Background: Innate immune responses induced by in vitro stimulation of primary mammary epithelial cells(MEC) using Gram-negative lipopolysaccharide(lPS) and Gram-positive lipoteichoic acid(lTA) bacterial cell wall components are well-characterized in bovine species. The objective of the current study was to characterize the downstream regulation of the inflammatory response induced by Toll-like receptors in primary goat MEC(pgMEC). We performed quantitative real-time RT-PCR(qPCR) to measure mRNA levels of 9 genes involved in transcriptional regulation or antibacterial activity: Toll-like receptor 2(TlR2), Toll-like receptor 4(TlR4), prostaglandin-endoperoxide synthase 2(PTGS2), interferon induced protein with tetratricopeptide repeats 3(IFIT3), interferon regulatory factor 3(IRF3), myeloid differentiation primary response 88(MYD88), nuclear factor of kappa light polypeptide gene enhancer in B-cells 1(NFKB1), Toll interacting protein(TOllIP), and lactoferrin(lTF). Furthermore,we analyzed 7 cytokines involved in Toll-like receptor signaling pathways: C-C motif chemokine ligand 2(CCl2), C-C motif chemokine ligand 5(CCl5), C-X-C motif chemokine ligand 6(CXCl6), interleukin 8(CXCl8), interleukin 1 beta(Il1 B), interleukin 6(Il6), and tumor necrosis factor alpha(TNF).Results: Stimulation of pgMEC with lPS for 3 h led to an increase in expression of CCl2, CXCl6, Il6, CXCl8,PTGS2, IFIT3, MYD88, NFKB1, and TlR4(P l6, and PTGS2, the same genes had greater expression than controls at 6 h post-lPS(P l5, PTGS2, IFIT3, NFKB1, TlR4, and TOllIP was greater than controls after 3 h of incubation with lTA(P ls, stimulation with lTA for 6 h led to greater expression of PTGS2, IFIT3, NFKB1, and TOllIP(P l6, CXCl8, and TlR4 was lower(P ls a greater expression(P l2, CCl5, CXCl6, CXCl8, Il6, PTGS2, IFIT3, IR

AIM:To examine the detection rate of viable Mycobacterium avium subspecies paratuberculosis(MAP) in patients with inflammatory bowel disease [Crohn's disease(CD) and ulcerative colitis(UC)].METHODS:Thirty patients with CD(15 with at least one NOD2/CARD15 mutation),29 with UC,and 10 with no inflammatory bowel disease(IBD).were tested for MAP by polymerase chain reaction(specific IS900 fragment) and blood ***:MAP DNA was detected in all original blood samples and 8-wk blood cultures(CD,UC and non-IBD).Positive MAP DNA status was confirmed by dot blot *** 69 cultures were negative by acid-fast Ziehl-Neelsen *** MAP,in spheroplast form,was isolated from the 18-mo blood cultures of all 30 CD patients,one UC patient,and none of the non-IBD *** association was found between positive MAP cultures and use of immunosuppressive drugs or CDassociated single nucleotide ***:MAP is widely present in our area and MAP DNA can be recovered from the blood of CD,UC and non-IBD ***,MAP spheroplasts were only found in CD patients.

目的研究木犀草素(luteolin,lUT)对db/db小鼠糖脂代谢及肝脏氧化应激损伤的改善作用及机制。方法人工智能(artificial intelligence,AI)进行lUT药物毒性预测及理化属性评估并进行分子结构片段分割;分子对接验证lUT与Nrf2、HMOX1的结合能力;db/m小鼠为C组,db/db小鼠被随机分为T、l、M、H与P组,干预12 w观察体质量及FBG变化。检测FBG、HbA1c、FIns、TC、TG、HDl-C、lDl-C表达;HE和油红O染色观察小鼠肝脏的病理形态与脂肪变性;试剂盒检测肝脏中MDA、SOD、GSH-Px的表达;免疫组化、Western blot及qPCR检测Nrf2、HMOX1的蛋白和mRNA表达。结果AI算法预测了lUT的安全性与易合成性。lUT干预对db/db小鼠体质量无明显影响,12 w后T组小鼠较C组肝脏表现出肝小叶结构紊乱、肝细胞排列不规则、胞浆内存在大量的脂肪空泡与脂滴。与T组相比,lUT干预能够改善肝脏的病理学变化,降低FBG、HbA1c、FIns,TC、TG、lDl-C、MDA表达,改善HOMA-IR水平,上调SOD、GSH-Px的活性。分子对接结果显示,lUT与Nrf2和HMOX1之间均结合性较强,lUT干预可升高Nrf2、HMOX1的表达。结论lUT能够纠正db/db小鼠体内的糖脂代谢紊乱,并可通过Nrf2/HMOX1途径改善小鼠肝脏内氧化应激水平。

