Elucidating the Degradation Pathways of human Insulin in the Solid State

while there have been significant advances in the development of peptide oral dosage forms in recent years, highlighted by the clinical and commercial success of approved peptides such as Rybelsus?, there remain several barriers in the way of broad range applicability of this approach to peptide delivery. One such barrier includes the poor physical and chemical stability inherent to their structures, which persists in the solid state although degradation typically occurs at different rates and via different pathways in comparison to the solution state. Using insulin as a model peptide, this work sought to contribute to the development of analytical techniques for investigating common insulin degradation pathways. Chemically denatured,deamidated and aggregated samples were prepared and used to benchmark circular dichroism spectroscopy, reverse phase hPLC and size exclusion chromatography methods for the investigation of unfolding, chemical modifications and covalent aggregation of the insulin molecule respectively. Solid state degraded samples were prepared by heating insulin powder at 60 °C and 75% relative humidity for 1, 3, 5 and 7 d, and the degradation profiles of the samples were evaluated and compared with those observed in solution. while no unfolding was observed to occur, significant deamidation and covalent aggregation were detected. Reductive disulfide bond cleavage using dithiothreitol allowed for separation of the insulin A-and B-chains,offering a facile yet novel means of assessing the mechanisms of deamidation and covalent aggregation occurring in the solid state.

Between Euthanasia and Dysthanasia: The Ethical Issue of Sedation in the Terminal Phase of Illness

Background: Deep sedation, euthanasia and therapeutic relentlessness lead us today to rethink the paradigm of life and the contingency of human existence. Between therapeutic relentlessness, the unreasonable care which uses heavy therapeutic means which are often disproportionate to the expected benefit, namely keeping alive a patient whose condition is considered medically hopeless;and euthanasia which would precipitate the process of death would be sedation in the terminal phase of the illness. Should doctors and families of comatose patients decide the “life” and “death” of their patients? For anti-euthanasia associations, doctors, relatives of terminally ill patients and the State itself, if they accept the principle of euthanasia, they are “murderers”, while for pro-euthanasists, the dignity of the human being would recommend that the days of patients in situations considered critical be shortened, to avoid unnecessary suffering and humiliation. Methods: A systematic review of the literature was carried out to identify relevant articles relating to euthanasia, dysthanasia and sedation in the terminal phase of illness. The search was conducted in French or English in three databases: PubMed, Google Scholar and Science Direct. Objectives: The objectives of this article are: 1) define the terminologies and concepts of palliative sedation, deep sedation, deep and continuous sedation until death, euthanasia and dysthanasia;2) present aspects of the meaning of life and the human person in African cultures;and 3) propose an ethical reflection on the value of life. Results: After precisely defining the concepts of euthanasia, dysthanasia and sedation, this research presented the African anthropological and ethical approach to the mysteries of life and death. Conclusion: with this in mind, the golden rule of medicine always remains as such “Primum non nocere”.

Mantle Driven Early Eocene Magmatic Flare-up of the Gangdese Arc, Tibet: A Case Study on the Nymo Intrusive Complex

Magmatic periodicity is recognized in continental arcs worldwide, but the mechanism responsible for punctuated arc magmatism is controversial. Continental arcs in the Trans-himalayan orogenic system display episodic magmatism and the most voluminous flare-up in this system was in early Eocene during the transition from subduction to collision. The close association of the flare-up with collision is intriguing. Our study employs zircon Lu-hf and bulk rock Sr-Nd isotopes, along with mineral geochemistry, to track the melt sources of the Nymo intrusive complex and the role of mantle magma during the early Eocene flare-up of the Gangdese arc, Tibet. The Nymo intrusive complex is composed of gabbronorite, diorite, quartz diorite, and granodiorite which define an arc-related calc-alkaline suite. Zircon U-Pb ages reveal that the complex was emplaced between ~50–47 Ma. Zircon hf isotopes yield εhf(t) values of 8.2–13.1, while whole-rock Sr and Nd isotopes yield εNd(t) values of 2.7–6.5 indicative of magmatism dominated by melting of a juvenile mantle source with only minor crustal assimilation(~15%–25%) as indicated by assimilation and fractional crystallization modeling. Together with published data, the early Eocene magmatic flare-up was likely triggered by slab breakoff of subducted oceanic lithosphere at depths shallower than the overriding plate. The early Eocene magmatic flare-up may have contributed to crustal thickening of the Gangdese arc. This study provides important insights into the magmatic flare-up and its significant role in the generation of large batholiths during the transition from subduction to collision.

Basalt fiber reinforcement in polymer matrix composites is becoming more and more popular because of its environmental friendliness and mechanical qualities that are comparable to those of synthetic *** fiber strengthened vinyl ester matrix polymeric composite with filler addition of nano-and micro-sized silicon carbide(SiC)element spanning from 2 weight percent to 10 weight percent was studied for its mechanical and wear *** application of Artificial Neural Network(ANN)to correlate the filler addition composition for optimum mechanical properties is required due to the non-linear mechanical and tribological features of *** stuffing blend and composition of the composite are optimized using the hybrid model and Genetic Algorithm(GA)to maximize the mechanical and wear-resistant *** predicted and tested ANN–GA optimal values obtained for the composite combination had a tensile,flexural,impact resilience,hardness and wear properties of 202.93 MPa,501.67 MPa,3.460 J/s,43 hV and 0.196 g,respectively,for its optimum combination of filler and *** can be noted that the nano-sized SiC filler particle enhances most of the properties of the composite which diversifies its *** predicted mechanical and wear values of the developed ANN–GA model were in closer agreement with the experimental values which validate the model.

Ultrafast 3D imaging is indispensable for visualizing complex and dynamic biological *** scanning-based techniques necessitate an inherent trade-off between acquisition speed and space-bandwidth product(SBP).Emerging single-shot 3D wide-field techniques offer a promising alternative but are bottlenecked by the synchronous readout constraints of conventional CMOS systems,thus restricting data throughput to maintain high SBP at limited frame *** address this,we introduce EventLFM,a straightforward and cost-effective system that overcomes these challenges by integrating an event camera with Fourier light field microscopy(LFM),a state-of-theart single-shot 3D wide-field imaging *** event camera operates on a novel asynchronous readout architecture,thereby bypassing the frame rate limitations inherent to conventional CMOS *** further develop a simple and robust event-driven LFM reconstruction algorithm that can reliably reconstruct 3D dynamics from the unique spatiotemporal measurements captured by *** results demonstrate that EventLFM can robustly reconstruct fast-moving and rapidly blinking 3D fluorescent samples at khz frame ***,we highlight EventLFM’s capability for imaging of blinking neuronal signals in scattering mouse brain tissues and 3D tracking of GFP-labeled neurons in freely moving *** believe that the combined ultrafast speed and large 3D SBP offered by EventLFM may open up new possibilities across many biomedical applications.
