Diversity and Morphometrics of Palm Weevils of the Genus Rhynchophorus in Maniema Province, Democratic Republic of Congo

An inventory of Rhynchophorus species was carried out to determine their diversity, biogeographical variability and morphometrics. A shotgun trapping method was used at nine sites in three districts over a four-month period, identifying three species: R. quadrangulus, R. phoenicis, R. ferrugineus and a probable variant of R. phoenicis. Of these species, R. phoenicis stands out as the most widespread palm weevil, while the probable variant of R. phoenicis is considered the rarest subspecies in the Maniema region. The color of the head and elytra is a criterion for distinguishing the different species of Rhynchophorus spp. Morphological criteria indicate that the probable subspecies of R. phoenicis is the largest palm weevil in the Maniema region and even globally. It is followed by R. quadrangulus and, by far, by R. phoenicis. The smallest palm weevil in Maniema is R. ferrugineus. Moreover, these criteria reveal that, for all three species identified, the female is generally larger than the male. This study has the merit of extending the distribution of R. ferrugineus from the Maghreb to the central East of the DRC, and highlighting a subspecies of R. phoenicis. Recognition of the sexes on the basis of the shape of the tip of the abdomen (oval in the male and flat in the female) is an additional contribution.

In Vivo Subacute Liver Toxicity of “Attoté”: A Herbal Drink

The herbal drink “Attoté” has been widely used in the Abidjan district to treat a number of illnesses, notably erectile dysfunction. Despite the popularity of its therapeutic effects, very few studies have been carried out on its effects on the health of users. The aim of this study was to identify the constituents contained in the phytomedicinal product and to assess their potential adverse effects in vivo. Phytochemical screening was conducted to identify the bioactive molecules in “Attoté” and to evaluate its hepatic effects in vivo. Forty (40) Wistar rats, randomly divided into 4 groups, with 10 animals per group (5 males and 5 females) were used to study potential hepatotoxic effects. Group 1 animals (control group) received distilled water. Batches I, II and III received by gavage a volume of Attoté extract corresponding to 1 ml/100 g body weight at 200 mg/kg, 400 mg/kg and 800 mg/kg, respectively. Attoté extract was administered daily at the same time for 28 days, and serum was collected every two weeks to assess hepatic biochemical markers by spectrophotometry using a Cobas C311® HITACHI biochemistry system. After one month of study, the rats were euthanized by ether overdose and the livers were harvested for morphological and histopathological analysis. Phytochemical screening revealed the presence of alkaloids, polyphenols, leucoanthocyanes, anthraquinones and quinones. Hepatic biochemical and hematological parameters such as red globular, hemoglobin, alanine aminotransferase (ALAT), aspartate aminotransferase (ASAT), alkaline phosphatase (ALPs) and gamma glutamyl transferase (GGT) showed no significant change (p > 0.05) in the treated rat group compared with controls. However, these variations were moderate and transient, with values remaining almost within their standard limits. Rhizosphere Soil Microbial Characteristics and Enzyme Activities under Cajanus cajan and Milletia laurentii Grown in Scientific Center of Brazzaville, Republic of the Congo

This study was conducted at the scientific center of Brazzaville. The objective was to assess the microbial characteristics and enzymes activities in the rhizosphere soil of Cajanus cajan and Milletia lauurentii. These plants have great importance in food and forestry. Microbial diversity management in the rhizosphere is the key for sustainable crop production or forest durability. DNA metagenomic sequencing was used to analyze the whole bacterial diversity, the microbial biomass was determinate by the fumigation-extraction method and the enzymes by the p-nitrophenol-β-D-glucoside for β-glucosidase, the p-nitrophenyl-N-acetyl-β-D-glucosaminide for β-Glucosaminidase. Dehydrogenase and acid phosphatase were quantified using 2,3,5-tryphenyl tetrazolium chloride and p-nitophenylphosphate respectively. The results show that, in Cajanus cajan culturable bacteria genera were mainly Acidobacterium, Skermanella, Rhodoplanes, Bacillus, Chloroflexus, Steroidobacter, Sphingomonas and Bradyrhizobium while in Milletia laurentii: Rhodoplanes, Bradyrhizobium, Bacillus, Sphingobacterium, Acidobacterium, Mesorhizobium, Nitrospira were the principal genera. In the two rhizosphere soils investigated, the uncultured bacteria exhibited relatively higher abundance, often for the same genera, than culturable bacteria. Metagenomic studies have revealed more bacterial diversity in each compared to when culturable bacteria were taken into account alone. The MBC and MBN were higher in the rhizosphere of Milletia than in rhizosphere of Cajanus. The same trend was observed with the enzyme activities. PCA of culturable and NMDS of unculturable soil bacteria genera shows that factors mainly involved in the carbon cycle such as MBC, members of the microbial community i.e. Acidobacterium, Skermanella, Chloroflexus, sand, C, β-glucosaminidase and dehydrogenase, were strongly correlated with Cajanus cajan. Cytotoxicity and Hepatic Effects in Vivo of the Phytomedicinal Antipalu Used by the Population of Abidjan

