Pharmacokinetic enhancement of oncolytic virus M1 by inhibiting JAK-STAT pathway

Oncolytic viruses(OVs),a group of replication-competent viruses that can selectively infect and kill cancer cells while leaving healthy cells intact,are emerging as promising living anticancer *** traditional drugs composed of non-replicating compounds or biomolecules,the replicative nature of viruses confer unique pharmacokinetic properties that require further *** some pharmacokinetics studies of OVs,mechanistic insights into the connection between OV pharmacokinetics and antitumor efficacy remain ***,we characterized the pharmacokinetic profile of oncolytic virus M1(OVM)in immunocompetent mouse tumor models and identified the JAK-STAT pathway as a key modulator of OVM *** suppressing the JAK-STAT pathway,early OVM pharmacokinetics are ameliorated,leading to enhanced tumor-specific viral accumulation,increased AUC and Cmax,and improved antitumor *** than compromising antitumor immunity after JAK-STAT inhibition,the improved pharmacokinetics of OVM promotes T cell recruitment and activation in the tumor microenvironment,providing an optimal opportunity for the therapeutic outcome of immune checkpoint blockade,such as *** together,this study advances our understanding of the pharmacokinetic-pharmacodynamic relationship in OV therapy.

Mirizzi syndrome is a serious complication of gallstone *** is caused by the impacted stones in the gallbladder neck or cystic *** of the features of Mirizzi syndrome is severe inflammation or dense fibrosis at the Calot’s *** our clinical practice,bile duct,branches of right hepatic artery and right portal vein clinging to gallbladder infundibulum are often observed due to gallbladder infundibulum adhered to right hepatic *** intraoperative damage of branches of right hepatic artery occurs more easily than that of bile duct,all of which are hidden pitfalls for *** resonance cholangiopancreatography(MRCP)and endoscopic retrograde cholangiopancreatography(ERCP)are the preferable tools for the diagnosis of Mirizzi *** cholecystectomy in Mirizzi syndrome is easy to damage branches of right hepatic artery and bile duct due to gallbladder infundibulum adhered to right hepatic *** cholecystectomy is an easy,safe and definitive approach to Mirizzi *** combined with the application of ERCP,a laparoscopic management of Mirizzi syndrome by well-trained surgeons is feasible and *** objective of this review was to highlight its existing problems:(1)low preoperative diagnostic rate,(2)easy to damage bile duct and branches of right hepatic artery,and(3)high concomitant gallbladder ***,the review aimed to discuss the possible therapeutic strategies:(1)to enhance its preoperative recognition by imaging findings,and(2)to avoid potential pitfalls during surgery.

The small brown planthopper(SBPH,Laodelphax striatellus)is a significant rice pest,responsible for transmitting rice stripe virus(RSV)in a persistent and propagative *** is one of the most detrimental rice viruses,causing rice stripe disease,which results in considerable loss of rice grain *** RNA interference and gene knockout techniques have enabled gene downregulation in SBPH,no system currently exists for the overexpression of endogenous or exogenous ***,the development of a protein expression system for SBPH is imperative to serve as a technical foundation for pest control and gene function *** study aimed to construct an expression vector using the promoter of the constitutive-expressed tubulin gene of SBPH,and promoter of human cytomegalovirus(CMV).Fluorescence experiments demonstrated that both tubulin and CMV promoter could drive green fluorescent protein(GFP)expression in SBPH,and could also facilitate the expression of a nucleocapsid protein(NP)-GFP fusion protein containing viral NP with comparable *** expression vector optimization,we have identified that the 3 tandem CMV promoters display a significantly higher promoter activity compared with both the 2 tandem CMV promoters and the single CMV *** addition,the incorporation of Star polycation nanoparticles significantly enhanced the expression efficiency in *** results provide a promising technical platform for investigating gene functions in SBPH.

