sexually reproducing animals require an orchestrated communication between spermatozoa and the egg to generate a new individual. Capacitation, a maturational complex phenomenon that occurs in the female reproductive tract, renders spermatozoa capable of binding and fusing with the oocyte, and it is a requirement for mammalian fertilization. Capacitation encompasses plasma membrane reorganization, ion permeability regulation, cholesterol loss and changes in the phosphorylation state of many proteins. Novel tools to study sperm ion channels, image intracellular ionic changes and proteins with better spatial and temporal resolution, are unraveling how modifications in sperm ion transport and phosphorylation states lead to capacitation. Recent evidence indicates that two parallel pathways regulate phosphorylation events leading to capacitation, one of them requiring activation of protein kinase A and the second one involving inactivation of ser/thr phosphatases. This review examines the involvement of ion transporters and phosphorylation signaling processes needed for spermatozoa to achieve capacitation. Understanding the molecular mechanisms leading to fertilization is central for societies to deal with rising male infertility rates, to develop safe male gamete-based contraceptives and to preserve biodiversity through better assisted fertilization strategies.

Effects of absolute fasting on reproduction and survival of the invasive apple snail Pomacea canaliculata in its native range

A south American freshwater gastropod, the apple snail Pomacea canaliculata, has become a driver of ecosystemic changes in wetlands and an important rice pest after its introduction to various parts of the world, mainly Asia. The objective of this study was to study the effect of an abrupt interruption in food availability in the short term （up to 4 weeks） and long term （up to 8 months） on survival and re- productive activity. The main results indicate that short-term fasting mainly affects the survival of males, but only when they are raised together with females, probably due to a greater mate-searching activity that increases mortality in the individuals with lower reserves. The number of copulating snails or egg-laying females shows an abrupt drop when fasting and a rapid recovery after the food supply is restored. The strategy of discontinuing reproductive activity prioritizes energy conservation for the survival of the females. Interpopulation variation in resistance to starvation was observed in adults, which can be explained to some extent by the food availability that they experienced in their natural environment. No interpopulational differences in survival were seen in hatchlings. The mean maximum values of survival under starvation were 52.6 days in hatchlings and the 3.3% of adults survive over than 200 days, which may be a relevant trait in dispersal and establishment in new habitats.

Identification of Candidate Genes Related to Polyploidy and/or Apomixis in <i>Eragrostis curvula</i>

This work was aimed at identifying genes that show altered expression profiles in response to changes in ploidy and/or reproductive mode (from sexual to apomictic) in the African grass Eragrostis curvula. A differential display analysis was performed on leaf and flower transcriptomes from a series of genetically related euploid plants, including tetraploid apomictic, diploid sexual, and tetraploid sexual plants. More than 100 primer combinations were used to generate 11,864 total markers, yielding 1293 differential bands. Of these bands, 11.84% to 6.74% were related to ploidy and 0.71% to 2.17% to the reproductive mode, depending on the tissue. A small percentage of bands showed similar expressions between the tetraploid apomictic and the diploid sexual plants. Expression-based similarity dendrograms were constructed. Our data suggested that ploidy is more decisive than tissue type in defining the transcriptome structure. Out of 102 fragments sequenced, 50 showed strong homology to known genes. The differentially expressed genes were mapped in silico onto maize chromosomes. several candidates mapped within the linkage group syntenic to the Tripsacum dactyloides diplospory-governing region. The evidence indicates that expression of genes located around the diplospory-associated region may be strongly influenced by ploidy and may be silenced in the apomictic genotype. These findings are discussed in the context of diplospory molecular control and its connection with ploidy.

Boron （B） is an essential nutrient for normal growth of higher plants, and B availability in soil and irrigation water is an important determinant of agricultural production. To date, a primordial function of B is undoubtedly its structural role in the ceil wall; however, there is increasing evidence for a possible role of B in other processes such as the maintenance of plasma membrane function and several metabolic pathways. In recent years, the knowledge of the molecular basis of B deficiency and toxicity responses in plants has advanced greatly. The aim of this review is to provide an update on recent findings related to these topics, which can contribute to a better understanding of the role of B in plants.

Dealing with a hyper-successful neighbor: effects of the invasive apple snail Pomacea canaliculata on exotic and native snails in south America

Pomacea canaliculata is a successful invader and also a competitor and predator of other snails and may play a key role in structuring freshwater snail communities both in its native and invaded range. In the present study we evaluated the contact and distant effects of P. canaliculata in its native range on exotic (Melanoides tuberculata and Physa acuta) and native snails (Heleobia parchappii, Biomphalaria peregrina, and Chilina parchappii). Habitat use was affected in P. acuta, H. parchappii, and B. peregrina by contact effects of P. canaliculata, whereas survival was only affected in P. acuta through combined con tact and dista nt effects. Fee undity was reduced in P. acuta and B. peregrina by combined con tact and distant effects;evidence of egg mass predation was also observed in both species. Melanoides tuberculata was not affected at all by P. canaliculata. The snail species with higher withdrawal resp on ses to con tacts with P. canaliculata were those that suffered less mortality by corporal contact, whereas snails with high crawling away responses suffered from higher mortality. The effects of P. canaliculata seem to be highly negative to small non operculate sn ails that lay gelati nous egg masses, whereas large operculate ovoviviparous snails are not affected in their survival and reproducti on. This apple snail may exert biotic resista nee agai nst P. acuta but could favor the establishme nt of M. tuberculata and other function ally similar species in new habitats in south America.

The Immune system and the Role of Inflammation in Perinatal Depression

Major depression during pregnancy is a common psychiatric disorder that arises from a complex and mul- tifactorial etiology. Psychosocial stress, sex, hormones, and genetic vulnerability increase the risk for triggering mood disorders. Microglia and toll-like receptor 4 play a crucial role in triggering wide and varied stress-induced responses mediated through activation of the inflammasome; this leads to the secretion of inflammatory cytokines, increased serotonin metabolism, and reduction of neurotransmitter availability along with hypothalamic-pituitary-adrenal axis hyperactivity. Dysregulation of this intricate neu- roimmune communication network during pregnancy modifies the maternal milieu, enhancing the emergence of depressive symptoms and negative obstetric and neu- ropsychiatric outcomes. Although several studies have clearly demonstrated the role of the innate immune system in major depression, it is still unclear how the placenta, the brain, and the monoaminergic and neuroendocrine systems interact during perinatal depression. Thus, in the present review we describe the cellular and molecular interactions between these systems in major depression during preg- nancy, proposing that the same stress-related mechanisms involved in the activation of the NLRP3 inflammasome in microglia and peripheral myeloid cells in depressed patients operate in a similar fashion in the neuroimmune placenta during perinatal depression. Thus, activation of Toll-like receptor 2 and 4 signaling and the NLRP3 inflammasome in placental immune cells may promote a shift of the Thl/Th2 bias towards a predominant Thl/Thl7 inflammatory response, associated with increased secretion of pro-inflammatory cytokines, among other secreted autocrine and paracrine mediators, which play a crucial role in triggering and/or exacerbating depressive symptoms during pregnancy.
