Reliability of the pronator quadratus fat pad sign to predict the severity of distal radius fractures

AIM To evaluate the reliability of pronator quadratus fat pad sign to detect distal radius fracture and to predict its *** Retrospectively we identified 89 consecutive patients(41 female, mean age 49 ± 18 years) who had X-ray(CR) and computed tomography(CT) within 24 h following distal forearm trauma. Thickness of pronator quadratus fat pad complex(PQC) was measured using lateral views(CR) and sagittal reconstructions(CT). Pearson's test was used to determine the correlation of the PQC thickness in CR and CT. A positive pronator quadratus sign(PQS) was defined as a PQC > 8.0 mm(female) or > 9.0 mm(male). Frykman classification was utilized to assess the severity of *** Forty-four/89 patients(49%) had a distal radius fracture(Frykman Ⅰ n = 3, Ⅱ n = 0, Ⅲ n = 10, Ⅳ n = 5, Ⅴ n = 2, Ⅵ n = 2, Ⅶ n = 9, Ⅷ n = 13). Mean thickness of the PQC thickness can reliably be measured on X-ray views and was 7.5 ± 2.8 mm in lateral views(CR), respectively 9.4 ± 3.0 mm in sagittal reconstructions(CT), resulting in a significant correlation coefficientof 0.795. A positive PQS at CR was present in 21/44 patients(48%) with distal radius fracture and in 2/45 patients(4%) without distal radius fracture, resulting in a specificity of 96% and a sensitivity of 48% for the detection of distal radius fractures. There was no correlation between thickness of the PQC and severity of distal radius *** A positive PQS shows high specificity but low sensitivity for detection of distal radius fractures. The PQC thickness cannot predict the severity of distal radius fractures.

Proximate composition and seed lipid components of “kabuli”-type chickpea (<i>Cicer arietinum</i>l.) from Argentina

Chickpea is an important pulse crop with a wide range of potential nutritional benefits because of its chemical composition. The purpose of the current work was to provide the chemical composition of “kabuli”-type chickpea (Cicer arietinum l.) developed in Argentina for nutritional purpose. Protein, oil and ash contents, fatty acid, tocopherol and mineral element compositions were studied. Among the studied genotypes, protein content ranged from 18.46 to 24.46 g/100g, oil content ranged from 5.68 to 9.01 g/100g and ash from 3.55 to 4.46 g/100g. linoleic, oleic and palmitic acids were the most abundant fatty acids. The average oleic-to-linoleic ratio was 0.62 and average iodine value was 117.82. Tocopherols, well-established natural antioxidants, were found in chickpea seeds in relatively similar amounts across all genotypes. Mineral element analysis showed that chickpea was rich in macronutrients such as K, P, Mg and Ca. The nutritional composition of chickpea genotypes developed and grown in Argentina provides useful information for breeding programs, food marketing and consumers and establishes chickpea as component of a balanced human diet.

Development and Validation of Two Methods to Quantify Volatile Acids (C2-C6) by GC/FID: Headspace (Automatic and Manual) and liquid-liquid Extraction (llE)

The concentration of the volatile fatty acids (VFA) is an important indicator of the status of an-aerobic processes, but most of the existing methods require sample pretreatment and are labor-intensive. It was developed and validated a rapid Gas Chromatographic (GC) method to quantify seven VFA (acetic, propionic, isobutyric, butyric, isovaleric, valeric and caproic), acetone, methanol, ethanol and n-butanol by headspace (automatic and manual) and liquid-liquid extraction (llE) with diethyl ether (only VFA). The determination was made in a Shimadzu Gas Chromatograph equipped with a Flame Ionization Detector (GC/FID), a headspace auto-sampler and an HP-INNOWAX column. Isobutanol and crotonic acid were utilized as internal standards (IS). The validation parameters evaluated were: precision (coefficient of variation—C.V.% for the retention times, from 0.02 to 0.87), linearity (R2 = 0.9291 - 0.9997), limits of detection (from 3.97 to 36.45 mg·l﹣1) and instrumental precision (from 0.01 to 0.53), which provide evidence that the methods are adequate to determine these analytes in samples from anaerobic reactors and from the environment.

