Deciphering the molecular mechanisms of Simiaowan in the treatment of hyperuricemia: in vivo and in silico approaches

Background:Simiaowan(SMW),a well-known traditional Chinese medicine,has been employed to treat hyperuricemia(HUA)and gout for ***,the bioactive components and underlying mechanisms have not been *** objective of this study was to identify the active components and potential mechanisms of SMW by integrating pharmacological experimentation,serum pharmacochemistry,network pharmacology and molecular ***:HUA rats modelling by high-fat/high-sugar diet and potassium oxonate/adenine oral administration were used to evaluate the pharmacodynamic effects of ***-Q-Exactive-MS/MS was employed to detect the bioactive components present in SMW-containing *** pharmacology and molecular docking were utilized to elucidate the potential targets and underlying ***:SMW effectively ameliorated HUA rats via the inhibition of uric acid(UA)production,promotion of UA excretion,improvement of lipid and glucose metabolic abnormalities,antioxidant,anti-inflammatory and anti-insulin resistance effects.A total of 73 compounds detected in SMW-containing serum were identified as potential active components,with alkaloids,flavonoids,organic acids,and terpenoids emerging as the primary active *** 203 corresponding targets were obtained as SMW anti-HUA/gout targets,which mainly participated in apoptosis,insulin resistance,TNF,PI3K-Akt,HIF-1,NF-κB,MAPK,IL-17 and TLR signaling *** docking indicated that active compounds(***,phellodendrine,quercetin,formononetin,ferulic acid)had superior binding abilities to the key targets(*** carrier family 22 member 12(URAT1),solute carrier family 22 member 6(OAT1),ATP-binding cassette sub-family G member 2(ABCG2),solute carrier family 2,facilitated glucose transporter member 9(GLUT9),xanthine dehydrogenase/oxidase(XDH),transcription factor p65(RELA),toll-like receptor 4(TLR4),prostaglandin G/H synthase 2(PTGS2),caspase-3(CASP3),insulin(INS)).Conclusion:S

Improving slurryability, rheology,and stability of slurry fuel from blending petroleum coke with lignite

Petroleum coke and lignite are two important fossil fuels that have not been widely used in China. Petroleum coke-lignite slurry （PCLS）, a mixture of petro- leum coke, lignite, water, and additives, efficiently utilizes the two materials. In this study, we investigate the effects of the proportion （7） of petroleum coke on slurryability, rheo- logical behavior, stability, and increasing temperature characteristics of PCLSs. The results show that the fixed- viscosity solid concentration （COo） increases with increasing 7. The ~Oo of lignite-water slurry （LWS, ~ = 0） is 46.7 %, compared to 71.3 % for the petroleum coke-water slurry （PCWS, c~ --- i00 %）, while that of PCLS is in between the two values. The rheological behavior of PCLS perfectly fits the power-law model. The PCWS acts as a dilatant fluid. As decreases, the slurry behaves first as an approximate Newtonian fluid, and then turns into a pseudo-plastic fluid that exhibits shear-thinning behavior. With increasing ct, the rigid sedimentation and water separation ratio （WSR） increase, indicating a decrease in the stability of PCLS. When α is 60-70 %, the result is a high-quality slurry fuel for industrial applications, which has high slurryability （ω0 = 57-60 %）, good stability （WSR 〈 2 %）, and superior pseudo-plastic behavior （n = 0.9）.

Organic nanophotocatalysts are promising candidates for solar fuels production,but they still face the challenge of unfavorable geminate recombination due to the limited exciton diffusion ***,we introduce a binary nanophotocatalyst fabricated by blending two polymers,PS-PEG5(PS)and PBT-PEG5(PBT),with matched absorption and emission spectra,enabling a F€orster resonance energy transfer(FRET)process for enhanced *** heterostructure nanophotocatalysts are processed using a facile and scalable flash nanoprecipitation(FNP)technique with precious kinetic control over binary nanoparticle *** resulting nanoparticles exhibit an exceptional photocatalytic hydrogen evolution rate up to 65 mmol g^(-1) h^(-1),2.5 times higher than that single component *** through fluorescence spectra and transient absorption spectra confirm the hetero-energy transfer within the binary nanoparticles,which prolongs the excited-state lifetime and extends the namely“effective exciton diffusion length”.Our finding opens new avenues for designing efficient organic photocatalysts by improving exciton migration.

Effect of h-BN rods on high-temperature properties of alumina-carbon refractory

h-BN rods modified low-carbon alumina-carbon(Al_(2)O_(3)-C)refractories were prepared,and the effect of h-BN rod addition on the high-temperature properties was investigated and compared with commercial h-BN flake,carbon black,and carbon nanotubes *** results demonstrated that Al_(2)O_(3)-C refractories containing h-BN rods exhibited optimal high-temperature service performances,including 25%higher hot modulus of rupture,21.3%higher thermal shock strength residual ratio,20.9%lower in oxidation and 44.3%less in slag corrosion,compared to the counterpart specimens without ***,benefiting from the synergistic enhancement of the rod-like h-BN and in-situ generated SiC whiskers,the high-temperature service performances of h-BN rods containing specimens outperformed counterpart specimens containing commercial h-BN flake,carbon black,and carbon nanotubes,respectively.