Antipalu is a phytomedicinal medicinal beverage that is popular in the District of Abidjan, particularly for the treatment of malaria. However, Antipalu could present potential health effects on patients, and few toxicological studies have been conducted before its use. In order to determine the cytotoxicity of Antipalu, two complementary tests, LDH activity and the MTT cell proliferation assay, were used using Vero cells. Vero cells were exposed to increasing concentrations of Antipalu and incubated for 24, 48 and 72 hours. In addition, forty (40) rats distributed randomly into 4 groups, including 10 animals per group (5 males and 5 females) were used for the potential hepatoxic effects. Animals in group 1 received distilled water and were used as a control group. On the other hand, Lot I, II and III received by gavage a volume of the Antipalu extract corresponding to 1 ml/100 g of body weight at 200 mg/kg, 400 mg/kg, 800 mg/kg, respectively. The extract was administered daily at the same time for 28 days and serum was collected once a week to evaluate hepatic biochemical markers. After 28 days of study, all rats were euthanized by an overdose of ether and the liver of the rats was removed for gross morphological and histopathological analysis. The results of the cell supernatant assay showed an increasing extracellular LDH enzyme activity with lethal concentrations at 10% and 50% (LC10 = 111 µg/mL and LC50 = 555 µg/mL, respectively). In addition, the MTT assay showed a decrease in mitochondrial activity and thus cell proliferation after 24, 48 and 72 H of incubation. Our study showed that Antipalu caused alterations in the plasma membranes of the cells, resulting in the release of lactase dehydrogenase (LDH) into the external environment and a decrease in the mitochondrial activity of the Vero cells. The Outcomes of Pregnancy in Patients with Cirrhosis in a Low-Income Country

Pregnancy in cirrhotic-woman is a rare event [1] [2]. It causes particular risks related to liver disease and its treatments, with maternal and fetal consequences. The largest reported series to date is by Welton’s with 13 cases [3]. In Africa, few cases were reported. The purpose of this work was to report the hepatic and obstetric complications in pregnant women in Burkina Faso, a low-income country. It was a descriptive cross-sectional study with retrospective data collection covering the period from January 1, 2013 to December 31, 2023. All the women diagnosed with cirrhosis through clinical basis, biological and imaging arguments associated with a pregnancy confirmed by ultrasound were included. A total of ten women were included in the study. Among them, two were known cirrhotics and were being followed. In the other eight cases, cirrhosis was discovered during pregnancy. Eight women were HBsAg positive and two of them had only the contact marker of hepatitis B virus (anti- HBc antibody). The evolution was, ascitic decompensation in nine cases, digestive hemorrhage due to rupture of esophageal varices in two cases, nine cases of anemia, two cases of ascitic fluid infection and one case of conjunctival jaundice. Two cases of probable hepatocellular carcinoma were noted with one case of extensive portal thrombosis. The evolution was without maternal complications in one case. Regarding fetal and neonatal complications, there were three cases of hypotrophy, one case of prematurity, one case of acute neonatal distress, two cases of fetal death in utero and one case of fetal tachycardia. Four pregnancies had a favorable evolution without any complications. The association of cirrhosis and pregnancy is an uncommon event, and maternal and fetal complications can occur, making this pregnancy a high-risk pregnancy.