Regulatory T(Treg) cells characterized by the transcription factor FoxP3 have a fundamental role in the establishment and maintenance of peripheral tolerance, and have transcriptional and functional phenotypes that distinguish them from conventional CD4+ T cells(Tconv). There have been reports that these two CD4+ T cell subsets differ in the signaling downstream of T cell receptor-triggered activation, with different requirements for some signaling factors intermediates and lower responsiveness in Treg cells. Seeking a more comprehensive view, we found that Treg cells have a broadly dampened activation of several pathways and signaling nodes, with low phosphorylation of CD3ζ, SlP76, Erk1/2, AKT, or S6 and lower calcium influx, upon TCR-mediated activation. Examination of STAT responses to cytokines highlighted striking variations in Treg vs Tconv responses to interferons, Il2 or Il6, but no general Treg signaling defect. Treg/Tconv differences in responses were not due to different TCR levels, but much of the difference could be attributed to lower responsiveness of cells with a CD44 or CD62 l cells, which form a greater proportion of the Treg pool. Testing putative candidate regulators, the Treg/Tconv differential could not be explained by Treg over-expression of the signaling modulator CD5, the co-inhibitors PD- and CTlA4, or the regulatory phosphatase DUSP4. However, gene expression profiling in Dusp4-deficient mice showed that DUSP4 enhances the expression of a segment of the canonical Treg transcriptional signature, a module which overlaps in part with the TCR-dependent Treg gene set. Taken together, our findings show Treg cell signaling to be tuned down, plausibly because of their intrinsically higher reactivity to self, but comparatively more attuned to cytokines or other inter-cellular signals.

Fluoxetine induces cytotoxic endoplasmic reticulum stress and autophagy in triple negative breast cancer

AIM: To investigate the mechanism of action of lipophilic antidepressant fluoxetine(FlX) in representative molecular subtypes of breast ***: The anti-proliferative effects and mechanistic action of FlX in triple-negative(SUM149PT) and luminal(T47D and Au565) cancer cells and nontransformed MCF10 A were investigated. Reverse phase protein microarray(RPPM) was performed with and without 10 μmol/l FlX for 24 and 48 h to determine which proteins are significantly changed. Viability and cell cycle analysis were also performed to determine drug effects on cell growth. Western blotting was used to confirm the change in protein expression examined by RPPM or pursue other signaling proteins. RESUlTS: The FlX-induced cell growth inhibition in all cell lines was concentration- and time-dependent but less pronounced in early passage MCF10 A. In comparison to the other lines,cell growth reduction in SUM149 PT coincided with significant induction of endoplasmic reticulum(ER) stress and autophagy after 24 and 48 h of 10 μmol/l FlX,resulting in decreased translation of proteins along the receptor tyrosine kinase/Akt/mammalian target of rapamycin pathways. The increase in autophagy marker,cleaved microtubule-associated protein 1 light chain 3,in SUM149 PT after 24 h of FlX was likely due to increased metabolic demands of rapidly dividing cells and ER stress. Consequently,the unfolded protein response mediated by double-stranded RNA-dependent protein kinase-like ER kinase resulted in inhibition of protein synthesis,growth arrest at the G1 phase,autophagy,and caspase-7-mediated cell ***: Our study suggests a new role for FlX as an inducer of ER stress and autophagy,resulting in death of aggressive triple negative breast cancer SUM149 PT.

Seasonal tropical cyclone(TC)forecasting has evolved substantially since its commencement in the early ***,present operational seasonal TC forecasting services still do not meet the requirements of society and stakeholders:current operational products are mainly basin-scale information,while more detailed sub-basin scale information such as potential risks of TC landfall is anticipated for decision *** fill this gap and make the TC science and services move forward,this paper reviews recent research and development in seasonal tropical cyclone(TC)*** particular,this paper features new research topics on seasonal TC predictability in neutral conditions of El Ni˜no–Southern Oscillation(ENSO),emerging forecasting techniques of seasonal TC activity including Machine learning/Artificial Intelligence,and multi-annual TC *** also review the skill of forecast systems at predicting landfalling statistics for certain regions of the North Atlantic,Western North Pacific and South Indian oceans and discuss the gap that remains between current products and potential user's *** knowledge and advanced forecasting techniques are expected to further enhance the capability of seasonal TC forecasting and lead to more actionable and fit-for-purpose products.

Alzheimer’s disease (AD) is a neurodegenerative disease characterized by the progressive loss of cognitive functions in affected individuals. Brain tissue pathology is associated with the formation of senile plaques which result from the over-production of amyloid β (Aβ), due to the cleavage of a membrane bound glycoprotein. It is unclear what causes AD and its associated pathologies, but age and genetic predisposition play an import role in the likelihood of disease development. Studies have shown that the reactivation of latent herpes simplex virus 1 (HSV-1) infection can lead to the neuropathy of acute herpes simplex encephalitis (HSE), which causes similar symptoms to AD. HSV-1 infection is a known risk factor for the development of AD, but no study has determined a definitive causal relationship. Using the Qiagen Ingenuity Pathway Analysis (IPA) tool, the inhibitory relationship between therapeutics for AD and HSV-1 were explored. Thirteen drugs developed to decrease Aβ buildup in AD and 32 drugs that act as HSV antivirals were retrieved from the data in the Qiagen Knowledge Base. These drugs were analyzed displayed as two separate networks. While many promising Aβ aggregation-targeting drugs have been discontinued due to lack of efficacy, HSV drugs could serve as potential therapeutics for those with AD.
