Adoptive cell therapy(ACT)is an immunotherapy strategy for cancer that has seen widespread clinical *** ACT,patient-derived lymphocytes are stimulated with the antigen of interest ex vivo,proliferated,then returned to the patient to initiate an antigen-specific antitumor *** effective,this process is resource-intensive and logistically impossible for many *** artificial antigen presenting cells(aAPCs)offer a potential“off-the-shelf”alternative to ex vivo *** particulate aAPCs perform well in vitro,they have had limited success in vivo due to poor bioavailability after *** to bioavailability include rapid clearance,unfavorable biodistribution,and inadequate interactions with CD8+T cells at sites of *** properties such as elasticity have been shown to vastly impact the bioavailability and particle-cell interactions,but this has yet to be investigated in the context of aAPCs for in vivo T-cell *** literature likewise indicates that biomaterial properties,especially elasticity,can modulate T-cell activation in *** the goal of creating a more biomimetic,next-generation particulate aAPC,we developed a poly(ethylene)glycol hydrogel particle platform with tunable elasticity to investigate the impact of elasticity on antigen-specific T cell activation for in vivo adoptive *** this knowledge,we were able to gain more precise control over in vivo T cell activation and investigate possible mechanisms including the effects of aAPC elasticity on T cell binding,macrophage uptake,and the protein corona.

This work explores the potential of a triple combination of meropenem(MEM),a novel metallo-blactamase(MBL)inhibitor(indole-2-carboxylate 58(InC58)),and a serine-b-lactamase(SBL)inhibitor(avibactam(AVI))for broad-spectrum activity against carbapenemase-producing bacteria.A diverse panel comprising MBL-and SBL-producing strains was used for susceptibility testing of the triple combination using the agar dilution *** frequency of resistance(FoR)to MEM combined with InC58 was *** were sequenced and tested for cross resistance,fitness,and the stability of the resistance *** with the double combinations of MEM plus an SBL or MBL inhibitor,the triple combination extended the spectrum of activity to most of the isolates bearing SBLs(oxacillinase-48(OXA-48)and Klebsiella pneumoniae carbapenemase-2(KPC-2))and MBLs(New Delhi metallo-blactamases(NDMs)),although it was not effective against Verona integron-encoded metallo-blactamase(VIM)-carrying Pseudomonas aeruginosa(***)and OXA-23-carrying Acinetobacter baumannii(***).The FoR to MEM plus InC58 ranged from 2.22×10^(-7)to 1.13×10^(-6).The resistance correlated with mutations to ompC and comR,affecting porin C and copper permeability,*** mutants manifested a fitness cost,a decreased level of resistance during passage without antibiotic pressure,and cross resistance to another carbapenem(imipenem)and a b-lactamase inhibitor(taniborbactam).In conclusion,compared with the dual combinations,the triple combination of MEM with InC58 and AVI showed a much wider spectrum of activity against different carbapenemaseproducing bacteria,revealing a new strategy to combat b-lactamase-mediated antimicrobial resistance.

Early life adversity as a risk factor for cognitive impairment and Alzheimer’s disease

Neurological conditions,including cognitive impairment and Alzheimer’s disease(AD),impose a huge burden on society,affecting millions of people *** addition to genetic factors,recent studies indicate that environmental and experiential factors may contribute to the pathogenesis of these *** life adversity(ELA)has a profound impact on brain function and health later in *** rodent models,exposure to ELA results in specific cognitive deficits and aggravated AD *** concerns have been raised regarding the higher risk of developing cognitive impairments in people with a history of *** this review,we scrutinize findings from human and animal studies focusing on the connection of ELA with cognitive impairment and *** discoveries suggest that ELA,especially at early postnatal stages,increases susceptibility to cognitive impairment and AD later in *** terms of mechanisms,ELA could lead to dysregulation of the hypothalamus-pituitary-adrenal axis,altered gut microbiome,persistent inflammation,oligodendrocyte dysfunction,hypomyelination,and aberrant adult hippocampal *** among these events may synergistically contribute to cognitive impairment later in ***,we discuss several interventions that may alleviate adverse consequences of *** investigation into this crucial area will help improve ELA management and reduce the burden of related neurological conditions.

