate high-quality,stress-resistant larches.According to the transcriptome data for L.olgensis,the genes Uni-gene81490 and Unigene70699 with complete ORFs(open reading frames)were obtained by conserved domain analy-sis and named LoNAC1 and LoNAC2,respectively.The cDNAs of LoNAC1 and LoNAC2 were 1971 bp and 1095 bp in length,encoding 656 and 364 amino acids,respectively.The molecular weights of the proteins encoded by the two genes were predicted to be 72.61 kDa and 41.13 kDa,and subcellular localization analysis indicated that the proteins were concentrated in the nucleus.The results of real-time quantitative PCR analysis showed that at different growth stages and in different tissues of L.olgensis,the relative expression levels of the two NAC genes were highest in the stem,and the expression differences were more obvious in non-lignified tissues.After drought,salt and alkali stress and hormone treatment,expression was induced to different degrees.The expression levels of LoNAC1 and LoNAC2 in semi-lignified L.olgensis were higher than in the other two periods(non-lignified and lignified),and expression levels significantly increased under drought and salt stress.Relative expression levels changed under hormone treatment.It is speculated that these two genes may not only be related to drought and salt stress and secondary growth but may also be induced by hormones such as abscisic acid.Overall,LoNAC1 and LoNAC2 are genetic materials that can be used for molecular breeding of larch.

Model-based clustering is popularly used in statistical literature, which often models the data with a Gaussian mixture model. As a consequence, it requires estimation of a large amount of parameters, especially when the data dimension is relatively large, in this paper, reduced-rank model and group-sparsity regularization are proposed to equip with the model-based clustering, which substantially reduce the number of parameters and thus facilitate the high-dimensional clustering and variable selection simultaneously. We propose an EM algorithm for this task, in which the M-step is solved using alternating minimization. One of the alternating steps involves both nonsmooth function and nonconvex constraint, and thus we propose a linearized alternating direction method of multipliers （ADMM） for solving it. This leads to an efficient algorithm whose subproblems are all easy to solve. In addition, a model selection criterion based on the concept of clustering stability is developed for tuning the clustering model. The effectiveness of the proposed method is supported in a variety of simulated and real examples, as well as its asymptotic estimation and selection consistencies.

A synergetic strategy of NIR-Ⅱ squaraine dyes with ultrahigh photothermal conversion efficiency for photothermal therapy

Recent research on photothermal therapy(PTT) has sparked significant interest in the development of new organic photothermal agents(PTAs),ranging from single-molecule to aggregated levels.However,controlling aggregation pathways for PTAs with ultrahigh photothermal conversion efficiency(PCE) remains a major challenge.Herein,a two-pronged approach utilizing “Haggregation” and “intramolecular motion” was employed to enhance the PCE of an acceptor-substituted squaraine dye(NSQs).The C2vmolecular symmetry of the NSQs,which possess a ground state dipole moment(μg),promotes H-dimeric aggregates through dipole-dipole counteraction.Peripheral triphenylethylene or diphenylamine groups were added to this H-dimeric nanoplatform.This was done to enhance intramolecular motions for heat generation and also to extend conjugation,which redshifted the optical absorption and balanced the blue-shift induced by H-aggregation.With this technique,an organic PTA with NIR-II absorption was developed,and its nanoparticle achieved a remarkable PCE of 86.3% under 1,064 nm laser excitation.Femtosecond transient absorption spectroscopy and quantum mechanical calculations demonstrated the accelerated internal conversion process in NIR-II PTAs for rapid heat generation.The NSQs nanoparticles exhibit superior photothermal therapeutic properties for in vivo photoacoustic imaging-guided PTT,demonstrating the potential of bottom-up design to enable synergistic engineering strategies towards efficient phototheranostic agents.

