BTG2 interference ameliorates high glucose-caused oxidative stress, cell apoptosis, and lipid deposition in HK-2 cells

Objective: Diabetic nephropathy (DN) is a deleterious microangiopathy of diabetes, constituting a critical determinant of fatality in diabetic patients. This work is purposed to disclose the effects and modulatory mechanism of BTG anti-proliferation factor 2 (BTG2) during the pathological process of DN. Methods: BTG2 expression in kidney tissues of diabetic mice and high glucose (HG)-exposed human proximal tubular cell line HK-2 was assessed with Western blot and RT-qPCR. The diabetic mice model was constructed by streptozotocin injection and con fi rmed by the blood glucose level beyond 16.7 mmol/L. Hematoxylin and eosin (H&E) staining and measurement of kidney function hallmarks were conducted to assess kidney injury. Cell counting kit (CCK)-8 method and TUNEL assay appraised cell activity and apoptosis. Oil red O staining assayed lipid accumulation. Relevant commercial kits were used to estimate oxidative stress-related factors. Co-immunoprecipitation (Co-IP) assay testi fi ed the binding relationship of BTG2 with protein arginine methyltransferase 1 (PRMT1). Results: BTG2 expression was signi fi cantly raised in renal tissues of diabetic mice and HK-2 cells exposed to HG. BTG2 de fi ciency improved viability and extenuated the apoptosis, lipid deposition as well as oxidative stress in HK-2 cells following HG exposure. In addition, PRMT1 was also overexpressed in HK-2 cells exposed to HG. BTG2 interacted with PRMT1 and positively modulated PRMT1 expression. The effects of BTG2 interference on viability, apoptosis, lipid deposition, and oxidative stress in HG-challenged HK-2 cells were partially abrogated by PRMT1 overexpression. Conclusion: Altogether, BTG2 might aggravate HK-2 cell injury in response to HG by binding with PRMT1, providing a novel target for the therapeutic strategy of DN.

Binaural rendering is of great interest to virtual reality and immersive media. Although humans can naturally use their two ears to perceive the spatial information contained in sounds, it is a challenging task for machines to achieve binaural rendering since the description of a sound field often requires multiple channels and even the metadata of the sound sources. In addition, the perceived sound varies from person to person even in the same sound field. Previous methods generally rely on individual-dependent head-related transferred function(HRTF)datasets and optimization algorithms that act on HRTFs. In practical applications, there are two major drawbacks to existing methods. The first is a high personalization cost, as traditional methods achieve personalized needs by measuring HRTFs. The second is insufficient accuracy because the optimization goal of traditional methods is to retain another part of information that is more important in perception at the cost of discarding a part of the information. Therefore, it is desirable to develop novel techniques to achieve personalization and accuracy at a low cost. To this end, we focus on the binaural rendering of ambisonic and propose 1) channel-shared encoder and channel-compared attention integrated into neural networks and 2) a loss function quantifying interaural level differences to deal with spatial information. To verify the proposed method, we collect and release the first paired ambisonic-binaural dataset and introduce three metrics to evaluate the content information and spatial information accuracy of the end-to-end methods. Extensive experimental results on the collected dataset demonstrate the superior performance of the proposed method and the shortcomings of previous methods.

