Synthesis and Evaluation of Folate-Conjugated Phenanthraquinones for Tumor-Targeted Oxidative Chemotherapy

Almost all cells are easily killed by exposure to potent oxidants. Indeed, major pathogen defense mechanisms in both animal and plant kingdoms involve production of an oxidative burst, where host defense cells show an invading pathogen with reactive oxygen species (ROS). Although cancer cells can be similarly killed by ROS, development of oxidant-producing chemotherapies has been limited by their inherent nonspecificity and potential toxicity to healthy cells. In this paper, we describe the targeting of an ROS-generating molecule selectively to tumor cells using folate as the tumor-targeting ligand. For this purpose, we exploit the ability of 9,10-phenanthraquinone (PHQ) to enhance the continuous generation of H2O2 in the presence of ascorbic acid to establish a con-stitutive source of ROS within the tumor mass. We report here that incubation of folate receptor-expressing KB cells in culture with folate-PHQ plus ascorbate results in the death of the cancer cells with an IC50 of ~10 nM (folate-PHQ). We also demonstrate that a cleavable spacer linking folate to PHQ is significantly inferior to a noncleavable spacer, in contrast to most other folate-targeted therapeutic agents. Unfortunately, no evidence for folate-PHQ mediated tumor regression in murine tumor models is obtained, suggesting that unanticipated impediments to generation of cytotoxic quantities of ROS in vivo are encountered. Possible mechanisms and potential solutions to these unanticipated results are offered.

Magnetism of Cr Impurities Embedded in Cu fcc Matrix： Density Functional Calculations

We present extensive first principles density functional theory (DFT) calculations dedicated to analyze the magnetic properties of Crn (n = 2 and 3) impurities embedded in a Cu fcc matrix. We consider several dimers and trimers having different interatomic distances. In all cases the Cr atoms have been embedded as substitutional impurities in the metallic network. For the case of the dimers we have considered two magnetic configurations: Ferromagnetic (Antiferromagnetic) when the atomic magnetic moments are parallel (antiparallel) each other, for this case we have found that the lowest-energy state corresponds to the antiferromagnetic coupling between Cr atoms whose interatomic distance is a√2. In the case of the Cr trimers we have considered three or four magnetic configurations, depending on the Cr impurity geometry, for this case we have found that the lowest-energy state corresponds to the antiferromagnetic Cr trimer AF (↑,↓,↓) forming an isosceles right angle triangle whose hypotenuse and legs length are a√2 and a, respectively, with magnetic moments c (2) = 3.45/-3.32/3.45 μB.

AIM To investigate any changing trends in the etiologies of hepatocellular carcinoma(HCC) in Argentina during the last years. METHODS A longitudinal cohort study was conducted by 14 regional hospitals starting in 2009 through 2016. All adult patients with newly diagnosed HCC either with pathology or imaging criteria were included. Patients were classified as presenting non-alcoholic fatty liver disease(NAFLD) either by histology or clinically, provided that all other etiologies of liver disease were ruled out, fatty liver was present on abdominal ultrasound and alcohol consumption was excluded. Complete follow-up was assessed in all included subjects since the date of HCC diagnosis until death or last medical *** A total of 708 consecutive adults with HCC were included. Six out of 14 hospitals were liver transplant centers(n = 484). The prevalence of diabetes mellitus was 27.7%. Overall, HCV was the main cause of liver disease related with HCC(37%) including cirrhotic and non-cirrhotic patients, followed by alcoholic liver disease 20.8%, NAFLD 11.4%, cryptogenic 9.6%, HBV 5.4% infection, cholestatic disease and autoimmune hepatitis 2.2%, and other causes 9.9%. A 6-fold increase in the percentage corresponding to NAFLDHCC was detected when the starting year, i.e., 2009 was compared to the last one, i.e., 2015(4.3% vs 25.6%; P < 0.0001). Accordingly, a higher prevalence of diabetes mellitus was present in NAFLD-HCC group 61.7% when compared to other than NAFLD-HCC 23.3%(P < 0.0001). Lower median AFP values at HCC diagnosis were observed between NAFLD-HCC and non-NAFLD groups(6.6 ng/m L vs 26 ng/m L; P = 0.02). Introduction: Post-hospital syndrome (PHS) is defined as a period of vulnerability during the first 30 days after a patient is discharged from hospital, in which multiple factors come into play. Hyponatremia is the most frequent hydroelectrolytic disorder in hospitalized patients and may be related to the appearance of PHS. Objective: The objective is to estimate the prevalence of PHS that is assessed as the rate of readmissions in the first 30 days after discharge, in patients with hyponatremia. Material and Methods: It is a descriptive observational study of patients with hyponatremia who were discharged from 1 September 2010 to 2 February 2020 at the Internal Medicine Service of the Hospital University of San juan (Alicante, Spain). Results: Of the 25 included patients, 5 (20%) were readmitted within a month of discharge, after a mean of 11.4 days (standard deviation [SD] 5.1). The overall mortality of the study was 20% (n = 5), with one case of death in the first 30 days post-hospitalization (4%). In 12 patients (48%) the origin of the hyponatremia was undetermined. The most frequently recorded etiology for the condition was pharmacological (n = 7, 28%), and there was pronounced variability in its clinical and laboratory study. The most widely used corrective measure was drug withdrawal, in 16 patients (64%). Water intake restriction was the most common treatment after discharge (5 patients, 20%), followed by urea (2 patients, 8%), while tolvaptan was not used. Conclusion: Hyponatremia may be the cause of PHS, which could increase the rate of early readmission. Hyponatremia is an underdiagnosed and undertreated entity, so it is necessary to apply an appropriate system to optimize its management and, in future studies, to assess its impact on PHS.

