Joint Contact Forces with Changes in Running stride Length and midsole stiffness

The purpose of this study was to determine if lower extremity joint loading was influenced by stride length or shoe midsole *** subjects completed 10 trials of overground running at an average speed of 4.43 m/s in each of three condi-tions:normal running,running with a stride length(sL)reduced by 10%of normal,and running with a cushioned midsole stiffness(i.e.,mechanical impact reduction of 13.7-10.9 g).Reaction forces calculated from inverse dynamics were summed with muscle forces estimated from a musculoskeletal model using static optimization to obtain joint contact forces at the hip,knee and ankle *** components of the contact forces[axial,anterior-posterior,and medial-lateral(mL)]were examined using parametric statistics(α=0.05).Reducing stride length resulted in significant decreases in absolute peak ankle contact forces in the axial direction(normal:-14.5±1.5 BW;reduced sL:-14.0±1.6 BW)and the mL direction(normal:0.67±0.23 BW;reduced sL:0.61±0.21 BW).Reducing stride length also reduced the peak absolute axial forces at the knee(normal:-10.6±1.3 BW;reduced sL:-9.8±1.2 BW)and the hip(normal:-7.26±2.24 BW;reduced sL:-6.75±2.10 BW).The cushioned shoe did not statistically reduce the peak absolute contact forces from the normal stride condition at any of the *** hoc stress analysis suggested that the observed changes in anterior hip force would increase stress more than any of the other statistically significant *** in stride length appear to decrease some joint contact variables but cushioning in the heel region of the shoe does not.

Introduction:Tidal marsh systems along the Pacific coast of the United states have experienced substantial stress and loss of area and ecosystem function,which we examined by using the endangered California Ridgway’s Rail,Rallus obsoletus obsoletus(‘rail’)as an indicator of its tidal marsh habitat in the san Francisco *** organized a collection of historical(1885-1940)and modern(2005-2014)rail feathers and analyzed the feather isotope means for delta carbon(δ^(13)C),sulfur(δ^(34)s),and nitrogen(δ^(15)N)by region and time ***:Feather isotopes represented the primary foraging habitat during historical then modern time *** individual nor regional rail feather isotopes suggested freshwater or terrestrial foraging by the *** regions with both historic and modern feather isotopes revealed non-uniform spatial shifts in isotope levels consistent with a marine based food web and significantδ^(15)N ***:Our results supported the rail’s status as a generalist forager and obligate tidal marsh species throughout the historic *** variable isoscape trends generated from feather isotope means illustrated a modern loss of the isotopic homogeneity between regions of historical tidal marsh,which correlated with spatially-explicit habitat alterations such as increasing biological invasions and sewage effluent over ***:These findings have reinforced the importance of tidal marsh conservation in the face of ongoing underlying changes to these important ecosystems.

In the past, Crohn’s disease (CD) has been understood primarily as an immunologic disorder characterized by an abnormal T-cell response. Recent in vitro and in vivo data suggests that CD may instead be precipitated by innate immune dysfunction resulting from a combination of genetic and environmental factors. some reports have demonstrated a defective immune response in a variety of other cellular components, including neutrophils, monocytes and dendritic cells. Recent studies of granulocyte-macrophage colony-stimulating factor (Gm- CsF) in CD, aiming to stimulate the innate immune system with the conception that an innate immune defect underlies the development of the disease, have been demonstrated a clinical benefit and reinforce this evolving understanding of the disease.

Identification of epididymis-specific transcripts in the mouse and rat by transcriptional profiling

