Metabolic collaboration between cells in the tumor microenvironment has a negligible effect on tumor growth

The tumor microenvironment is composed of a complex mixture of different cell types interacting under conditions of nutrient deprivation,but the metabolism therein is not fully understood due to difficulties in measuring metabolic fluxes and exchange of metabolites between different cell types in ***-scale metabolic modeling enables estimation of such exchange fluxes as well as an opportunity to gain insight into the metabolic behavior of individual cell ***,we estimated the availability of nutrients and oxygen within the tumor microenvironment using concentration measurements from blood together with a metabolite diffusion *** addition,we developed an approach to efficiently apply enzyme usage constraints in a comprehensive metabolic model of human *** combined modeling reproduced severe hypoxic conditions and the Warburg effect,and we found that limitations in enzymatic capacity contribute to cancer cells’preferential use of glutamine as a substrate to the citric acid ***,we investigated the common hypothesis that some stromal cells are exploited by cancer cells to produce metabolites useful for the cancer *** identified over 200 potential metabolites that could support collaboration between cancer cells and cancer-associated fibroblasts,but when limiting to metabolites previously identified to participate in such collaboration,no growth advantage was *** work highlights the importance of enzymatic capacity limitations for cell behaviors and exemplifies the utility of enzyme-constrained models for accurate prediction of metabolism in cells and tumor microenvironments.

Oncologic outcomes with and without amniotic membranes in robotic-assisted radical prostatectomy:A propensity score matched analysis

Objective:Placement of human placenta derived grafts during robotic-assisted radical prostatectomy(RARP)hastens the return of continence and *** long-term impact on the oncologic outcomes remains to be *** objective was to determine the oncologic outcomes of patients with dehydrated human amnion chorion membrane(dHACM)at RARP compared to a matched ***:In a referral centre,from August 2013 to October 2019,599 patients used dHACM in bilateral nerve-sparing *** excluded patients with less than 12 months follow-up,simple prostatectomy,and unilateral *** with dHACM(amnio group)were 529,and were propensity score matched 1:1 to 2465 patients without dHACM(non-amnio group)and a minimum follow-up of 36 *** the time of RARP,dHACM was placed around the neurovascular bundle in the amnio *** and categorical variables in matched groups was tested by two-sample Kolmogorov-Smirnov test and Fisher's exact test *** measured were biochemical recurrence(BCR),adjuvant and salvage therapy ***:Propensity score matching resulted in two groups of 444 *** incidence functions for BCR did not show a difference between the groups(p=0.3).Patients in the non-amnio group required salvage therapy more frequently than the amnio group,particularly after partial nerve-sparing RARP(6.3%vs.2.3%,p=0.001).limitations are the absence of prospective ***:The data suggest that using dHACM does not have a negative impact on BCR in *** of cancer specific and overall survival will require follow-up study to increase our understanding of these grafts’impact on prostate cancer biology.

Predictive Factors of Renal Failure in COVID-19 Patients at the Anti-COVID Center in lome, Togo

Background: Angiotensin-converting enzyme 2 has been identified as the receptor that allows the entry of SarsCov2 into the human cell. Its expression in the kidney is 100 times higher than in the lung;thus, making the kidney an excellent target for SarsCov2 infection manifesting as renal failure (RF). The objective of this study was to determine the predictive factors of RF during COVID-19 in the Togolese context. Patients and Methods: This was a retrospective descriptive and analytical study conducted at the lomé Anti-COVID Center including the records of patients hospitalized for COVID-19, of age ≥ 18 years and having performed a creatinemia. RF was defined by a GFR 2 calculated according to the MDRD formula. Patients were randomized into 2 groups according to GFRResults: 482 patients were selected for this study with a mean age of 58.02 years. Sixty-five percent of the patients were men, i.e., a sex ratio of 1.88. Fifty-two patients had RF, i.e., a frequency of 10.8%.There were 65% men (315 cases), for a sex ratio (M/F) of 1.88. Risk factors for renal failure in COVID-19 were age ≥ 65 years (ORa 2.42;CIa95% [1.17 - 4.95];p = 0.016), anemia (ORa 2.49;CIa95% [1.21 - 5.26];p = 0.015), moderate (ORa 13;CIa95% [2.30 - 2.44];p = 0.017), severe (ORa 26.2;CIa95% [4.85 - 4.93];p = 0.002) and critical (ORa 108;CIa95% [16.5 - 21.76];p Conclusion: Renal failure would therefore be related to the severity of COVID-19 and is the most formidable factor, conditioning the course of the disease and the patient’s vital prognosis.

Purpose The transition into full-time training represents a key period in the development of young soccer *** we compared the weekly training loads(matches,field-,and resistance-training)of English Premier-league Academy soccer players from under-16(U16),under-18(U18)and under-23(U23)age-groups during a training *** Forty players(U16 n=13,U18 n=15 and U23 n=12)were monitored using global navigation satellite systems and differential ratings of perceived exertion(dRPE).External load metrics were total distance,high-speed running distance,[absolute:≥19.8 km/h,relative:≥87%of 30-15 final-velocity(vIFT)],sprint distance(absolute:≥25.2 km/h,relative:≥80%maximal sprint speed),and dynamic stress *** load metrics were dRPE training *** Other than relative sprint distance,overall weekly external training loads were substantially greater for U18s and U23s when compared with U16s[effect size range:1.09-1.99(moderate to large);±90%confidence limits~0.45].When compared with U16s,overall internal loads were substantially greater for U18s[0.69-0.95(moderate);±~0.40],but not *** in weekly training loads between U18s and U23s were *** Substantial differences in training loads between elite U16 players and their older counterparts,indicates the need for planned increases in training loads in anticipation of the transition into full-time training.

The Prostate Cancer Prevention Trial （PCPT） is a seminal study in the field of urology. More than 10years after its initial publication, updated data from this trial continue to shape our understanding of prostate cancer. Among the major findings from the PCPT has been the demonstration that prostate cancer is common in men with prostate-specific antigen （PSA） once thought to be in the normal range,~ finasteride prevents the development of benign prostatic hypertrophy,2 it increases the sensitivity of PSA3 and digital rectal examination.4 Furthermore the PCPT helped to establish the link between erectile dysfunction and cardiovascular disease,5 and perhaps most importantly finasteride demonstrated a 25% relative risk reduction in the diagnosis of prostate cancer compared with placebo.

lHCb gets closer to discovering the second doubly charmed baryon

Recently the lHCb Collaboration [1] published the results of a search for the doubly charmed baryon ■. No significant signal is seen in the mass range from 3.4 to3.8 Ge V. To put this result in context, the ■baryon was seen by lHCb in decay modes ΛcK-π^+π^+[2](2017) and ■^+cπ^+[3](2018). The weighted average of the ■^++cc mass is 3621.24±0.65 (stat.)±0.31 (syst.) MeV [3].
