Non-invasive sensing techniques to phenotype multiple apple tree architectures

Tree fruit architecture results from combination of the training system and pruning and thinning processes across multiple growth and development ***,the tree fruit architecture contributes to the light interception and improves tree growth,fruit quality,and fruit yield,in addition to easing the process of orchard management and *** tree architectural traits are measured manually by researchers or growers,which is labor-intensive and *** this study,the remote sensing techniques were evaluated to phenotype critical architectural traits with the final goal to assist tree fruit breeders,physiologists and growers in collecting architectural traits efficiently and in a standardized *** this,a consumer-grade red–green–blue(RGB)camera was used to collect apple tree side-images,while an unmanned aerial vehicle(UAV)integrated RGB camera was programmed to image tree canopy at 15 m above ground level to evaluate multiple tree fruit *** sensing data were compared to ground reference data associated with tree orchard blocks within three training systems(spindle,V-trellis,Biaxis),two rootstocks(‘WA 38 trees grafted on G41 and M9-Nic29)and two pruning methods(referred as bending and click pruning).The data were processed to extract architectural features from ground-based 2D images and UAV-based 3D digital surface *** traits extracted from sensing data included box-counting fractal dimension(DBs),middle branch angle,number of branches,trunk basal diameter,and tree row volume(TRV).The results from ground-based sensing data indicated that there was a significant(Ps between spindle and V-trellis training systems,and correlations between DBs with tree height(r=0.79)and total fruit yield per unit area(r=0.74)were significant(Ps between average or total TRV and ground reference data,such as tree height and total fruit yield per unit area,were significant(P<0.05).With the r

Effect of different exogenous fatty acids on the cytosolic triacylglycerol content in bovine mammary cells

The objective of this study was to determine how cytosolic triacylglycerols(TAG) are stored in mammary cells and whether this depends on the individual chemical configuration of fatty acids(FA). This objective was accomplished by addition of different FA to a FA-free medium used to culture mammary alveolar cells-large T antigen cells(MAC-T). Treatments consisted of adding FA(palmitate, stearate, oleate, linoleate,rumenic acid [CLA], elaidate and vaccinate) solutions to the medium at 100, 200, 300 and 400 mmol/L concentrations for a 24-h incubation period. At the end of each incubation period, cytosolic TAG, DNA and protein contents were measured. Palmitate, vaccenate, linoleate and CLA increased(P solic TAG(μg/mg protein). Palmitate and CLA increased(P solic TAG adjusted for DNA content. Overall, effects on cytosolic TAG accumulation depended on individual FA structure(chain length, degree of saturation, and number and orientation of FA double bonds). In addition, the long-chain FA used in this study did not have a detrimental effect on MAC-T cells as indicated by cytosolic protein and DNA contents reflecting their biological role in lipid accumulation.

Investigating lodging in spearmint with overhead sprinklers compared to drag hoses using entropy values from low altitude RGB-imagery

Lodging occurs when the crop canopy is too heavy for the strength of the stem and it fallsover onto the ground. This decreases crop yield and quality, and it makes harvest difficult.A research experiment was set up in a spearmint field on a center pivot with mid elevationspray application (MEsA) overhead sprinklers, where the water was applied from a “midelevation” of 2 m above the ground level (AGL), and low elevation precision application(LEPA) sprinklers, where the water was emitted directly onto the soil surface through draghoses without wetting the crop canopy. Every-other span of this full-size center pivot wasconfigured with MEsA and LEPA sprinklers alternatively. In 2018, imagery was collectedwith an unmanned aerial vehicle (UAV) from a cross section of this field. In 2019, a crosssection was again collected, but in addition UAV imagery was collected from marked lodgedand un-lodged areas of the field to validate the lodging detection method. These UAV-basedimagery data were captured with a ground sample distance (GsD) of 0.03 m. This researchintroduces using the texture feature, which is based on image entropy, was used to evaluate the degree of lodging. The results from 2018 showed that the average entropy of thegrayscale image from LEPA (5.5 (mean) ± 0.27 (standard deviation)) was significantly(P sA. Also, the entropy valueextracted from the images in 2019 from the marked un-lodged locations were significantlyhigher compared to that of the lodged areas. Overall, the LEPA irrigation treatment was significantly less lodged compared to MEsA. Moreover, the entropy value, or texture feature, isa viable method for estimating lodging using low altitude RGB imagery.