Objective:Adalimumab(ADA)is expensive and rarely used among ***,its cost for moderate to severe chronic plaque psoriasis can now be reimbursed since its addition to the National Reimbursement Drug List(NRDL)in January ***,we conducted this study to estimate the medical cost of ADA and its influencing factors in patients with psoriasis using ADA and thus gain a better understanding of the effect of the NRDL adjustment in ***:This retrospective study evaluated the medical cost of ADA before and after the addition of ADA to the NRDL(2018—2022)among 320 patients in a large professional dermatology specialty *** analysis,Welch’s analysis of variance,and multiple regression analysis were used to evaluate the medical cost and influencing ***:Before ADA was added to the NRDL,patients’mean total cost and ADA cost were¥24,243.56±16,346.53 and¥20,391.11±14,440.69,*** ADA became covered by the NRDL,the above 2 expenses decreased to¥10,461.19±7,554.66 and¥8,775.27±7,092.16,*** significant decrease in the cost of ADA resulted in a significant 110.5-times increase in ADA *** significant differences were found in the total cost,patient type(outpatient/inpatient),insurance status(yes/no),reimbursement status(before/after),and *** insurance reimbursement(P<0.001,β=-0.468,before reimbursement=1,after reimbursement=2)was the most significant influencing factor,followed by patient type(P=0.018,β=-0.215,outpatient=1,inpatient=2)and age(P=0.0046,β=-0.174).Conclusion:Medical insurance reimbursement is the most important factor affecting patients’total cost of ADA by reducing the economic burden,enhancing the availability of ADA,and stimulating the need for treatment.

The Spatial Differences and Influencing Factors of Tourism Resources in Ningxia,China

The analysis of the spatial distribution of tourism resources and the identification of its influencing factors are crucial for supporting the sustainable development of regional *** study established a comprehensive database of tourism resources in Ningxia Hui Autonomous Region(Ningxia)through a combination of literature review and field *** examined the quantitative,qualitative,and categorical characteristics of tourism resources in Ningxia,and determined the spatial patterns based on kernel density and spatial association *** study also comprehensively evaluated the societal,economic,and environmental factors influencing the spatial distribution of tourism resources in the entire region by employing the geographical detector model to quantify the influence of each *** following results were obtained.(1)There were 29218 individual tourism resources in Ningxia,comprising eight main types,23 subtypes,and 105 fundamental types,and they exhibit a hierarchical pyramidal structure.(2)The tourism resources in Ningxia displayed characteristics of“widespread regional dispersion and limited regional agglomeration”.The spatial distribution of tourism resources was highly imbalanced,and most types of tourism resources exhibit strong positive spatial correlation.(3)The altitude,annual precipitation,population density,distance from urban centers,urbanization rate,and per capita GDP were identified as significant factors influencing the spatial distribution of tourism resources in *** on the results,we recommend that the government should formulate tourism development policies in Ningxia based on local conditions to effectively address the spatial imbalances,enhance the sustainability of tourism development,and continue to promote high-quality tourism development in Ningxia.

The use of metal-organic frameworks(MOFs)as solid adsorption materials for carbon capture is promising,but achieving efficient and reversible adsorption with a balance of capacity and selectivity for carbon dioxide(CO_(2))over N_(2) remains a *** take full advantage of the strong channel traffic and robustness of MOFs with relatively small pores,it is highly necessary to employ a defect-engineering strategy to construct a broader channel structure that can facilitate the loading of functional motif-rich amino acids(AAs).This strategy can greatly enhance the CO_(2) adsorption performance of *** this study,motif-rich amino acids are loaded into the defective and robust porous frameworks via combined defect-engineering and post-synthetic *** defective Zr/Hf-MOF-808s modified with AAs,especially for the 18 mol%4-nitroisophthalic acid,generated defective products allowing for the loading of L-serine(L-Ser).This modification resulted in a significant improvement in both the adsorption capacity(248%improvement at 298 K,100 kPa)and the selectivity of CO_(2)/N_(2) using the ideal adsorbed solution theory(IAST),with the selectivity increasing to 120.55 and 38.27 at 15 and 100 kPa,respectively,while maintaining good cycling *** functional theory(DFT)simulation,CO_(2) temperature-programmed desorption(CO_(2)-TPD),and in situ Fourier transform infrared spectroscopy(FTIR)were further employed to have a better understanding of the enhanced CO_(2) adsorption ***,unlike the AAs loaded pristine MOF-808s that showed the best CO_(2) adsorption capacity with the loading of short and small glycine(Gly),the broadened channel size in our work enables the loading of functional motif-rich L-serine,which brings more active binding sites,improving CO_(2) adsorption.