As part of our efforts to identify novel contraceptive targets in the epididymis we performed transcriptional profiling on each of the 10 and 19 segments of the mouse and rat epididymidis, respectively, using Affymetrix whole genome microarrays. A total of 17 096 and 16 360 probe sets representing transcripts were identified as being expressed in the segmented mouse and rat epididymal transcriptomes, respectively. Comparison of the expressed murine transcripts against a mouse transcriptional profiling database derived from 22 other mouse tissues identified 77 transcripts that were expressed uniquely in the epididymis. The expression of these genes was further evaluated by reverse transcription polymerase chain reaction （RT-PCR） analysis of RNA from 21 mouse tissues. RT-PCR analysis confirmed epididymis-specific expression of Defensin Beta 13 and identified two additional genes with expression restricted only to the epididymis and testis. Comparison of the 16 360 expressed transcripts in the rat epididymis with data of 21 other tissues from a rat transcriptional profiling database identified 110 transcripts specific for the epididymis. sixty-two of these transcripts were further investigated by qPCR analysis. Only Defensin 22 （E3 epididymal protein） was shown to be completely specific for the epididymis. In addition, 14 transcripts showed more than 100-fold selective expression in the epididymis. The products of these genes might play important roles in epididymal and/or sperm function and further investigation and validation as contraceptive targets are warranted. The circadian timing system plays a key role in orchestrating lipid metabolism. In concert with the solar cycle, the circadian system ensures that daily rhythms in lipid absorption, storage, and transport are temporally coordinated with rest-activity and feeding cycles. At the cellular level, genes involved in lipid synthesis and fatty acid oxidation are rhythmically activated and repressed by core clock proteins in a tissue-specific manner. Consequently, loss of clock gene function or misalignment of circadian rhythms with feeding cycles （e.g., in shift work） results in impaired lipid homeostasis. Herein, we review recent progress in circadian rhythms research using lipidomics, i.e., large-scale profiling of lipid metabolites, to characterize circadian-regulated lipid pathways in mammals. In mice, novel regulatory circuits involved in fatty acid metabolism have been identified in adipose tissue, liver, and muscle. Extensive diversity in circadian regulation of plasma lipids has also been revealed in humans using lipidomics and other metabolomics approaches. In future studies, lipidomics platforms will be increasingly used to better understand the effects of genetic variation, shift work, food intake, and drugs on circadian-regulated lipid pathways and metabolic health.

The epididymis is divided into caput, corpus and cauda regions, organized into intraregional segments separated by connective tissue septa （CTs）. In the adult rat and mouse these segments are highly differentiated. Regulation of these segments is by endocrine, lumicrine and paracrine factors, the relative importance of which remains under investigation. Here, the ability of the CTs to limit signaling in the interstitial compartment is reviewed as is the effect of 15 days of unilateral efferent duct ligation （EDL） on ipsilateral segmental transcriptional profiles. Inter-segmental microperifusions of epidermal growth factor （EGF）, vascular endothelial growth factor （VEGFA） and fibroblast growth factor 2 （FGF2） increased phosphorylation of mitogen activated protein kinase （mAPK） in segments 1 and 2 of the rat epididymis and the effects of all factors were limited by the CTs separating the segments. microan＇ay analysis of segmental gene expression determined the effect of 15 days of unilateral EDL on the transcriptome-wide gene expression of rat segments 1-4. Over 11 000 genes were expressed in each of the four segments and over 2 000 transcripts in segment 1 responded to deprivation of testicular lumicrine factors. segments 1 and 2 of control tissues were the most transcriptionally different and EDL had its greatest effects there. In the absence of lumicrine factors, all four segments regressed to a transcriptionally undifferentiated state, consistent with the less differentiated histology. Deprivation of lumicrine factors could stimulate an individual gene＇s expression in some segments yet suppress it in others. such results reveal a higher complexity of the regulation of rat epididymal segments than that is generally appreciated. （Asian J Androl 2007 July; 9： 565-573）

Pelvic region bone density,soft tissue mass,and injury frequency in female professional ballet dancers and soccer athletes

We recently observed a high prevalence of low pelvic bone mineral density(BmD)in female professional ballet *** this population is susceptible to musculoskeletal overuse injuries,we aimed to determine which regions of the pelvis may be at greatest risk compared to general population females(GENPOP)as well as professional female soccer players(sOCCER,a comparison to other elite athletes regularly subjected to high degrees of loading).Three groups of age-matched females[(GENPOP;n=38,27±1yrs),(BALLET;single com-pany,n=36,26±3yrs),(sOCCER;single NWsL■club,n=34,25±1yrs)]consented to have their BmD and body composition assessed(DEXA,GE■).In addition to soft tissue and total and regional BmD analyses,a segmental analysis of the pelvis was performed to determine site-specific BmD for the iliac fossa,iliac fossa/iliac crest/ilium combined,pubic bone,ischium,and sacrum.A mixed-model ANOVA followed by a Tukey's post-hoc test was used to compare the groups(Type-I error;α=0.05).The BALLET group had lower pelvic BmD for all measures(Avg.%Diff.=15%-27%,pmpared to the sOCCER group and for the ischium(Avg.%Diff.=8%;p=0.007)and sacrum(Avg.%Diff.=7%;p=0.028)compared to the GENPOP *** BALLET group had lower lean mass for all measures compared to the other groups(Avg.%Diff.=12%-18%;pssional ballet performers exhibit reduced pelvic region soft tissue and site-specific BmD not previously detected using standard DEXA *** findings highlight which pelvic regions may benefit from preventative strength training and/or nutritional interventions.
