Development of alternatively activated （M2） macrophage phenotypes is a complex process that is coordinately regulated by a plethora of pathways and factors. Here, we report that RBP-j, a DNA-binding protein that integrates signals from multiple pathways including the Notch pathway, is critically involved in polarization of M2 macrophages. Mice deficient in RBP-j in the myeloid compartment exhibited impaired M2 phenotypes in vivo in a chitin-induced model of M2 polarization. Consistent with the in vivo findings, M2 polarization was partially compromised in vitro in Rbpj-deficient macrophages as demonstrated by reduced expression of a subset of M2 effector molecules including arginase 1. Functionally, myeloid Rbpj deficiency impaired M2 effector functions including recruitment of eosinophils and suppression of T cell proliferation. Collectively, we have identified RBP- jas an essential regulator of differentiation and function of alternatively activated macrophages.

Alkaline-earth dihalide nanocrystals(NCs) such as srFCl, owing to their high chemical stability and low phonon energy, are excellent host materials for lanthanide(Ln3+) doping to achieve desirable optical properties for various bioapplications, Herein, we report a novel strategy for the synthesis of sub-10 nm Ln3+-doped srFCl NCs with efficient upconverting and downshifting luminescence through a facile onestep hot-injection method. Utilizing the temperature-dependent upconverting luminescence(UCL) from the thermally coupled 2H11/2 and 4s3/2 levels of Er3+, we showed the potential of srFCl:Yb,Er NCs as sensitive UCL nanoprobes for non-contact thermal sensing with a maximum detection sensitivity of 0.0066 K-1, which is among the highest values for thermal sensing based on Er3+-activated UCL nanoprobes. Furthermore, by employing the intense downshifting luminescence from Tb3+ and Eu3+, we demonstrated the successful use of biotinylated srFCl:Ce,Tb and srFCl:Eu3+ nanoprobes for biotin receptor-targeted cancer cell imaging, thus revealing the great promise of srFCl:Ln3+ nanoprobes for versatile bioapplications.

To detect anaerobic bacteria Clostridium sp . and Bacteroides fragilis in intrahepatic stones by molecular genetic method Methods DNA was extracted from 59 stone samples and subjected to polymerase chain reaction (PCR) amplification targeting the 16s rRNA gene of Clostridium sp . and the glutamine synthetase gene of Bacteroides fragilis single-strand conformational polymorphism (ssCP) analysis was performed to identify the Clostridium sp Results 16s rRNA gene sequences for Clostridium sp. were identified in 49 stones (83%, 49/59) The two most common groups were detected in 19 (41%) and 17 (37%) of the 46 samples using ssPC analysis, and 25/59 (42%) stones were tested positive for Bacteroides fragilis Conclusions Anaerobes such as Clostridium sp and Bacteroides fragilis present in intrahepatic stones and may play a role in stone formation PCR is a useful technique to detect fastidious pathogens, which are difficult to culture ssCP of PCR products is a rapid method in differentiating bacterial species

This study provided insights into the persistent yellowish color in biological and tertiary effluents of municipal wastewater through a multi-characterization approach and fluorescence excitation-emission matrix-parallel factor(EEM-PARAFAC)*** characterization was performed on three to five full-scale municipal wastewater treatment plants(WWTPs),including differential log-transformed absorbance(DLn A)spectroscopy,resin fractionation,size-exclusion chromatography for apparent molecular weight analysis(sECAMW),and X-ray photoelectron spectroscopy(XPs)*** acids(HPOA)were abundant in visible colored dissolved organic matter(DOM).The sEC-AMW result showed that the molecular weight of the colored substances in the secondary effluents is mainly distributed in the range of 2–3 k *** XPs analysis,C-O/C-N and pyrrolic/pyridonic(N-5)were found to be positively correlated with *** component models were built on biological(two components)and tertiary effluent(three components)and the correlation analysis revealed that PARAFAC component 2 in biological effluent(BE-C2)and component 1in tertiary effluent(TE-C1),which were ascribed to Hydrophobic acids and Humic acid-like,were the responsible visible colored DOM components cause yellowish *** addition,component similarity testing found that the identified visible colored DOM PARAFAC BE-C2,and PARAFAC TE-C1 were identical(0.96)in physicochemical properties,with 4%removal efficacy on average,compared with 11%for invisible colored *** implied that tertiary effluents containing colorants(TE-C1)were resistant to degradation/removal using different disinfection and filtration processes in advanced *** sheds light on many physicochemical aspects of PARAFAC-identified visible colored DOM components and provides spectral data to build an online monitoring system.

