Whole blood platelet aggregometry in HIV-infected patients on treatment with abacavir

Purpose: To assess platelet activity in HIV-patients with and without antiretroviral therapy (ART), analysing the influence of the presence or absence of ABC in the regimen. Methods: Observational, pilot study, including 30 asymptomatic HIV-patients: 20 on ART for at least 24 weeks and with undetectable HIV viral load - 10 on ABC, 10 on tenofovir (TDF) - and 10 na?ve patients, and a control group of 10 HIV-negative subjects. No subject was receiving drugs with antiagregant activity. Platelet activity was assessed by measuring time-dependent platelet aggregometry (electrical impedance on fasting whole blood), induced by ADP (1.25, 2.5 μM), collagen (0.5, 1 μg/mL), arachidonic acid (100, 200 μM), and U46619 (receptor agonist of the tromboxano A2) (1.25, 2.5 μM). Statistic program: SPSS, 16.0. Results: Demographic, anthropometric data, and cardiovascular risk factors were similar in all groups, but older age and longer time of HIV infection in the ABC group (50.4 vs 36.1, 34.2 and 42.7 years, respectively;p < 0.05, and 140.3 vs 88.1 and 48.3 months in the two other groups of HIV patients;p < 0.05). Mean CD4 cells count was 564/mm3. Platelet aggregation with exposure to U46619 was higher in the ABC compared with the TDF group (11.1 vs 4.4%;p = 0.007), na?ve patients (11.1 vs 5.7%;p = 0.014), and the HIV-negative group (11.1 vs 6.5%;p = 0.04). These differences remained significant when controlled for age and time of HIV infection. Conclusions: ABC increases platelet aggregability possibly in relation with the receptor of tromboxano. Wider studies are needed to confirm this hypothesis.

Translational pancreatic cancer research:a comparative study on patient-derived xenograft models

AIM To assess the viability of orthotopic and heterotopic patient-derived pancreatic cancer xenografts implanted into nude *** This study presents a prospective experimental analytical follow-up of the development of tumours in mice upon implantation of human pancreatic adenocarcinoma samples. Specimens were obtained surgically from patients with a pathological diagnosis of pancreatic adenocarcinoma. Tumour samples from pancreatic cancer patients were transplanted into nude mice in three different locations(intraperitoneal, subcutaneous and pancreatic). Histological analysis(haematoxylin-eosin and Masson's trichrome staining) and immunohistochemical assessment of apoptosis(TUNEL), proliferation(Ki-67), angiogenesis(CD31) and fibrogenesis(α-SMA) were performed. When a tumour xenograft reached the target size, it was reimplanted in a new nude mouse. Three sequential tumour xenograft generations were generated(F1, F2 and F3).RESULTS The overall tumour engraftment rate was 61.1%. The subcutaneous model was most effective in terms of tissue growth(69.9%), followed by intraperitoneal(57.6%) and pancreatic(55%) models. Tumour development was faster in the subcutaneous model(17.7 ± 2.6 wk) compared with the pancreatic(23.1 ± 2.3 wk) and intraperitoneal(25.0 ± 2.7 wk) models(P = 0.064). There was a progressive increase in the tumour engraftment rate over successive generations for all three models(F1 28.1% vs F2 71.4% vs F3 80.9%, P < 0.001). There were no significant differences in tumour xenograft differentiation and cell proliferation between human samples and the three experimental models among the sequential generations of tumour xenografts. However, a progressive decrease in fibrosis, fibrogenesis, tumour vascularisation and apoptosis was observed in the three experimental models compared with the human samples. Endoscopic recommendations for colorectal cancer screening and surveillance in patients with inflammatory bowel disease:Review of general recommendations

