Comprehensive molecular characterization to predict immunotherapy response in advanced biliary tract cancer:a phase II trial of pembrolizumab

Background:Immune checkpoint inhibitors(ICIs)are effective in a subset of patients with metastatic solid ***,the patients who would benefit most from ICIs in biliary tract cancer(BTC)are still *** and methods:We molecularly characterized tissues and blood from 32 patients with metastatic BTC treated with the ICI pembrolizumab as second-line ***:All patients had microsatellite stable(MSS)type *** of the 32 patients achieved partial response(PR),with an objective response rate(ORR)of 9.4%(95%confidence interval[CI],2.0–25.2)and nine showed stable disease(SD),exhibiting a disease control rate(DCR)of 37.5%(95%CI,21.1–56.3).For the 31 patients who had access to PD-1 ligand 1(PD-L1)combined positive score(CPS)testing(cut-off value≥1%),the ORR was not different between those who had PD-L1-positive(PD-L1+;1/11,9.1%)and PDL1-(2/20,10.0%)tumors(p=1.000).The tumor mutational burden(TMB)of PD-L1+BTC was comparable to that of PD-L1-BTC(p=0.630).TMB and any exonic somatic mutations were also not predictive of pembrolizumab *** analysis of blood and tumor samples demonstrated a relatively high natural killer(NK)cell proportion in the peripheral blood before pembrolizumab treatment in patients who achieved tumor ***,the tumors of these patients presented high enrichment scores for NK cells,antitumor cytokines,and Th1 signatures,and a low enrichment score for cancer-associated ***:This study shows the molecular characteristics associated with the efficacy of pembrolizumab in BTC of the MSS type.

The electrical characteristics of hybrid super capacitor were evaluated by synthesizing LTO(Li_(4)Ti_(5)O_(12))using TiO_(2) having a hydrogen titanate nanowire *** of the hydrogen titanate nanowire was implemented by using TiO_(2) having size of 60 nm and NaOH,and performing synthesis at 70℃for 6 h with a sonochemical *** compound was synthesized at 150℃for 36 h and at 180℃for 36 h respectively by using the hydrogen titanate nanowire and LiOH·H2O as starting materials with a hydrothermal *** final LTO compound was synthesized at 700℃for 6 h using a solid-state *** a result of manufacturing the hybrid super capacitor using LTO synthesized at 180℃for 36 h with the hydrothermal method,a capacity of 198 mA·h/g has been achieved compared to a theoretical capacity of 172 mA·h/g of existing LTO,and thus,the capacity has been increased by about 13%.Further,such excellent cycle performance has ensured its possibility as a high-capacity capacitor.

Nitric oxide(NO)has been shown to promote revascularization and nerve regeneration after peripheral nerve ***,in vivo application of NO remains challenging due to the lack of stable carrier materials capable of storing large amounts of NO molecules and releasing them on a clinically meaningful time ***,a silica nanoparticle system capable of reversible NO storage and release at a controlled and sustained rate was *** this study,NO-releasing silica nanoparticles(NO-SNs)were delivered to the peripheral nerves in rats after acute crush injury,mixed with natural hydrogel,to ensure the effective application of NO to the *** using a polymer dye and immunohistochemical staining for the detection of CD34(a marker for revascularization)results showed that NO-releasing silica nanoparticles increased revascularization at the crush site of the sciatic *** sciatic functional index revealed that there was a significant improvement in sciatic nerve function in NO-treated *** and anatomical analyses showed that the number of myelinated axons in the crushed sciatic nerve and wet muscle weight excised from NO-treated rats were ***,muscle function recovery was improved in rats treated with *** together,our results suggest that NO delivered to the injured sciatic nerve triggers enhanced revascularization at the lesion in the early phase after crushing injury,thereby promoting axonal regeneration and improving functional recovery.