Influence of surface modified mixed metal oxide nanoparticles on the electrochemical and mechanical properties of polyurethane matrix

Newly synthesized functional nanoparticles,3-amino-1,2,4-triazole(ATA)/SiO_(2)—TiO_(2)were introduced to the polyurethane(PU)*** techniques were used to investigate the barrier properties of the synthesized PU—ATA/SiO_(2)—TiO_(2)nanocomposite coated steel *** natural seawater,electrochemical impedance spectroscopy experiments indicated outstanding protective behaviour for the PU—ATA/SiO_(2)—TiO_(2)coated *** coating resistance(Rcoat)of PU—ATA/SiO_(2)—TiO_(2)was determined to be 2956.90 kΩ·cm^(−2).The Rcoat of the PU—ATA/SiO_(2)—TiO_(2)nanocomposite coating was found to be over 50%higher than the PU *** current measured along the scratched surface of the PU—ATA/SiO_(2)—TiO_(2)coating was found to be very low(1.65 nA).The enhanced ATA/SiO_(2)—TiO_(2)nanoparticles inhibited the entry of electrolytes into the coating interface,as revealed by scanning electron microscopy/energy dispersive X-ray spectroscopy and X-ray diffraction analysis of the degradation *** contact angle testing validated the hydrophobic nature of the PU—ATA/SiO_(2)—TiO_(2)coating(θ=115.4°).When the concentration of ATA/SiO_(2)—TiO_(2)nanoparticles was 2 wt%,dynamic mechanical analysis revealed better mechanical ***,the newly synthesised PU—ATA/SiO_(2)—TiO_(2)nanocomposite provided excellent barrier and mechanical properties due to the addition of ATA/SiO_(2)—TiO_(2)nanoparticles to the polyurethane,which inhibited material degradation and aided in the prolongation of the coated steel’s life.

Mitotic phosphorylation of PRC1 at Thr470 is required for PRC1 oligomerization and proper central spindle organization

During cell division, chromosome segregation is orchestrated by the interaction of spindle microtubules with thecentromere. A dramatic remodeling of interpolar microtubules into an organized central spindle between the separatingchromatids is required for the initiation and execution of cytokinesis. Central spindle organization requires mitotic kine-sins, the chromosomal passenger protein complex, and microtubule bundling protein PRC1. PRC1 is phosphorylated byCdc2 at Thr470 and Thr481 during mitosis. However, the functional relevance of PRC1 phosphorylation at Thr470 hasremained elusive. Here we show that expression of the non-phosphorylatable mutant PRC1but not the phospho-mimi-cking mutant PRC1causes aberrant organization of the central spindle. Immunoprecipitation experiment indicatesthat both PRC1and PRC1mutant proteins associate with wild-type PRC1, suggesting that phosphorylationof Thr470 does not alter PRC1 self-association. In addition, in vitro co-sedimentation experiment showed that PRC1binds to microtubule independent of the phosphorylation state of Thr470. Gel-filtration experiment suggested that phos-phorylation of Thr470 promotes oligomerization of PRC1. Given the fact that prevention of the Thr470 phosphorylationinhibits PRC1 oligomerization in vitro and causes an aberrant organization of central spindle in vivo, we propose thatthis phosphorylation-dependent PRC1 oligomerization ensures that central spindle assembly occurs at the appropriatetime in the cell cycle.

Toll-like receptor signaling in hematopoietic homeostasis and the pathogenesis of hematologic diseases

Toll-like receptors （TLRs）, which are found in innate immune cells, are essential mediators of rapid inflammatory responses and appropriate T-cell activation in response to infection and tissue damage. Accumulating evidence suggests that TLR signaling is involved in normal hematopoiesis and specific hematologic pathologies. Particular TLRs and their downstream signaling mediators are expressed not only in terminally differentiated innate immune cells but also in early hematopoietic progenitors. Sterile activation of TLR signaling is required to generate early embryonic hematopoietic progenitor cells. In adult animals, TLR signaling directly or indirectly promotes differentiation of myeloid cells at the expense of that of lymphoid cells and the self renewal of hematopoietic stem cells during infection and tissue damage. Activating mutations of the MyD88 gene, which codes for a key adaptor involved in TLR signaling, are commonly detected in B-cell lymphomas and other B-cell hematopathologies. Dysregulated TLR signaling contributes to the pathogenesis of many hematopoietic disorders, including bone marrow failure, myelodysplastic syndrome, and acute myeloid leukemia. Complete elucidation of the molecular mechanisms by which TLR signaling mediates the regulation of both normal and pathogenic hematopoiesis will prove valuable to the development of targeted therapies and strategies for improved treatment of hematopoietic disorders.