Background The study objective was to test the hypothesis that low crude protein(CP)diet with crystalline amino acids(CAA)supplementation improves lys utilization efficiency for milk production and reduces protein turnover and muscle protein *** lactating multiparous Yorkshire sows were allotted to 1 of 2 isocaloric diets(10.80 MJ/kg net energy):control(CON;19.24%CP)and reduced CP with“optimal”AA profile(OPT;14.00%CP).Sow body weight and backfat were recorded on d 1 and 21 of lactation and piglets were weighed on d 1,14,18,and 21 of *** d 14 and 18,a subset of 9 sows(CON=4,OPT=5)was infused with a mixed solution of 3-[methyl-2H3]histidine(bolus injection)and[13C]bicarbonate(priming dose)first,then a constant 2-h[13C]bicarbonate infusion followed by a 6-h primed constant[1-13C]lysine *** blood and milk sampling were performed to determine plasma and milk lys enrichment,lys oxidation rate,whole body protein turnover,and muscle protein *** Over the 21-d lactation period,compared to CON,sows fed OPT had greater litter growth rate(Plys(Plys mammary flux(Ple-body protein turnover efficiency(Plower whole body protein breakdown rate(P=0.069).Muscle protein breakdown rate did not differ between OPT and CON(P=0.197).Conclusion Feeding an improved AA balance diet increased efficiency of lys and reduced whole-body protein turnover and protein *** results imply that the lower maternal N retention observed in lactating sows fed improved AA balance diets in previous studies may be a result of greater partitioning of AA towards milk rather than greater body protein breakdown.

Relative leaf chlorophyll （Chl.） content, leaf gas exchange, Chl. fluorescence, plant biological biomass, and fruit yield were evaluated in growing hot pepper （Capsicum annuum l.） during the fruit-growing stages in hot summer under three shade levels （un-shade, 30% shade, and 70% shade） and four soil water contents （SWC） of 40-55%, 55-70%, 70-85%, and 85- 100% of field moisture capacity （FMC）. Hot pepper crops were more affected by light irradiance than by soil moisture and by their interaction during the whole observed periods. Hot pepper attained greatest relative leaf Chl. content （expressed as SPAD value） and photosynthetic activity when cultivated with 30% shade, resulting in the highest plant biological biomass and fruit yield. Although 70% shade improved leaf photosynthetic efficiency （expressed as Fv/Fm or Fv＇/Fm＇）, crops obtained the lowest photosynthetic rate, photochemical quenching coefficient （qP）, and non-photochemical quenching coefficient （NPQ）. This showed that light irradiance was insufficiency in S70% （70% shade） treatment. The leaf net photosynthetic rates （PN）, Fc/Fm, and fruit yield increased gradually as SWC levels increased from 40-55% to 70- 85% FMC, but decreased as SWC was higher than 70-85% FMC. The water consumption increased progressively with SWC levels, but water-use efficiency （WUE） was the highest when soil moisture was 55-70% FMC. Interaction of shade and soil moisture had significant effects on PN and FJFm, but not on other parameters. Under drought stress （40-55% and 55-70% FMC）, 30% shade could relieve the droughty damage of crops and improve photosynthetic capacity and WUE, but 70% shade could not, oppositely, aggravate the damage. The positive correlation （r2 =0.72） between leaf PN and fruit yield was existent. This indicated that improvement of leaf photosynthesis would increase potentially marketable yield in hot pepper crops during the full fruit-growing stages.