Synoptic-Scale Analysis on Development and Maintenance of the 19–21 July 2021 Extreme Heavy Rainfall in Henan,Central China

In this paper,synoptic-scale analyses of frontogenesis,moisture budget,and tropospheric diabatic heating are performed to reveal the development and maintenance mechanisms for the extreme heavy rainfall in Henan Province of central China from 19 to 21 July 2021,based on station observations and the ECMWF Reanalysis version 5(ERA5)data.The results demonstrate that owing to the blocking effect of local topography,low-level wind convergence in Henan appeared underneath high-level divergence,conducive to development and maintenance of a midtropospheric low-pressure system saddled by the Asian continental high and the western Pacific subtropical high(WPSH),during the extreme heavy rainfall.In the lower troposphere,frontogenesis occurred in the θ_(se) intensive region,as a result of the divergence and horizontal deformation(which play equally important roles),generating frontal secondary circulation with strong vertical motion favorable to heavy rainfall.Moisture budget analysis reveals that 1)with the continuous strengthening of the easterly wind from the north side of Typhoon In-Fa(2106),strong wind shear and orographic uplift led to abnormally strong convergence of water vapor flux in the boundary layer in Henan;2)there occurred extremely strong net inflow of moisture in the boundary layer from the east.Horizontally,both the apparent heat source and the moisture sink coincided with the area of heavy rainfall;vertically,however,Q_(1)exhibited a single peak with the heating center in the middle and upper troposphere,while large Q_(2)values evenly resided over 850–400 hPa;and Q_(1)(Q_(2))was dominated by vertical(horizontal)transport of potential temperature(moisture).These indicate that the latent heat release from condensation of initial heavy rainfall provided a positive feedback,leading to increasingly heavy precipitation.All these synoptic settings sustained the extreme rainfall process.

Ordered mesoporous carbon（OMC） with high specific surface area and large pore volume was synthesized and tested for use as an adsorbent for volatile organic compound（VOC）disposal. Benzene, cyclohexane and hexane were selected as typical adsorbates due to their different molecular sizes and extensive utilization in industrial processes. In spite of their structural differences, high adsorption amounts were achieved for all three adsorbates, as the pore size of OMC is large enough for the access of these VOCs. In addition, the unusual bimodal-like pore size distribution gives the adsorbates a higher diffusion rate compared with conventional adsorbents such as activated carbon and carbon molecular sieve. Kinetic analysis suggests that the adsorption barriers mainly originated from the difficulty of VOC vapor molecules entering the pore channels of adsorbents. Therefore, its superior adsorption ability toward VOCs, together with a high diffusion rate, makes the ordered mesoporous carbon a promising potential adsorbent for VOC disposal.

A Prototype for Diagnosis of Psoriasis in Traditional Chinese Medicine

Psoriasis is a chronic,non-communicable,painful,disfiguring and disabling disease for which there is no cure,with great negative impact on patients’quality of life(QoL).Diagnosis and treatment with traditional Chinese medical technique based on syndrome differentiation has been used in practice for a long time and proven effective,though,up to now,there are only a few available studies about the use of semantic technologies and the knowledge systems that use Traditional Chinese Medicine(TCM)-syndrome differentiation for information retrieval and automated reasoning.In this paper we use semantic techniques based on ontologies to develop a prototypical system for the diagnosis of Psoriasis.For this purpose,a domain ontology is developed for syndrome differentiation of psoriasis vulgaris(PV).This ontology is founded on an adapted version of the general formal ontology(GFO),with the evidence-based clinical practice guideline of TCM for psoriasis vulgaris(Guideline 2013)as the primary data sources.The implemented prototype,called ONTOPV,contains this domain ontology and is aimed at a decision support system for diagnosis and treatment of PV.This system uses a case-database for Case Based Reasoning(CBR),combined with fuzzy pattern recognition.Experimental results show that the ONTOPV realizes the basic functionalities of data collection,querying,browsing and navigation,and supports rule-based knowledge reasoning,and integrates fuzzy pattern recognition.It can provide users with clinical decision support for TCM syndrome differentiation in diagnosis of psoriasis.