Screening for colorectal cancer(CRC) in patients with inflammatory bowel disease(IBD) is recommended by all scientific societies. However, there are differences in the recommendations they make regarding screening and surveillance. We address a series of questions that come up in the daily clinical practice of a physician. The first two questions that are raised are:(1) Who should be offered screening for CRC? and(2) When should the first colonoscopy be performed? The next step is to decide who should undergo endoscopic surveillance and at what intervals they should be performed. Chromoendoscopy is emerging as the recommended endoscopic technique for screening and surveillance. The terminology for describing lesions detected with endoscopy is also changing. The management of visible lesions or non-visible dysplasia is also a motive for the review. We end the review by addressing the followup for endoscopically resected lesions. These questions often cannot be answered easily due to the varying degrees of evidence available; therefore, we have made some general recommendations based on those made by the various guidelines and consensuses. The first screening colonoscopy should be offered 8 years after a IBD diagnosis and we recommend that patients be stratified according to the individual risk for each for endoscopic surveillance intervals.

RNAi technology targeting PbGP43 and PbP27 in Paracoccidioides brasiliensis

Efficient technologies for gene silencing would be important to carry out functional analysis with P. brasiliensis genes, as well as for a better understanding of the biology and pathogenesis of this pathogenic fungus. Due to the fact that homologous recombination is unusual in P. brasiliensis, the development of knockout isolates is currently non-feasible. The goal of this work was to assess RNA interference (RNAi) technology as an alternative tool for gene silencing previously employed successfully in H. capsulatum. For this purpose, we built different inverted repeat transgenic hairpin constructs to down-regulate the PbGP43 and PbP27 genes known to codify for two fungal immunogenic proteins that elicit a strong immune response during experimental paracoccidioidomycosis. Using the RNAi strategy, a reduction in the mRNA levels of the PbGP43 and PbP27 genes was observed during the first 20 days after selection;however, in the transformed yeast cells, the gene silencing status proved non-stable through the assay. We demonstrated that electrotransformation was suitable to transform P. brasiliensis yeast cells and integrate the hairpin constructions;nonetheless, gene silencing was not stable along the experimental time. A detailed analysis of the underlying molecular RNAi machinery may provide further insights into the intracellular mechanism that governs this reverse genetic tool.

Support Vector Machines for Regression: A Succinct Review of Large-Scale and Linear Programming Formulations

Support Vector-based learning methods are an important part of Computational Intelligence techniques. Recent efforts have been dealing with the problem of learning from very large datasets. This paper reviews the most commonly used formulations of support vector machines for regression (SVRs) aiming to emphasize its usability on large-scale applications. We review the general concept of support vector machines (SVMs), address the state-of-the-art on training methods SVMs, and explain the fundamental principle of SVRs. The most common learning methods for SVRs are introduced and linear programming-based SVR formulations are explained emphasizing its suitability for large-scale learning. Finally, this paper also discusses some open problems and current trends.

Multinational Measles Outbreak in Post-Elimination Era,Involves Three Countries of North America and a European Country in a Short Transmission Chain

Background: Four measles cases in Canada and one in the United States are linked to international importation of measles in Playa del Carmen, Quintana Roo, Mexico. Objective: To describe characteristics of transmission and not spillover to local population in Mexico. Material and Methods: The outbreak investigation was based on active search of cases and in the rapid monitoring of vaccination coverage in children aged 1 - 7 years old. Laboratory confirmation by Enzyme-Linked Immunosorbent Assay (ELISA) and molecular detection by Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR) assay in throat swab and urine were done. Results: One transmission chain with three generations of cases was identified. The primary case was a 9-year-old boy who was infected in Wales, UK. His sisters aged 7 and 15 years old respectively, were the first generation of cases in Mexico. The second generation was related to the imported cases, and affected two Canadian tourists and an American woman aged 39 years old. A third generation occurred in Canada and affected an infant of 15 months of age and his sister aged 4 years old. The genotype D8 which was circulating in UK was identified in these patients. One probable case was detected in Quintana Roo, but was discarded by laboratory testing. The coverage with at least one dose of Measles-Mumps-Rubella (MMR) vaccine ranged from 95% to 99.5%. Conclusion: International travelers with no history of vaccination are at risk of acquiring measles even in countries that have interrupted endemic transmission. The high immunization coverage of measles containing vaccine could explain the absence of cases in Mexican population. Highlights: Multinational measles outbreak in a country without endemic transmission. The findings exhibit the importance of immunization in international travelers in the post-elimination era.