高温环境中运动会引起体温调节和其它生理压力,继而可导致耐力运动能力的损害。本共识性声明的目的是提供最新的建议以使热环境中体育活动时的运动能力最优化。可用于降低热应激压力和优化运动能力的最重要干预方式是热习服,其应包括1~2周以上反复的运动—高温环境暴露。此外,运动员应在正常水合状态下开始比赛和训练,并将运动中的脱水最小化。随着商用降温系统（如降温背心）的发展,在高温环境中训练或比赛前,运动员可以采取降温策略来促进热的散发或提高蓄热能力。而且,赛事组织者应该设计大面积的遮阳区域,并提供降温和补水设施,按照最小化运动员的健康风险来安排赛事,尤其是在大众参与的赛事中及一年之中炎热天气开始之初。以最近的2008年奥运会和2014年国际足联世界杯为例,当比赛在高温环境中举行时,赛事主管机构应考虑在比赛中或比赛之间允许额外的（或更长的）恢复期以提供补水和降温的时机。

Dear Editor Human pluripotent stem cells including human embryonic stem cells （ESCs） and induced pluripotent stem cells （iPSCs） are cells displaying abilities of unlimited self-renewal and differentiation into any somatic cell type. These unique properties make them increasingly attractive for novel applications in disease modeling, drug discovery, and cell therapy （Buganim et al., 2014; Liu et al., 2011; Liu et al., 2012; sanchez Alvarado and Yamanaka, 2014）. Moreover, iPSCs hold great potential for personalized cell therapy as they avoid some of the ethical concerns as well as the immunological rejection issues ascribed to ESCs.

CdS, CdSe and PbS films were grown onto glass by chemical bath deposition (CBD). Different constant deposition temperatures (Td) were employed in the range 0-80 °C with subintervals of 10 °C. Concentration of reagents, pH, and stirring were maintained constants. A microcrystalline mixture CdCO3 and CdS were grown. At intermediates Td values a mixture of both materials present. i. e., For Td? 40 °C, the intensity of the main of X-ray Diffraction (XRD) CdS peak is abruptly reduced. By scanning electronic microscopy (SEM) with Td = 80 °C, concentration atomic: Cd = 49.10, S = 51.90, for Td? 70 °C, increase of carbon and oxygen. The CdSe, average grain size (GS) increased monotonic with Td, reaching saturation of 70 °C, with GS values of 5-16 nm. At low Td values (0-20 °C), the structural phase was found, and the high Td = 70 °C on material 5-16 nm was appreciated transition to phase: Wurzite (W)? Zinbclende (ZB) and phases mixture: W/ZB, grain size (GS) 9-27 nm. By (XRD), PbS grow in W phase at all films, for Td = 60 °C, was observed preferential orientation in plane [111] with (GS) of 19-26 nm also a Gibbs free energy calculation is included.