Accurate prediction of thermodynamic properties requires an extremely accurate representation of the free-energy *** are twofold—first,the inclusion of the relevant finite-temperature mechanisms,and second,a dense volume–temperature grid on which the calculations are performed.A systematic workflow for such calculations requires computational efficiency and reliability,and has not been available within an ab initio framework so ***,we elucidate such a framework involving direct upsampling,thermodynamic integration and machine-learning potentials,allowing us to incorporate,in particular,the full effect of anharmonic *** improved methodology has a five-times speed-up compared to state-of-the-art *** calculate equilibrium thermodynamic properties up to the melting point for bcc Nb,magnetic fcc Ni,fcc Al,and hcp Mg,and find remarkable agreement with experimental data.A strong impact of anharmonicity is observed specifically for *** introduced procedure paves the way for the development of ab initio thermodynamic databases.

A modular and efficient synthesis of the biologically significant C-methylisoflavones isosideroxylin（1）,6,8-dimethylgenistein（2） and their analogues（3-8） is established for the first *** synthesis is realized in 7-8 steps in overall yields of 16%-24%from commercially inexpensive phloroglucinol and features a high yielding Vilsmeier-Haack reaction,Friedel-Crafts acylation,Gammill＇s protocol and Suzuki coupling as the pivotal ***,these compounds evaluated for their inhibitory potency on the production of nitric oxide（NO） in lipopolysaccharide（LPS）-activated RAW-264.7 cells as an indicator of anti-inflammatory *** results showed that all the compounds decreased NO production in a dose-dependent manner without marked cytotoxicity and IC_（50） values are found in the range of 10.17-33.88 μmol/*** note,compounds 3 followed by 1,7 and 8 show comparable inhibitory activity with positive control（N-monomethyl-L-arginine,L-NMMA）.

Hydrogen Generation from the Dissociation of Water Using Microwave Plasmas

Hydrogen is produced by direct dissociation of water vapor,i.e.,splitting water molecules by the electrons in water plasma at low pressure(<10–50 Torr)using microwave plasma *** condition generates a high electron temperature,which facilitates the direct dissociation of water molecules.A microwave plasma source is developed,utilizing the magnetron of a microwave oven and a TE10 rectangular *** quantity of the generated hydrogen is measured using a residual gas *** electron density and temperature are measured by a Langmuir probe,and the neutral temperature is calculated from the OH line intensity.

This study uses the smart phone with the Android system to construct a cloud-side smart switch system in the client-server architecture with the Android open platform system, using microprocessors(MCU 16F690) as the thermostat controller and combined with Raspberry Pi. The computing technology extends the control of the constant temperature from the local to the cloud, allowing the user to view the temperature status recorded by the cloud server using the smart phone application(App) or web browser. It can even remotely control the heating power of the smart switch for the heating device in the Internet control environment and use the proportional-integral-derivative(PID) control and the pulse-width modulation(PWM) technology to achieve intelligent constant temperature *** proposed control system uses the expert PID control as the core to calculate the duty cycle of the PWM signal to control the power output of the smart switch for the constant temperature water tank. The experiment results verify the effectiveness of the proposed system.

The carbon dioxide （CO2） removal efficiency, reaction rate, and CO2 loading into aqueous blended monoethanolamine （MEA）＋ 2-amino-2-methyl-l-propanol （AMP） solutions to enhance absorption characteristics of MEA and AMP were carried out by the absorption/regeneration process. As a result, compared to aqueous MEA and AMP solutions, aqueous blended MEA ＋ AMP solutions have a higher CO2 loading than MEA and a higher reaction rate than AMP. The CO2 loading of rich amine of aqueous 18 wt.% MEA ＋ 12 wt.% AMP solution was 0.62 mol CO2/mol amine, which is 51.2% more than 30 wt.% MEA （0.41 mol CO2/mol amine）. Consequently, blending MEA and AMP could be an effective way to design considering economical efficiency and used to operate absorber for a long time.