Plant peroxisomes have the capacity to gen erate different reactive oxygen and nitrogen species(ROs and RNs),such as H2O2,superoxide radical(O2^·-),nitric oxide and peroxynitrite(ONOO^-).These organelles have an active nitrooxidative metabolism which can be exacerbated by adverse stress *** sulfide(H2s)is a new signaling gasotransmitter which can mediate the posttranslational modification(PTM)*** used Arabidopsis thaliana transgenic seedlings expressing cyan fluorescent protein(CFP)fused to a canonical peroxisome targeting signal 1(PTs1)to visualize peroxisomes in living cells,as well as a specific fluorescent probe which showed that peroxisomes contain H2s.H2s was also detected in chloroplasts under glyphosate-induced oxidative stress *** enzyme activities,including catalase,photorespiratory H2O2-generating glycolate oxidase(GOX)and hydroxypyruvate reductase(HPR),were assayed in vitro with a H2s *** line with the persulfidation of this enzyme,catalase activity declined significantly in the presence of the H2s *** corroborate the inhibitory effect of H2s on catalase activity,we also assayed pure catalase from bovine liver and pepper fruit-enriched samples,in which catalase activity was *** together,these data provide evidenee of the presence of H2s in plant peroxisomes which appears to regulate catalase activity and,consequently,the peroxisomal H2O2 metabolism.

Diet switch and omega-3 hydroxy-fatty acids display differential hepatoprotective effects in an obesity/nonalcoholic fatty liver disease model in mice

AIM To study the effect of 18-hydroxy-eicosapentaenoic acid(18-HEPE) and 17-hydroxy-docosahexaenoic acid(17-HDHA) in a murine model of obesity/nonalcoholic fatty liver *** C57 BL/6 mice were fed with standard chow diet(CD) or high-fat, fructose-enriched diet(HFD) for 16 wk. Then, three groups were treated for 14 d with either, diet switch(HFD for CD), 18-HEPE, or 17-HDHA. Weightand fasting glucose were recorded on a weekly basis. Insulin tolerance test was performed at the end of treatment. Histological analysis(HE and Masson's trichrome stain) and determination of serum insulin, glucagon, glucagon-like peptide 1(GLP-1), glucose-dependent insulinotropic polypeptide, adiponectin and resistin were carried out as well as liver proteins by western *** Mice treated with hydroxy-fatty acids 18-HEPE and 17-HDHA displayed no weight loss or improved insulin sensitivity. However, these mice groups showed a significant amelioration on serum GLP-1, adiponectin and resistin levels. Also, a significant reduction on inflammatory infiltrate was observed at both portal and lobular zones. Furthermore, up-regulation of PPARα/γ protein levels was observed in liver tissue and it was associated with decreased levels of NF-κB also determined by western blot analysis. On the other hand, diet switch regimen resulted in a marked improvement in most parameters including: weight loss, increased insulin sensitivity, decreased steatosis, restored levels of insulin, glucagon, leptin, adiponectin and resistin. However, no significant changes were observed regarding inflammatory infiltrate in this last *** 18-HEPE and 17-HDHA differentially exert hepatoprotective effects through up-regulation of nuclear receptors PPARα/γ and amelioration of serum adipokines profile.

META-R(multi-environment trial analysis in R)is a suite of R scripts linked by a graphical user interface(GUI)designed in Java *** objective of META-R is to accurately analyze multi-environment plant breeding trials(METs)by fitting mixed and fixed linear models from experimental designs such as the randomized complete block design(RCBD)and the alpha-lattice/lattice ***-R simultaneously estimates the best linear and unbiased estimators(BLUEs)and the best linear and unbiased predictors(BLUPs).Additionally,it computes the variance-covariance parameters,as well as some statistical and genetic parameters such as the least significant difference(LsD)at 5%significance,the coefficient of variation in percentage(CV),the genetic variance,and the broad-sense *** parameters are very important in the selection of top performing genotypes in plant ***-R also computes the phenotypic and genetic correlations among environments and between traits,as well as their statistical *** genetic correlations between environments or traits can be visualized in a biplot graph or a tree diagram(dendrogram).Genetic correlations are very important for identifying environments with similar behavior or making indirect selection and identifying the most highly associated ***-R performs multi-environment analyses by using the residual maximum likelihood(REML)method;these analyses can be done by environment,across environments by grouping factors(stress conditions,nitrogen content,etc.)and across environments;the analyses across environments can be done with a pre-defined degree of heritability.