A comparison of lower limb stiffness and mechanical muscle function in ACL-reconstructed,elite,and adolescent alpine ski racers/ski cross athletes

Purpose： The aim of this study was to compare mechanical muscle function in the eccentric/concentric phases of vertical bilateral jumping in anterior cruciate ligament-reconstructed （ACLR）, elite （ELITE）, and adolescent （ADOL） alpine ski racers and ski cross athletes. Methods： Alpine ski racers/ski crossers （ACLR： n = 12, age = 26.7 ± 3.8 years; ELITE： n = 12, age = 23.9 ± 3.0 years; ADOL： n = 12, age = 17.8 ± 0.7 years; females： n = 6 per group, males： n = 6 per group） performed 5 maximal countermovement jumps （CMjs） and 5 squat jumps. The ground reaction forces for each limb were analyzed using dual force plate recording to obtain body center of mass （BCM） velocity, displacement, and power. The eccentric deceleration （ECC） and concentric phases were determined from BCM velocity. CMj net concentric and ECC impulses were calculated （body mass normalized） along with the peak and mean BCM power and maximal vertical jump height. CMj lower limb stiffness （LLS） was determined by the slope of the ground reaction forces vs. the BCM displacement curve over the ECC phase. Concentric and ECC asymmetry indices were calculated for each leg, and the left vs. right LLS was compared. Outcome measures （reported as mean ± SD） calculated as a 5-jump mean were normalized to body mass and compared using an analysis of variance. Results： No between-group differences were found for peak and mean power or jump heights. There were no group differences for LLS or net concentric phase impulse, but the net ECC impulse was lower in the ADOL group compared with ELITE skiers （ADOL： 1.33 ± 0.32 Ns/kg; ELITE： 1.59 ± 0.16 Ns/kg; p 〈 0.05）. Although no group differences were found for ECC asymmetry indices, a group × limb interaction was found for LLS （p 〈 0.01）, which was systematically higher in the right vs. the left limb of ADOL skiers （right： 54.1 ± 17.9 N/m/kg; left： 48.7 ± 15.7 N/m/kg;p 〈 0.01）. Sickle Cell Anemia(SCA)is a prevalent genetic condition in Saudi Arabia,particularly in the jazan *** study’s purpose was to assess the prevalence of de-pression among individuals with SCA in this area and to identify the factors associated with this *** study involved 391 adult participants,all patients in the hematology center of Prince Mohammed bin Nasser Hospital(PMNH).Data was collected through an anonymous interview questionnaire–the Arabic version of the Patient Health Questionnaire-9(PHQ-9)–between March–May *** questionnaire covered socio-demographic information and variables related to the participant’s SCA history,such as pain levels,frequency of pain,hospitalization duration,and frequency of blood *** variables underwent Chi-square and One-way ANOVA testing,followed by multivariate *** study found a 42%prevalence of depression among the SCA *** significantly linked with heightened depression levels included being widowed or divorced,experiencing frequent and severe pain,and longer duration of hospital *** educational level of at least a bachelor’s degree was also a significant *** study reveals a high prevalence of depression among SCA patients in the jazan *** underscores the need for policymakers to educate both physicians and patients about the importance of mental health in SCA *** study also highlights the need for more research into the specific causes and effects of depressive symptoms in SCA patients to inform the creation of effective management plans.

Interference from HONO in the measurement of ambient air NO2 via photolytic conversion and quantification of NO

The reference method to quantify mixing ratios of the criteria air pollutant nitrogen dioxide(NO_(2)) is NO-O_(3) chemiluminescence(CL), in which mixing ratios of nitric oxide(NO) are measured by sampling ambient air directly, and mixing ratios of NO_(x)(= sum of NO and NO_(2))are measured by converting NO_(2) to NO using, for example, heated molybdenum catalyst or,more selectively, photolytic conversion(P-CL).In this work, the nitrous acid(HONO) interference in the measurement of NO_(2) by P-CL was *** with two photolytic NO2 converters are *** first used radiation centered at 395 nm, a wavelength region commonly utilized in *** second used light at 415 nm, where the overlap with the HONO absorption spectrum and hence its photolysis rate are *** ratios of NO_(2),NO_(x) and HONO entering and exiting the converters were quantified by Thermal Dissociation Cavity Ring-down Spectroscopy(TD-CRDS).Both converters exhibited high NO_(2) conversion efficiency(CF_(NO2);> 90%) and partial conversion of *** of CF against flow rate were consistent with photolysis frequencies of 4.2 s^(-1) and 2.9 s^(-1) for NO_(2) and 0.25 s^(-1) and 0.10 s^(-1) for HONO at 395 nm and 415 nm, *** larger than predicted from the overlap of the emission and HONO absorption *** results imply that measurements of NO2 by P-CL marginally but systematically overestimate true NO_(2) concentrations,and that this interference should be considered in environments with high HONO:NO_(2) ratios such as the marine boundary layer or in biomass burning plumes.