Interactive response of photosynthetic characteristics in Haloxylon ammodendron and Hedysarum scoparium exposed to soil water and air vapor pressure deficits

C4 plants possess better drought tolerance than C3 plants. However, Hedysarum scoparium, a C3 species, is dominant and widely distributed in the desert areas of northwestern China due to its strong drought tolerance. This study compared it with Haloxylon ammodendron, a C4 species, regarding the interactive effects of drought stress and different leaf–air vapor pressure deficits. Variables of interest included gas exchange, the activity levels of key C4 photosynthetic enzymes, and cellular anatomy. In both species, gas exchange parameters were more sensitive to high vapor pressure deficit than to strong water stress, and the net CO2 assimilation rate（A n） was enhanced as vapor pressure deficits increased. A close relationship between A n and stomatal conductance（g s） suggested that the species shared a similar response mechanism. In H. ammodendron, the activity levels of key C4 enzymes were higher, including those of phosphoenolpyruvate carboxylase（PEPC） and nicotinamide adenine dinucleotide phosphate-malate enzyme（NADP-ME）, whereas in H. scoparium, the activity level of nicotinamide adenine dinucleotide-malate enzyme（NAD-ME） was higher.Meanwhile, H. scoparium utilized adaptive structural features, including a larger relative vessel area and a shorter distance from vein to stomata, which facilitated the movement of water. These findings implied that some C4 biochemical pathways were present in H. scoparium to respond to environmental challenges.

Lysine acetylation,a reversible and dynamic post-translational modification,plays pivotal roles in regulating many biological processes in plants.However,the information of lysine acetylation during flower development of woody angiosperm remains unclear.Herein,we identified a total of 667 lysine acetylation sites in 467 proteins in flower buds of Catalpa Bungei.The acetylated proteins were mainly involved in the pathways of carbon metabolism,glycometabolism and oxidative phosphorylation.Using functional enrichment analysis,61 and 13 acetylated proteins were involved in the glycometabolism and oxidative phosphorylation pathways,respectively.This suggests that lysine acetylated proteins play critical roles in the energy metabolism pathways.Accordingly,the protein–protein interactions were also highly enriched in the carbon metabolic pathway.Integrative analysis of lysine acetylome and transcriptome showed that transcript level changes of seven key lysine-acetylated proteins/genes involved in energy metabolism were significant different during floral transition.Importantly,compared with the normal flowering variety,the expression levels of phosphoribulokinase,phosphoenolpyruvate carboxykinase,ATP synthase and pyruvate dehydrogenase complex genes were significantly up-regulated at the stages of vegetative and transition buds during early flowering of C.bungei.These data represent the first lysine acetylome in the C.bungei flowers,and can serve as an essential basis for exploring the function of lysine acetylated proteins in the flower bud development of plants.

Catalpa fargesii is an important economic tree species used for furniture and timber production because of its high density and hardness.Its survival and growth are severely aff ected and primarily limited by drought stress.Thus,to better understand the mechanism of drought resistance in C.fargesii,we used qRT-PCR to reveal signifi cantly diff erent expression of three plasma membrane intrinsic protein genes:CfPIP1-1,CfPIP1-2 and CfPIP1-4.We then cloned their full-length cDNA sequences and characterized the encoded proteins.We analyzed the genes phylogenetically and predicted conserved motifs,domains,and secondary and tertiary structures.To verify the function ofthe CfPIP1 genes further,we ectopically expressed CfPIP1 transgenes in Arabidopsis thaliana.The results showed that CfPIP1-1,CfPIP1-2 and CfPIP1-4 had several characteristics of aquaporins.The transgenic plants grew better than the WT plants did under drought stress,and overexpression of the CfPIP1 genes increased the plant water content and resistance to drought.Thus,CfPIP1-1,CfPIP1-2 and CfPIP1-4 of C.fargesii play key roles in regulating the intracellular and extracellular water balance and in mediating the plant response to drought.