Background:Deep body temperature is a critical indicator of heat ***,direct measures are often invasive,costly,and difficult to implement in the *** study assessed the agreement between deep body temperature estimated from heart rate and that measured directly during repeated work bouts while wearing explosive ordnance disposal(EOD)protective clothing and during ***:Eight males completed three work and recovery periods across two separate *** consisted of treadmill walking on a 1%incline at 2.5,4.0,or 5.5 km/h,in a random order,wearing EOD protective *** temperature and relative humidity were maintained at 24℃and 50%[Wet bulb globe temperature(WBGT)(20.9±1.2)℃]or 32℃and 60%[WBGT(29.0±0.2)℃]on the separate days,*** rate and gastrointestinal temperature(TGI)were monitored continuously,and deep body temperature was also estimated from heart rate(ECTemp).Results:The overall systematic bias between TGI and ECTemp was 0.01℃with 95%limits of agreement(Lo A)of±0.64℃and a root mean square error of 0.32℃.The average error statistics among participants showed no significant differences in error between the exercise and recovery periods or the environmental *** TGI levels of(37.0–37.5)℃,(37.5–38.0)℃,(38.0–38.5)℃,and>38.5℃,the systematic bias and±95%Lo A were(0.08±0.58)℃,(–0.02±0.69)℃,(–0.07±0.63)℃,and(–0.32±0.56)℃,***:The findings demonstrate acceptable validity of the ECTemp up to 38.5℃.Conducting work within an ECTemp limit of 38.4℃,in conditions similar to the present study,would protect the majority of personnel from an excessive elevation in deep body temperature(>39.0℃).

Identifying genetic risk factors for Alzheimer's disease(AD)is an important research *** date,different endophenotypes,such as imaging-derived endophenotypes and proteomic expression-derived endophenotypes,have shown the great value in uncovering risk genes compared to case-control ***,a co-varying pattern of different omics-derived endophenotypes could result from the shared genetic ***,existing methods mainly focus on the effect of endophenotypes alone;the effect of cross-endophenotype(CEP)associations remains largely *** this study,we used both endophenotypes and their CEP associations of multi-omic data to identify genetic risk factors,and proposed two integrated multi-task sparse canonical correlation analysis(inMTsCCA)methods,i.e.,pairwise endophenotype correlationguided MTsCCA(pcMTsCCA)and high-order endophenotype correlation-guided MTsCCA(hocMTsCCA).pcMTsCCA employed pairwise correlations between magnetic resonance imaging(MRI)-derived,plasma-derived,and cerebrospinal fluid(CsF)-derived endophenotypes as an additional *** used high-order correlations among these multi-omic data for *** figure out genetic risk factors at individual and group levels,as well as altered endophenotypic markers,we introduced sparsity-inducing penalties for both *** compared pcMTsCCA and hocMTsCCA with three related methods on both simulation and real(consisting of neuroimaging data,proteomic analytes,and genetic data)*** results showed that our methods obtained better or comparable canonical correlation coefficients(CCCs)and better feature subsets than *** importantly,the identified genetic loci and heterogeneous endophenotypic markers showed high ***,jointly using multi-omic endophenotypes and their CEP associations is promising to reveal genetic risk factors.

Geochemistry and origin of deep-seated cracked gas on the northern slope of the Dongying sag, shandong Province

Deep-seated cracked gas exploration was achieved great breakthrough in Es4L of the Minfeng area on the northern slope of the Dongying sag. Carbon isotopic and molecular compositions studies revealed the characteristics of wet gas and a normal trend of carbon isotopic composition. Empirical cutoff points of δ13C2 and δ13C3 and light hydrocarbon compositions distinguished the cracking gas as sapropelic gas. Variations in i/nC5, i/nC4 and δ13C2 fur-ther confirmed that the gas was cracked from residual kerogen. source characteristics indicated that the gas was de-rived from mature-highly mature source rocks of Es4 with kerogen type II being dominant mixed with some oil-cracking gas. Burial history modeling indicated that there were two hydrocarbon charging periods in Es4L reser-voirs. The first period refers to the Guantao-Minghuazhen stage dominated by oil charging, while the second period refers to the Minghuazhen stage and has been dominated by cracked gas charging till now.