Forecasting the Demand of Short-Term Electric Power Load with Large-Scale LP-SVR

This research studies short-term electricity load prediction with a large-scalelinear programming support vector regression (LP-SVR) model. The LP-SVR is compared with other three non-linear regression models: Collobert’s SVR, Feed-Forward Neural Networks (FFNN), and Bagged Regression Trees (BRT). The four models are trained to predict hourly day-ahead loads given temperature predictions, holiday information and historical loads. The models are trained on-hourly data from the New England Power Pool (NEPOOL) region from 2004 to 2007 and tested on out-of-sample data from 2008. Experimental results indicate that the proposed LP-SVR method gives the smallest error when compared against the other approaches. The LP-SVR shows a mean absolute percent error of 1.58% while the FFNN approach has a 1.61%. Similarly, the FFNN method shows a 330 MWh (Megawatts-hour) mean absolute error, whereas the LP-SVR approach gives a 238 MWh mean absolute error. This is a significant difference in terms of the extra power that would need to be produced if FFNN was used. The proposed LP-SVR model can be utilized for predicting power loads to a very low error, and it is comparable to FFNN and over-performs other state of the art methods such as: Bagged Regression Trees, and Large-Scale SVRs.

Studying the causes of parasite geographic distribution is relevant to understand ecological and evolutionary processes that affect host populations as well as for species conservation. Temperature is one of the most important environmental variables affecting parasite distribution, as raising temperatures positively affect development, reproduction, and rate of transmission of both endo- and ectoparasites. In this context, it is generally accepted that, in mountains, parasite abundance decreases with elevation. However, empirical evidence on this topic is limited. In the present study, we analyzed the elevational variation of hemoparasites and ectoparasites of a lizard, Psammodromus algirus, along a 2,200-m elevational gradient in Sierra Nevada （SE Spain）. As pre- dicted, ectoparasite （mites, ticks, mosquitoes, and sandflies） abundance decreased with elevation. However, hemoparasite prevalence and intensity in the lizard augmented with altitude, showing a pattern contrary to their vectors （mites）. We suggest that tolerance to hemoparasites may increase with elevation as a consequence of lizards at high altitudes taking advantage of increased body condition and food availability, and reduced oxidative stress. Moreover, lizards could have been selected for higher resistance against hemoparasites at lowlands （where higher rates of replication are expected）, thus reducing hemoparasite prevalence and load. Our findings imply that, in a scen- ario of climate warming, populations of lizards at high elevation may face increased abundance of ectoparasites, accompanied with strong negative effects.

SnO_(2) Dense Ceramic Microwave Sintered with Low Resistivity

The Hall-Héroult process is used for alumina reduction by the use of graphite anodes even though it involves a high emission of carbon dioxide (CO2) and several other organic compounds. Proposals have been made aiming at substituting graphite for a single-phase SnO2-based ceramic with low resistivity and chemical resistance to cryolite, which is characterized as an inconsumable anode, reducing pollutant emissions. To this end, a wide range of studies were carried out on SnO2-based ceramics modified with ZnO as a densification aid doped with the promoters of electrical conductivity such as Nb2O5, Al2O3 and Sb2O3 through a mixture of oxides and hybrid sintering in a microwave oven. The pressed pellets were sintered in a microwave oven up to 1050℃ under a constant heating rate of 10℃/min. After sintering, the density was determined by the Archimedes method, the phases were then characterized by X-ray diffraction, the microstructure and chemical composition resulting from the sintered SnO2-based ceramics were also investigated by field emission scanning electron microscopy (FE-SEM) coupled with an energy-dispersive X-ray spectroscopy (EDS) and the electrical properties were determined by the measurements of the electric field x current density. A single-phase ceramic was obtained with a relative density of above 90% and electrical resistivity of 6.1 Ω·cm at room temperature. The ceramics obtained in this study could be a potential candidate as an inconsumable anode to replace the current fused coke used in the reduction of alumina.