Pharmacokinetic studies of multi-bioactive components in rat plasma after oral administration of Xintiantai Ⅰ extract and effects of guide drug borneol on pharmacokinetics

Objective:To investigate the in vivo pharmacokinetic characteristics of 17 bioactive components including ginsenoside Rg1,Rb1,Rd,berberine,epiberberine,jatrorrhizine,palmatine,columbamine,coptisine,evodiamine,dehydroevodiamine,rutaecarpine,limonin,hyperin,curcumin,demethoxycurcumin and bisdemethoxycurcumin in rat plasma after oral administration of Xintiantai I extract powder(XI)and Xintiantai I without guide drug borneol extract powder(XI without borneol),and study the compatibility effects of guide drug borneol on the ***:A UHPLC-MS/MS method was established and fully validated for the comparative pharmacokinetics of 17 bioactive *** pharmacokinetics parameters of 17 bioactive components after oral administration of XI and XI without borneol were calculated by the software of DAS 3.0 and ***:The specificity,linearity,lower limit of quantification(LLOQ),precision,accuracy,extraction recovery rates,matrix effects,and stability of the UHPLC-MS/MS assay were good within the acceptance criteria from FDA *** drug borneol can significantly increase AUC of G-Rd,palmatine,hyperin,curcumin,demethoxycurcumin,bisdemethoxycurcumin and Cmaxof 16 bioactive components except for dehydroevodiamine(P<0.05),decrease Tmaxof G-Rd,berberine,columbamin,coptisine,limonin and MRT of 17 bioactive components in XI group(P<0.05).Conclusion:Guide drug borneol enhanced the absorption of G-Rd,palmatine,hyperin,curcumin,demethoxycurcumin and bisdemethoxycurcumin.

In recent years, biopharmaceuticals have witnessed remarkable advancements, transforming the landscape of therapeutic interventions. Biopharmaceuticals encompassing therapeutics generated through cutting-edge biotechnological methods have shown promising therapeutic outcomes. However, their clinical success hinges significantly on overcoming drug delivery challenges related to stability, intracellular delivery, immunogenicity, and pharmacokinetic properties. Herein, we provide an overview of various marketed macromolecules, including nucleic acids, and immunotherapeutic agents such as cytokines and monoclonal antibodies, as well as other therapeutic peptides/proteins like enzymes, hormones, and coagulation factors. Our primary focus is on elucidating the delivery challenges associated with these macromolecules and highlighting the pivotal role played by drug delivery platforms in the development of currently marketed products, offering valuable insights for both scientific research and the pharmaceutical industry.

Simultaneous immobilization of multiple heavy metal(loid)s in contaminated water and alkaline soil inoculated Fe/Mn oxidizing bacterium

Two strains of Fe/Mn oxidizing bacteria tolerant to high concentrations of multiple heavy metal(loid)s and efficient decontamination for them were *** surface of the bio-Fe/Mn oxides produced by the oxidation of Fe(II)and Mn(II)by Pseudomonas taiwanensis(marked as P4)and Pseudomonas plecoglossicida(marked as G1)contains rich reactive oxygen functional groups,which play critical roles in the removal efficiency and immobilization of heavymetal(loid)s in co-contamination *** isolated strains P4 and G1 can growwell in the following environments:pH 5-9,NaCl 0-4%,and temperature 20-30℃.The removal efficiencies of Fe,Pb,As,Zn,Cd,Cu,and Mn are effective after inoculation of the strains P4 and G1 in the simulated water system(the initial concentrations of heavy metal(loid)were 1 mg/L),approximately reaching 96%,92%,85%,67%,70%,54%and 15%,*** exchangeable and carbonate bound As,Cd,Pb and Cu are more inclined to convert to the Fe-Mn oxide bound fractions in P4 and G1 treated soil,thereby reducing the phytoavailability and bioaccessible of heavy metal(loid)*** research provides alternatives method to treat water and soil containing high concentrations of multi-heavy metal(loid)s.

Sky location of massive black hole binaries in the foreground of Galactic white dwarf binaries

For space-based gravitational wave(GW)detection,the main noise source for massive black hole binaries(MBHBs)is attributed to approximately 107 double white dwarf binaries in the *** a GW source,the amplitude of the detector response,recorded by a space-based gravitational wave detector,exhibits a modulation effect with a year period when observing the source from various orbital *** the adverse conditions mentioned above,where there is a strong foreground noise and annual modulation in the signals,we employed the wavelet transform and the strong-amplitude relevant orbital position search methods,which allows the weak MBHB sources to achieve higher locating *** detail,for two MBHB sources of lower intensity,the precision of luminosity distance,represented by the ratioΔD_(L)/D_(L) at the 95%confidence level,is enhanced by factors of∼*** angular resolutions,denoted byΔΩs,are enhanced by a factor of∼*** improvements increase the number of detectable GW sources,facilitate multi-messenger follow-up observations and provide constraints on the cosmological constant.
