Form.tion potential and analysis of 32 regulated and unregulated disinfection by-products: Two new sim.lified m.thods

Water disinfection is an essential process that provides safe water by inactivating pathogens that cause waterborne diseases. However, disinfectants react with organic m.tter naturally present in water, leading to the form.tion of disinfection by-products(DBPs). m.lti-analyte m.thods based on m.ss spectrom.try(m.) are preferred to quantify m.ltiple DBP classes at once however, m.st require extensive sam.le pre-treatm.nt and significant resources. In this study, two analytical m.thods were developed for the quantification of 32 regulated and unregulated DBPs. A purge and trap(P&T) coupled with gas chrom.tography m.ss spectrom.try(GC-m.) m.thod was optim.zed that autom.ted sam.le pre-treatm.nt and analyzed volatile and sem.-volatile com.ounds, including trihalom.thanes(THm.), iodinated trihalom.thanes(I-THm.), haloacetonitriles(HANs), haloketones(HKTs) and halonitrom.thanes(HNm.). LOQs were between 0.02-0.4 μg/L for m.st DBPs except for 8 analytes that were in the low μg/L range. A second m.thod with liquid chrom.tography(LC) tandem.m.ss spectrom.try(m./m.) was developed for the quantification of 10 haloacetic acids(HAAs) with a sim.le clean-up and direct injection. The LC-m./m. direct injection m.thod has the lowest detection lim.ts reported(0.2-0.5 μg/L). Both m.thods have a sim.le sam.le pre-treatm.nt, which m.ke it possible for routine analysis. Hyperchlorination and uniform.form.tion conditions(UFC) form.tion potential tests with chlorine were evaluated with water sam.les containing high and low TOC. Hyperchlorination form.tion potential test m.xim.zed THm. and HAAs while UFC m.xim.zed HANs. Ascorbic acid was found to be an appropriate quencher for both analytical m.thods. Disinfected drinking water from.four water utilities in Alberta, Canada were also evaluated.

Background:A key step in gene expression is the recognition of the stop codon to term.nate translation at the correct ***,it has been observed that ribosom.s can m.sinterpret the stop codon and continue the translation in the 3′UTR *** phenom.non is called stop codon read-through(SCR).It has been suggested that these events would occur on a program.ed basis,but the underlying m.chanism. are still not well ***:Here,we present a strategy for the com.rehensive identification of SCR events in the Drosophila m.lanogaster transcriptom. by evaluating the ribosom.l density *** associated ribosom.l leak rate was estim.ted for every event identified.A statistical characterization of the frequency of nucleotide use in the proxim.l region to the stop codon in the sequences associated to SCR events was ***:The results show that the nucleotide usage pattern in transcripts with the UGA codon is different from.the pattern for those transcripts ending in the UAA codon,suggesting the existence of at least two m.chanism. that could alter the translational term.nation ***,a linear regression m.dels for each of the three stop codons was developed,and we show that the m.dels using the nucleotides at inform.tive positions outperform. those m.dels that consider the entire sequence context to the stop ***:We report that distal nucleotides can affect the SCR rate in a stop-codon dependent m.nner.

Root-secreted phytochem.cals m.diate m.ltiple interactions in the rhizosphere. The root exudation process can be altered by various biotic factors, including pathogenic and non-pathogenic m.crobes, and abiotic factors like tem.erature and soil m.isture. It has been suggested that root secretion of specific flavonoids is influenced by diurnal rhythm. （by light or dark） but a com.rehensive analysis of the overall secretion of phytochem.cals in response to diurnal rhythm. has not been studied. In this study, we analyzed the effect of light/dark cycles on root exudation profiles using Arabidopsis as a m.del plant. Our results reveal that the root secretion of phytochem.cals is partly regulated by the diurnal light cycle and follows two m.in patterns of secretion：（1） the large m.jority of phytochem.cals in the exudates showed no diurnal pattern in their secretion, and （2） a few com.ounds showed a diurnal pattern in their secretion： three com.ounds increased in secretion only under light; two com.ounds increased in secretion only while it was dark; and two com.ounds increased in secretion during the transition from.dark to light. Root-specific ABC transporters have been im.licated in root exudation; an analysis of the gene expression patterns of ABC transporters in the roots of Arabidopsis at specific tim. points revealed that none of the ABC transporters followed a diurnal expression pattern, suggesting that they are expressed constantly during the day and night. Sim.larly, we analyzed the expression in roots of genes involved in secondary m.tabolite biosynthesis and found that som. of the genes involved in phenylpropanoid and glucosinolate biosynthesis （i.e. 4-coum.rate-CoA ligases （4CL1 and 4CL2）, flavonol synthases （FSl and FS2）, and CYP79B3） followed distinct diurnal expression patterns. We present an unusual case of portal vein throm.osis with a splanchnic hem.ngiom. secondary to acute biliary pancreatitis. We report a 45-year-old patient, who has system.c arterial hypertension in treatm.nt, was adm.tted for abdom.nal pain in the epigastrium. with irradiation to the right hypochondrium. accom.anied by nausea and vom.ting of 10 occasions of bile content, physical exam.nation with pain in the right hypochondrium. m.rphy positive. We have laboratory studies with a lipase of 788, so a diagnosis of pancreatitis is m.de with an etiology to be determ.ned. The laboratories suggestive of acute biliary pancreatitis (lipase 788.71);an im.ging study was subsequently perform.d (ultrasonography) with the result of stone in the com.on bile duct. A laparoscopy was perform.d with relative im.rovem.nt, so he was discharged and returned 20 days after surgery due to abdom.nal pain of the sam. intensity in the left hypochondrium. Ending his hospitalization with a splenectom. for splenic hem.ngiom. with portal vein throm.osis.

本研究的目的是i)确定伊比利亚半岛一种大疣蛛(m.crothele calpeiana)分布的气候相关性以预测其潜在分布,ii)详细阐述该物种在伊比利亚半岛的分布假说,iii)通过推断模型预测确认该物种在北非和整个地中海地区的适合区域,iv)预测气候变暖对蜘蛛潜在分布区的影响。基于物种的存在、远离目前环境条件下的可能缺失以及其它的气候参数,使用广义线性模型发展了潜在分布的可预测模型。蜘蛛在伊比利亚半岛上的潜在分布远大于目前已知的分布区,延伸到尚未发现蜘蛛分布的葡萄牙广大地区。本文提出了该种大疣蛛在适合分布区域内没有分布的历史因素。北非具有适合该物种的条件但却没有该属物种的分布支持了大疣蛛(m.crothele)祖先的东方起源假说。对地中海地区蜘蛛分布的推断突出显示了阿根廷地区有合适的分布区,该地区也发现有另一种欧洲大疣蛛。气候变暖将对现存于伊比利亚的***种群产生负面影响,因为气候变暖将减少并破碎化蜘蛛在北非的潜在栖息地。目前,急需确认在葡萄牙广大地区是否存在蜘蛛物种,发展对该属的系统发育研究以确定大疣蛛属起源和扩散史的理论。