Generation and regulation of electromagnetic pulses generated by femtosecond lasers interacting with multitargets

Ultrashort and powerful laser interactions with a target generate intense wideband electromagnetic pulses(EMPs).In this study,we report EMPs generated by the interactions between petawatt(30 fs,1.4×10^(20) W/cm^(2))femtosecond(fs)lasers with metal flat,plastic flat,and plastic nanowire-array(NWA)*** analyses are conducted on the EMPs in terms of their spatial distribution,time and frequency domains,radiation energy,and *** results indicate that EMPs from metal targets exhibit larger amplitudes at varying angles than those generated by other types of targets and are enhanced significantly for NWA *** a plastic target holder and increasing the laser focal spot can significantly decrease the radiation energy of the ***,the composite shielding materials indicate an effective shielding effect against *** simulation results show that the NWA targets exert a collimating effect on thermal electrons,which directly affects the distribution of *** study provides guidance for regulating EMPs by controlling the laser focal spot,target parameters,and target rod material and is beneficial for electromagnetic-shielding design.

A new interacting capillary bundle model on the multiphase flow in micropores of tight rocks

Surfactants are widely used in the fracturing fluid to enhance the imbibition and thus the oil recovery rate. However, current numerical models cannot capture the physics behind capillary imbibition during the wettability alteration by surfactants. Although the interacting capillary bundle(ICB) model shows potential in characterizing imbibition rates in different pores during wettability alteration, the existing ICB models neglect the influence of wettability and viscosity ratio on the imbibition behavior, making it difficult to accurately describe the oil-water imbibition behavior within the porous media. In this work,a new ICB mathematical model is established by introducing pressure balance without assuming the position of the leading front to comprehensively describe the imbibition behavior in a porous medium under different conditions, including gas-liquid spontaneous imbibition and oil-water *** the pore size distribution of a tight rock is known, this new model can predict the changes of water saturation during the displacement process in the tight rock, and also determine the imbibition rate in pores of different sizes. The water saturation profiles obtained from the new model are validated against the waterflooding simulation results from the CMG, while the imbibition rates calculated by the model are validated against the experimental observations of gas-liquid spontaneous imbibition. The good match above indicates the newly proposed model can show the water saturation profile at a macroscopic scale while capture the underlying physics of the multiphase flow in a porous medium at a microscopic scale. Simulation results obtained from this model indicate that both wettability and viscosity ratio can affect the sequence of fluid imbibition into pores of different sizes during the multiphase flow, where less-viscous wetting fluid is preferentially imbibed into larger pores while more-viscous wetting fluid tends to be imbibed into smaller pores. Influence of meteorological factors on open biomass burning at a background site in Northeast China

Biomass burning(BB)is a very important emission source that significantly adversely impacts regional air *** produces a large number of primary organic aerosol(POA)and black carbon(BC).Besides,BB also provides many precursors for secondary organic aerosol(SOA)*** this work,the ratio of levoglucosan(LG)to organic carbon(OC)and the fire hotspots map was used to identify the open biomass burning(OBB)events,which occurred in two representative episodes,October 13 to November 30,2020,and April1 to April 30,*** ratio of organic aerosol(OA)to reconstructed PM_(2.5)concentration(PM_(2.5)^(*))increased with the increase of LG/*** LG/OC ratio is higher than 0.03,the highest OA/PM_(2.5)^(*)ratio can reach 80%,which means the contribution of OBB to OA is *** to the ratio of LG to K^(+),LG to mannosan(MN)and the regional characteristics of Longfengshan,it can be determined that the crop residuals are the main *** occurrence of OBB coincides with farmers’preferred choices,i.e.,burning biomass in“bright weather”.The“bright weather”refers to the meteorological conditions with high temperature,low humidity,and without *** factors indirectly affect regional biomass combustion pollution by influencing farmers’active choices.

Adhesive Contact of Elastic Solids with Self-Affine Fractal Rough Surfaces

The elastic adhesive contact of self-affine fractal rough surfaces against a rigid flat is simulated using the finite element method. An array of nonlinear springs, of which the force-separation law obeys the Lennard–Jones potential, is introduced to account for the interfacial adhesion. For fractal rough surfaces, the interfacial interaction is generally attractive for large mean gaps, but turns repulsive as the gap continuously shrinks. The interfacial interactions at the turning point corresponding to the spontaneous contact are shown for various surfaces. For relatively smooth surfaces, the probability density distributions of repulsion and attraction are nearly symmetric. However, for rougher surfaces, the simulation results suggest a uniform distribution for attraction but a monotonously decreasing distribution with a long tail for repulsion. The pull-off force rises with increasing ratio of the work of adhesion to the equilibrium distance, whereas decreases for solids with a higher elastic modulus and a larger surface roughness. The current study will be helpful for understanding the adhesion of various types of rough solids.
